This study aimed at investigating the possible mechanisms of hepatic protective activity of Cichorium intybus L. (chicory) in acute liver injury. Pathological observation, reactive oxygen species (ROS) detection and measurements of biochemical indexes on mouse models proved hepatic protective effect of Cichorium intybus L. Identification of active compounds in Cichorium intybus L. was executed through several methods including ultra performance liquid chromatography/time of flight mass spectrometry (UPLC-TOF-MS). Similarity ensemble approach (SEA) docking, molecular modeling, molecular docking, and molecular dynamics (MD) simulation were applied in this study to explore possible mechanisms of the hepato-protective potential of Cichorium intybus L. We then analyzed the chemical composition of Cichorium intybus L., and found their key targets. Furthermore, in vitro cytological examination and western blot were used for validating the efficacy of the selected compounds. In silico analysis and western blot together demonstrated that selected compound 10 in Cichorium intybus L. targeted Akt-1 in hepatocytes. Besides, compound 13 targeted both caspase-1 and Akt-1. These small compounds may ameliorate liver injury by acting on their targets, which are related to apoptosis or autophagy. The conclusions above may shed light on the complex molecular mechanisms of Cichorium intybus L. acting on hepatocytes and ameliorating liver injury. 相似文献

As a giant leap in DNA self-assembly, DNA origami has exhibited an unprecedented ability to construct nanostructures with arbitrary shapes and sizes. In typical DNA origami, hundreds of short DNA staple strands fold a long, single-stranded (ss) DNA scaffold cooperatively into designed nanostructures. However, large numbers of DNA strands are expensive and would hinder applications such as pharmaceutical investigations because of the complicated components. Therefore, one challenge is how to reduce the number of staple strands needed to construct DNA origami. For a DNA origami structure, the scale-free folding pattern of the scaffold strand is determined by staple strands at the branching vertexes. Simple duplex regions help to define the size-related features of the origami geometry. In this study, we hypothesized that a scaffold strand can be correctly folded into a designed topology by using only staple strands involved in branching vertexes. After assembly, any remaining, flexible, single-stranded regions of the scaffold could be converted into rigid duplexes by DNA polymerase to achieve the designed geometric structures. To demonstrate the concept, we used only 18 staple strands (covering 15 % of the scaffold strand) to assemble a porous DNA nanostructure, which was visualized by atomic force microscopy (AFM). Context and objective: The aim of this study was to develop, characterize and evaluate a mucoadhesive caplet resulting from a polymeric blend (polymeric caplet) for intravaginal anti-HIV-1 delivery.Materials and methods: Poly(lactic-co-glycolic) acid, ethylcellulose, poly(vinylalcohol), polyacrylic acid and modified polyamide 6, 10 polymers were blended and compressed to a caplet-shaped device, with and without two model drugs 3′-azido-3′-deoxythymidine (AZT) and polystyrene sulfonate (PSS). Thermal analysis, infrared spectroscopy and microscopic analysis were carried out on the caplets employing temperature-modulated DSC (TMDSC), Fourier transform infra-red (FTIR) spectrometer and scanning electron microscope, respectively. In vitro and in vivo drug release analyses as well as the histopathological toxicity studies were carried out on the drug-loaded caplets. Furthermore, molecular mechanics (MM) simulations were carried out on the drug-loaded caplets to corroborate the experimental findings.Results and discussion: There was a big deviation between the Tg of the polymeric caplet from the Tg's of the constituent polymers indicating a strong interaction between constituent polymers. FTIR spectroscopy confirmed the presence of specific ionic and non-ionic interactions within the caplet. A controlled near zero-order drug release was obtained for AZT (20 d) and PSS (28 d). In vivo results, i.e. the drug concentration in plasma ranged between 0.012–0.332?mg/mL and 0.009–0.256?mg/mL for AZT and PSS over 1–28 d.Conclusion: The obtained results, which were corroborated by MM simulations, attested that the developed system has the potential for effective delivery of anti-HIV-agents. 相似文献

The falling down problem has become one of the very important issues of global public health in an aging society. The specific equipment was adopted as the detection device of falling-down in the early studies, but it is inconvenient for the elderly and difficult for future application. The smart phone more commonly used than the specific fall detection equipment is selected as a mobile device for human fall detection, and a fall detection algorithm is developed for this purpose. What the user has to do is to put the smart phone in his/her thigh pocket for falling down detection. The signals detected by the tri-axial G-sensor are converted into signal vector magnitudes as the basis of detecting a human body in a stalling condition. The Z-axis data sets are captured for identification of human body inclination and the occurrence frequencies at the peak of the area of use are used as the input parameters. A high-level fuzzy Petri net is used for the analysis and the development of identifying human actions, including normal action, exercising, and falling down. As an industrial pollutant, tar derived from biomass gasification is used as the precursor for fabricating a novel carbon-metal hydroxides composite electrode. A slurry (the mixture of tar, KOH and melamine) is daubed uniformly onto the nickel foam, which is directly carbonized to form NPC@LDH electrode material. This electrode is further coated with NiCo-LDH nanosheets using an electrodeposition method to form NF@NPC@LDH. The newly made NF@NPC@LDH electrode exhibits a high specific capacity of 9.6 F cm−2 at a current density of 2 mA cm−2 and good rate performance (55.3% retention). Furthermore, a hybrid NF@NPC@LDH//NF@PC all-solid-state supercapacitor is fabricated, and the device exhibits high energy density of 1.28 mWh cm−3 at a power density of 8.04 mW cm−3, low resistance and good cycling stability. 相似文献

Buffalo milk proteins (casein, co-precipitate or whey protein concentrate) were phosphorylated with phosphorus oxychloride (POCl3) at three different pH values (5.0, 7.0 and 9.0). The solubilities of phosphorylated milk proteins were examined over the pH range 3.0–9.0 in water and ionic (0.1 m NaCl or 10–70 mm Ca2+) systems. The solubilities of buffalo milk proteins decreased at pH 3.0, while there was an increase in the solubilities of casein and co-precipitate near their isoelectric points upon phosphorylation. Solubilities of these phosphorylated milk proteins were pH dependent in 0.1 m NaCl but there was a decrease in their solubilities with increase in calcium ion concentration. This alteration could be due to the shifting of isoionic points of phosphorylated buffalo milk proteins towards acidic pH. 相似文献

An updated and statistically-rigorous correlation is provided for crack-arrest toughness values versus normalized temperature for light-water nuclear reactor pressure vessel (RPV) steels. The database used in this effort is larger than applied heretofore and includes results from tests of laboratory-size specimens and from tests of large-scale specimens, which contain features prototypical of operating RPVs. The mathematical methodology used is based on a lognormal distribution, with its parameters calculated by orthogonal distance regression. This correlation was developed as one of several items updated for use in the US Nuclear Regulatory Commission's extensive program to evaluate and potentially revise its rule for ensuring structural integrity of operating RPVs when subjected to pressurized thermal-shock transients. 相似文献
