TRPM7 plays an important role in cellular Ca2+, Zn2+ and Mg2+ homeostasis. TRPM7 channels are abundantly expressed in ameloblasts and, in the absence of TRPM7, dental enamel is hypomineralized. The potential role of TRPM7 channels in Ca2+ transport during amelogenesis was investigated in the HAT-7 rat ameloblast cell line. The cells showed strong TRPM7 mRNA and protein expression. Characteristic TRPM7 transmembrane currents were observed, which increased in the absence of intracellular Mg2+ ([Mg2+]i), were reduced by elevated [Mg2+]i, and were inhibited by the TRPM7 inhibitors NS8593 and FTY720. Mibefradil evoked similar currents, which were suppressed by elevated [Mg2+]i, reducing extracellular pH stimulated transmembrane currents, which were inhibited by FTY720. Naltriben and mibefradil both evoked Ca2+ influx, which was further enhanced by the acidic intracellular conditions. The SOCE inhibitor BTP2 blocked Ca2+ entry induced by naltriben but not by mibefradil. Thus, in HAT-7 cells, TRPM7 may serves both as a potential modulator of Orai-dependent Ca2+ uptake and as an independent Ca2+ entry pathway sensitive to pH. Therefore, TRPM7 may contribute directly to transepithelial Ca2+ transport in amelogenesis.  相似文献

Kinetic and thermodynamic analyses of catalytic hydrodechlorinations in supercritical carbon dioxide (SC-CO2) were performed using 5% Pd supported on γ-Al2O3. The selected standard compounds used for the study represented chlorinated wood resins commonly found in pitch deposits; 1-chlorooctadecane (C18-Cl), 9,10-dichlorostearic acid (Stearic-Cl2), and 12,14-dichlorodehydroabietic acid (DHA-Cl2). The reaction utilized isopropanol as a hydrogen donor. Pressure, temperature, and the concentrations of isopropanol and palladium were varied to study the effect of each parameter and to optimize the dechlorination yield. The reaction in SC-CO2 was compared to the one in liquid solvents at atmospheric pressure. By applying a Langmuir–Hinshelwood kinetic model, the rate-determining step of the reaction was deduced to be adsorption of the chlorinated molecules on the palladium surface. The apparent activation energies of the reactions for C18-Cl, Stearic-Cl2, DHA-Cl2 were 43±5, 40±7, and 135±7 kJ mol−1, respectively, in SC-CO2. The relatively high activation energy for DHA-Cl2 was apparently due to structural differences from the other two compounds. The apparent activation energy of dechlorination of C18-Cl in liquid isopropanol at atmospheric pressure was determined to be 35±3 kJ mol−1, leading to the conclusion that the rate-determining step is the same for this compound in both fluid systems. The enthalpies of desorption of stearic acid and dehydroabietic acid were determined to be 18±2 and 12±2 kJ mol−1, respectively. These values being less than half of the apparent activation energies of dechlorination of their corresponding chlorinated compounds indicates that desorption of the dechlorinated products is not the rate-determining step of the reaction. This was consistent with the conclusion that the rate-determining step is adsorption, on the understanding that the reaction mechanism is same in both fluid systems.  相似文献

Classical Cartesian domain decompositions for parallel lattice Boltzmann simulations of fluid flow through heterogeneous porous media are doomed to workload imbalance as the number of processors increases, thus leading to decreasing parallel performance. A one-lattice lattice Boltzmann method (LBM) implementation with vector data structure combined with even fluid node partitioning domain decomposition and fully-optimized data transfer layout is presented. It is found to provide nearly-optimal workload balance, lower memory usage and better computational performance than classical slice decomposition techniques using sparse matrix data structures. Predictive memory usage and parallel performance models are also established and observed to be in very good agreement with data corresponding to numerical fluid flow simulations performed through 3-dimensional packings of cylinders and polydisperse spheres.  相似文献

In this study, a laboratory scale Univ. of Cape Town enhanced biological phosphorus removal process was operated under controlled conditions at a solids retention time of 15 days. Results are presented for the process performance and oxygen transfer parameters determined by applying the steady state oxygen uptake rate (OUR) and the changing power level (CPL) techniques, as per ASCE standard guidelines. The testing periods were temporally separated to eliminate interference of the tests. During the application of the CPL method, the sludge volume index gradually increased and higher values of the oxygen transfer rate and alpha were measured, in comparison to the data from the steady state OUR method, under similar process performance. Furthermore, the mass transfer rate decreased as the CPL method of testing continued. In contrast, the oxygen transfer parameters remained uniform during the time when the OUR method was applied. The data indicated that the CPL method resulted in higher and variable oxygen transfer parameters, even though the process performance remained unchanged. Therefore, a more rigorous evaluation of the CPL method is recommended to clarify the validity of the test.  相似文献

The use of the life history calendar (LHC) or the event history calendar as tools for collecting retrospective data has received increasing attention in many fields of social science and medicine. However, little research has examined the use of this method with web-based surveys. In this study, we adapted this method to an on-line setting to collect information about young adults' life histories, sexual behaviors, and substance use. We hypothesized that the LHC method would help respondents to date sensitive and non-sensitive events more precisely than when using a conventional questionnaire. We conducted an experimental design study comparing university students' responses to an on-line LHC and a conventional on-line question list. A test-retest design in which the respondents completed the survey again two weeks later was also applied to test the precision and reliability of the participants' dating of events. The results showed that whereas the numbers of sensitive and non-sensitive events were generally similar for the two on-line questionnaires, the responses obtained with the LHC were more consistent across the two administrations. Analyses of the respondents' on-line behavior while completing the LHC confirmed that respondents used the LHC's graphic interface to correct and reedit previous answers, thus decreasing data errors.  相似文献

Microgrids can provide an avenue for increasing the amount of distributed generation (DG) and delivery of electricity, where control is more dispersed and quality of service is locally tailored to end-use requirements, with applications from military bases to campuses to commercial office buildings. Many studies have been done to date on microgrid technology and operations, but fewer studies exist on demonstration programs and commercial microgrid development. As China prepares to launch the largest microgrid demonstration program in the world, we review progress made by demonstration programs across Europe, Asia, and the Americas as well as microgrid benefits and barriers. Through case studies, we highlight the difference in experience for microgrids developed under the auspices of a government-sponsored demonstration program versus those that were commercially developed. Lastly, we provide recommendations oriented towards creating a successful microgrid demonstration program.  相似文献

Electric recreational vehicles represent a new challenge in terms of power supply systems compared to the current light-duty electric vehicles, which achieve high performance and long-range. The recreational vehicles need to heed the limited dimension requirements while assuring the high requested power. This paper proposes an integration of Lithium-Ion Capacitor (LIC) with Fuel Cell (FC) without any power electronic device for a three-wheel electric motorcycle. Unlike other hybrid power supply systems, the proposed FC-LIC passive configuration is lighter, compact, more efficient, and simpler to implement. Due to the different impedance of the components the system is self-management, in which FC supplies the average power component and LIC operates as a low-pass filter. In this respect, a simulator is built based on experimental tests to study the system performance in terms of hydrogen consumption and FC degradation. Subsequently, the system is tested under three standard motorcycle driving cycles at three different FC system lifespan stages. The obtained results demonstrate that a passive topology can supply the requested power along different FC stages of life and reported just an increment of 12% of hydrogen consumption at the oldest condition compared to the new condition.  相似文献

Finnish food control authorities waived pre-inspections of food premises in 2011, leaving food business operators to begin operating with no pre-operation food control. This study aims to investigate the effects of this food policy change on the preconditions for Good Hygienic Practices (GHPs) on food premises. Of the 916 food premises that were included in this study, 379 were pre-approved whereas 537 merely notified their operations. The results show that notified food service premises (restaurants) preparing food displayed significantly more non-compliance pertaining to infrastructure than did restaurants pre-approved for food preparation (11.5% and 1.8% of the premises, respectively) (p < 0.05). Significant differences also emerged in the number of premises with non-compliance pertaining to cleaning facilities and equipment, and marked differences in the adequacy of hand and other washing sites. Such instances of non-compliance weaken the preconditions for GHPs. The results suggest that re-introducing pre-inspections of restaurants would strengthen the preconditions for GHPs and possibly provide a model for other countries with similar food control systems.  相似文献

Ready-to-eat (RTE) foods are not further treated before consumption in such a way that may significantly reduce the microbial load, therefore the risk of foodborne disease must be considered. In this regard, the use of natural antimicrobial compounds is an interesting method to be considered. On this topic, the antibacterial activity of cranberry juice concentrate (CJC) have been evaluated in vitro and in situ against 3 foodborne pathogenic bacteria. Results showed a high antimicrobial effect with a noticeable inhibition capacity against Escherichia coli O157:H7, Listeria monocytogenes, Salmonella typhimurium. Acid sensitivity studies of bacteria indicated that at the same pH level (pH = 2.4) in presence of organic acid solution (citric and quinic acids), cranberry juice concentrate showed greater antibacterial effects than the acids due to their phenolic compounds. In situ studies showed 2.5, 1.8 and 5 log reduction of E. coli, L. monocytogenes and S. typhimurium, respectively in presence of cranberry juice concentrate, on pre-cut red peppers after 7 days of storage at 4 °C. A total inhibition of L. monocytogenes on fresh cranberry fruits in primary day of storage, was observed. Cranberries treated with CJC also showed a 3 log reduction of S. typhimurium after 4 days of storage at 4 °C. The results suggest that CJC can be an effective preservation, source of natural antibacterial, to protect the RTE foods from foodborne pathogens contamination without effecting on sensorial properties of treated samples and allow to maintain the freshness, sensory and the nutritional quality of RTE foods.  相似文献
