Left censoring or left truncation occurs when specific failure information on machines is not available before a certain age. If only the number of failures but not the actual failure times before a certain age is known, we have left censoring. If neither the number of failures nor the times of failure are known, we have left truncation. A datacenter will typically include servers and storage equipment installed on different dates. However, data collection on failures and repairs may not begin on the installation date. Often, the capture of reliability data starts only after the initiation of a service contract on a particular date. Thus, such data may exhibit severe left censoring or truncation, since machines may have operated for considerable time periods without any reliability history being recorded. This situation is quite different from the notion of left censoring in non-repairable systems, which has been dealt with extensively in the literature. Parametric modeling methods are less intuitive when the data has severe left censoring. In contrast, non-parametric methods based on the Mean Cumulative Function (MCF), recurrence rate plots, and calendar time analysis are simple to use and can provide valuable insights into the reliability of repairable systems, even under severe left censoring or truncation. The techniques shown have been successfully applied at a large server manufacturer to quantify the reliability of computer servers at customer sites. In this discussion, the techniques will be illustrated with actual field examples.  相似文献

A brown mesoporous TiO2‐x/MCF composite with a high fluorine dopant concentration (8.01 at%) is synthesized by a vacuum activation method. It exhibits an excellent solar absorption and a record‐breaking quantum yield (Φ = 46%) and a high photon–hydrogen energy conversion efficiency (η = 34%,) for solar photocatalytic H2 production, which are all higher than that of the black hydrogen‐doped TiO2 (Φ = 35%, η = 24%). The MCFs serve to improve the adsorption of F atoms onto the TiO2/MCF composite surface, which after the formation of oxygen vacancies by vacuum activation, facilitate the abundant substitution of these vacancies with F atoms. The decrease of recombination sites induced by high‐concentration F doping and the synergistic effect between lattice Ti3+–F and surface Ti3+–F are responsible for the enhanced lifetime of electrons, the observed excellent absorption of solar light, and the photocatalytic production of H2 for these catalysts. The as‐prepared F‐doped composite is an ideal solar light‐driven photocatalyst with great potential for applications ranging from the remediation of environmental pollution to the harnessing of solar energy for H2 production.  相似文献

There has been increasing interest in finding natural antioxidants to prevent free radical damage and retard the progress of chronic inflammatory diseases. Our previous data demonstrated the strong antioxidant properties of polyphenolics in Mamao seed (MS) and Mamao marc (MM) extracts. In this study we further investigated the effect of MS and MM polyphenolics on hydrogen peroxide (H2O2)-induced apoptosis and tumour promoter 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced inflammation, using human breast epithelial (MCF10A) cells. MS and MM extracts conferred dose-dependent protection against H2O2-induced apoptosis by inhibiting PARP/caspase-3 cleavage, inducing anti-apoptotic Bcl-2 expression, and down-regulating pro-apoptotic Bax. Moreover, MS and MM polyphenolics inhibited TPA-induced COX-2 and NF-κB activation by blocking the degradation of cytoplasmic IκBα, as well as subsequent nuclear translocation of p65 and attenuation of the activation of ERK, but not JNK and p38. These data establish the molecular mechanism for the anti-apoptotic and anti-inflammatory effects of MS and MM polyphenolics.  相似文献

The combination of natural products with standard chemotherapeutic agents offers a promising strategy to enhance the efficacy or reduce the side effects of standard chemotherapy. Doxorubicin (DOX), a standard drug for breast cancer, has several disadvantages, including severe side effects and the development of drug resistance. Recently, we reported the potential bioactive markers of Australian propolis extract (AP-1) and their broad spectrum of pharmacological activities. In the present study, we explored the synergistic interactions between AP-1 and DOX in the MCF7 breast adenocarcinoma cells using different synergy quantitation models. Biochemometric and metabolomics-driven analysis was performed to identify the potential anticancer metabolites in AP-1. The molecular mechanisms of synergy were studied by analysing the apoptotic profile via flow cytometry, apoptotic proteome array and measuring the oxidative status of the MCF7 cells treated with the most synergistic combination. Furthermore, label-free quantification proteomics analysis was performed to decipher the underlying synergistic mechanisms. Five prenylated stilbenes were identified as the key metabolites in the most active AP-1 fraction. Strong synergy was observed when AP-1 was combined with DOX in the ratio of 100:0.29 (w/w) as validated by different synergy quantitation models implemented. AP-1 significantly enhanced the inhibitory effect of DOX against MCF7 cell proliferation in a dose-dependent manner with significant inhibition of the reactive oxygen species (p < 0.0001) compared to DOX alone. AP-1 enabled the reversal of DOX-mediated necrosis to programmed cell death, which may be advantageous to decline DOX-related side effects. AP-1 also significantly enhanced the apoptotic effect of DOX after 24 h of treatment with significant upregulation of catalase, HTRA2/Omi, FADD together with DR5 and DR4 TRAIL-mediated apoptosis (p < 0.05), contributing to the antiproliferative activity of AP-1. Significant upregulation of pro-apoptotic p27, PON2 and catalase with downregulated anti-apoptotic XIAP, HSP60 and HIF-1α, and increased antioxidant proteins (catalase and PON2) may be associated with the improved apoptosis and oxidative status of the synergistic combination-treated MCF7 cells compared to the mono treatments. Shotgun proteomics identified 21 significantly dysregulated proteins in the synergistic combination-treated cells versus the mono treatments. These proteins were involved in the TP53/ATM-regulated non-homologous end-joining pathway and double-strand breaks repairs, recruiting the overexpressed BRCA1 and suppressed RIF1 encoded proteins. The overexpression of UPF2 was noticed in the synergistic combination treatment, which could assist in overcoming doxorubicin resistance-associated long non-coding RNA and metastasis of the MCF7 cells. In conclusion, we identified the significant synergy and highlighted the key molecular pathways in the interaction between AP-1 and DOX in the MCF7 cells together with the AP-1 anticancer metabolites. Further in vivo and clinical studies are warranted on this synergistic combination.  相似文献

In order to improve the capacity of hydrogen storage using activated carbon nanofibers, metal and fluorine were introduced into the activated carbon nanofibers by electrospinning, heat treatment, and direct fluorination. The pore structure of the samples was developed by the KOH activation process and investigated using nitrogen isotherms and micropore size distribution. The specific surface area and total pore volume approached 2800 m2/g and 2.7 cc/g, respectively. Because of the electronegativity gap between the two elements (metal and fluorine), the electron of a hydrogen molecule can be attracted to one side. This reaction effectively guides the hydrogen molecule into the carbon nanofibers. The amount of hydrogen storage was dramatically increased in this metal–carbon–fluorine system; hydrogen content was as high as 3.2 wt%.  相似文献

Optical glasses used in a range of industrially important optoelectronic devices must be polished to nano-level roughness for proper device operation. Polishing process with magnetic compound fluid slurry (MCF polishing) under a rotary magnetic field is an influential candidate for the method to precisely polish optical glass. MCF slurry has been successfully utilized to polish a variety of materials, ranging from soft optical polymers to hard optical glasses. MCF was developed by mixing a magnetic fluid and a magnetorheological fluid with the same base solvent, and hence includes not only μm-sized iron particles but also nm-sized magnetite particles. To elucidate the behaviour of material removal in MCF polishing, this study measured the normal and shear forces generated in the polishing zone during polishing. From these measurements, the distributions of pressure P and shear stress τ were obtained. The distribution of material removal rate (MRR) was investigated through spot polishing of borosilicate glass. The effects of three process parameters, namely magnet revolution speed, MCF carrier rotational speed and working gap, on pressure P, shear stress τ and the MRR were also investigated. The results revealed that P is higher near the centre of the interacting area (i.e. the polishing spot centre) and the point of maximum shear stress τ appears at about 5 mm from the polishing spot centre. All of P, τ and MRR are sensitive to MCF carrier rotational speed and working gap but insensitive to magnet revolution speed. Shear stress is more sensitive to these process parameters than the pressure. Cross-sectional profiles of the polishing spots exhibit a characteristic symmetric W-shape; material removals are minimal at the spot centre and maximal at approximately 8.2–10.2 mm from the spot centre depending on the process parameters. MRR is proportional to the MCF carrier rotational speed and is negatively correlated with working gap. An MRR model involving both the pressure and shear stress in MCF polishing is proposed. In the model, MRR is more dominated by shear stress than by pressure.  相似文献
