Fermentable fibers such as psyllium increase volatile fatty acid (VFA) concentrations in the lower digestive tract and increase the gastrointestinal tract (GIT) mass of many mammals. We reasoned that psyllium inclusion in milk replacer might produce similar effects in neonatal dairy calves, which could lead to improved growth and health. Male Holstein calves were fed a milk replacer (22% crude protein, 20% fat) either without or with psyllium (1.1% of dry matter, DM) from 2 d through 28 d of age. Milk replacer was reconstituted to 12.5% DM and fed at 12% of calf body weight, adjusted weekly. Water was offered ad libitum but no starter was fed. Three calves per treatment were harvested weekly to sample digesta from the reticulo-rumen, abomasum, jejunum, proximal colon, and distal colon, and to determine length and mass of GIT components. Psyllium in milk replacer increased the proportion of butyrate in reticulo-rumen contents from 2.4 to 3.2% of total but did not affect total VFA concentrations. Total VFA concentrations were very low in the jejunum but psyllium tended to increase total VFA, acetate, and valerate concentrations; valerate accounted for 15.9 and 16.7% of total VFA (molar basis) for control and psyllium calves, respectively. Psyllium increased total VFA concentrations in the proximal and distal colon by 104.4 and 45.6%, respectively, but had little effect on the profile of VFA. Psyllium in milk replacer increased populations of bifidobacteria (from 9.7 to 10.3 log10 cfu/g of DM) and lactobacilli (from 8.2 to 9.4 log10 cfu/g of DM) in the reticulo-rumen, but did not affect populations in jejunum or colon. Calves fed psyllium had 12.0% greater total GIT mass and 9.4% greater GIT as a percentage of body weight. Psyllium tended to increase mass of the reticulo-rumen and significantly increased mass of duodenum (34.2%), jejunum (14.5%), and colon (14.6%). Density of intestinal tissues from calves fed psyllium-supplemented milk replacer was 25.9% greater in the jejunum and 25.3% greater in the ileum, and tended to be greater in duodenum and colon than tissue from control calves. Supplementation of psyllium to milk replacer increased fermentation in the colon, mass of the total GIT, and populations of bifidobacteria and lactobacilli in the reticulo-rumen.  相似文献

The objective of the study was to evaluate the effect of diets supplemented with fatty acids of different degrees of saturation, in the absence or presence of an antioxidant (AOX; Agrado Plus, Novus International Inc., St. Charles, MO), on dairy cow lactation performance. Calcium salts of long-chain fatty acids were supplemented as a source of lower saturation fatty acid, and a palm acid product was supplemented as the higher saturation fatty acid source. Sixty early-lactation Chinese Holstein cows (100 ± 23 d in milk) were randomly allocated to 4 dietary treatments in a 2 × 2 factorial design: (1) lower saturation fatty acid (LS), (2) LS and AOX, (3) higher saturation fatty acid (HS), and (4) HS and AOX. The Ca salts of long-chain fatty acids and palm acid product were supplied at 1.8 and 1.5% on a dry matter basis, respectively, to form isoenergetic diets. The AOX was added at 0.025% in the ration. The experiment lasted 9 wk, including 1 wk for adaptation. Lactation performance was recorded and milk was sampled and analyzed weekly. Blood samples were taken from the coccygeal vein to determine metabolism parameters on d 16, 36, and 56 during the experiment. Neither fatty acid type nor AOX supplementation showed a significant effect on dry matter intake during the study. Milk yield was lower in the LS-fed cows compared with the cows fed HS. Milk fat and milk protein concentrations were not affected by fatty acid type or AOX supplementation. Adding AOX increased the yield of milk in the LS-fed cows, but did not affect those fed HS. Activity of plasma superoxide dismutase was significantly lower, plasma glucose tended to be lower, and plasma malondialdehyde was higher in the LS-fed animals compared with those fed HS. Addition of AOX decreased both plasma nonesterified fatty acids and hydrogen peroxide contents and increased total antioxidant capacity across the fatty acid types. Plasma β-hydroxybutyrate was not affected by fatty acid type or AOX treatment. Cows fed LS had higher cis-9 C18:1 and trans-10, cis-12 C18:2 in milk at the expense of C18:0, whereas AOX addition increased milk cis-9 C18:1 at the expense of milk C12:0, C16:0, and trans-10, cis-12 C18:2. It is inferred that feeding LS resulted in inferior lactation performance, whereas addition of antioxidant partially alleviated these negative effects.  相似文献

Progesterone pharmacokinetics were analyzed for plasma hormone concentrations ranging from linear to saturated metabolism in lactating Holstein cows with differing daily milk yields. The adequacy of 2-coupled first-order (bi-exponential equation), hyperbolic (Michaelis-Menten equation), and sigmoidal (Hill equation) kinetic models to describe the experimental progesterone pharmacokinetic profiles was examined on a statistical basis. After nonlinear regression and statistical analysis of the data-fitting capability, a simple one-compartment model based on Hill equation proved to be most adequate. This model indicates an enzyme-catalyzed metabolism of progesterone involving cooperative substrate-binding sites, resulting from allosteric effects that yield a sigmoidal saturation rate curve. Kinetic parameters were estimated for 2 groups of lactating Holstein cows with different daily milk yields. We found, for the first time, a remarkable quantitative agreement of the Hill coefficient value with that reported in pharmacokinetic studies involving cytochrome P450, family 3, subfamily A (CYP3A)-mediated reactions in other mammals, humans included. It seems that positive cooperativity makes enzymes much more sensitive to plasma progesterone concentration, and their activities can undergo significant changes in a narrow range of concentration as characteristic of sigmoidal behavior. Therefore, the values of classical pharmacokinetic parameters, such as the elimination constant, half-life, and clearance rate, were found to be highly dependent on the plasma progesterone concentration.  相似文献

Udder cleft dermatitis (UCD) is an inflammatory skin condition affecting the anterior parts of the udder of dairy cows. The lesions may present as mild or severe skin lesions and have been associated with mastitis and digital dermatitis. The full etiology and pathogenesis are not understood and no large-scale studies have investigated prevalence and risk factors. Therefore, the main objectives of the study were to investigate the prevalence of mild and severe UCD in Swedish dairy herds and to identify risk factors associated with such lesions. We also wanted to investigate risk factors for all cases of UCD and to determine whether UCD increases the risk for mastitis and culling. A random sample of 100 freestall dairy herds were included in the study, and each herd was visited once. Cows were registered as having no, mild, or severe UCD. Additional cow and herd data were obtained via observations, interviews, and the Swedish Official Milk Recording Scheme. The data were analyzed using logistic regression models to identify risk factors for mild and severe UCD. In total, data from 3,479 cows in 99 herds were analyzed. The prevalence of mild and severe UCD was 19 and 9%, respectively. Lesions were found in 98 of 99 herds but the within-herd prevalence of mild (0–43%) and severe (0–33%) UCD varied notably between herds. Breed (Swedish Red compared with Swedish Holstein), certain udder conformation traits, and higher parity were risk factors associated with increased risk of UCD. In addition, cows with hock lesions and cows in herds with high incidence of culling due to hoof and leg diseases had a higher risk for mild UCD. More days in milk and high milk yield were cow-related risk factors associated with severe UCD. Three housing-related factors (shorter cubicles, mattress as cubicle base, and cubicles installed before 2001 compared with 2001–2005), a high incidence of veterinary-treated clinical mastitis and culling due to udder diseases, and a low incidence of culling of first-parity cows in early lactation were herd-related risk factors associated with increased risk for severe UCD. In addition, cows in herds with a high proportion of heifers older than 17 mo that were not inseminated were associated with lower risk of all UCD. Finally, UCD was not associated with the outcomes milk somatic cell count, veterinary-treated clinical mastitis, or culling in the multivariable analyses. The etiology of UCD is most likely multifactorial, involving udder conformation traits and other cow-related risk factors as well as herd-related risk factors. The high prevalence of severe UCD lesions in Swedish dairy cows emphasizes the need for preventive measures and efficient treatments.  相似文献

Multiple births or twinning in cattle is a naturally occurring reproductive phenomenon. For dairy cattle, twinning is considered a detrimental trait as it can be harmful to cow and calf as well as costly to the producer. The objective of this study was to examine recent US calving records for the Holstein breed to determine a current estimate of heritability for twinning rate along with effects of season and parity. Two models were used in this study: a linear sire model and a binary threshold-logit sire model. Both were mixed models considering fixed effects and random effects. Analyses were conducted using a restricted maximum likelihood method. Heritability estimates were 0.0192 ± 0.0009 and 0.1420 ± 0.0069 for the linear and threshold models, respectively. Repeatabilities from the linear and threshold-logit models were 0.0443 ± 0.0012 and 0.2310 ± 0.0072, respectively. The nonzero estimates of heritability indicate the potential to select against this trait for genetic improvement of Holstein cattle.  相似文献

The objective of the present study was to test the association between the presence of fluid in uterine lumen and the fertility of repeat breeder cows (RBC). Grazing Holstein cows that were clinically normal and were not pregnant after at least 3 consecutive inseminations were defined as RBC and included in the study (n = 358). A cut-point ≥2 mm of uterine lumen visible by ultrasonography was used as indicative of the presence of fluid in the uterus. The effect of this fluid on the fertility of RBC was assessed by mixed logistic regression. Twenty percent of the RBC had ≥2 mm of fluid in their uterus, and this finding was associated with a marked reduction in the odds of conception (odds ratio = 0.46). Additional intrauterine sampling revealed that 24.0% of RBC showed signs of subclinical endometritis [≥5% neutrophils in endometrial cytology (SCE)]. However, we found no agreement between the finding of uterine fluid and the diagnosis of SCE (kappa coefficient = 0.03, 95% confidence interval = ?0.07 to 0.14). In conclusion, the presence of fluid (≥2 mm) in uterine lumen, detected by ultrasonography, was associated with lower fertility in RBC, but was not related to the diagnosis of SCE. Thus, ultrasonographic evaluation is an important tool to assist in assessment of RBC.  相似文献

BACKGROUND: There remain technological deficiencies in the economical anaerobic digestion of the entire flow of flushed dairy manure on farms within cold climates. In order to develop an anaerobic digester tailored for this purpose, this study investigated the distributions of biomass and biogas generated in three particle‐size‐characterized zones of flushed dairy manure. RESULTS: Results demonstrate that comparable biogas generations are equally distributed between the solids and liquid phases of flushed dairy manure, implying that at least 50% of methane generation might be lost when utilizing existing processes that only selectively handle the liquid or solids phases. Microscopic and kinetic studies show that the majority of microorganisms in the manure are attached to the surface of undigested dairy manure fibrous solids. In particular, greater than 70% of acetate‐utilizing methanogens were found closely associated with the indigestible solids. This high microbial affinity to the dairy manure solids phase can be traced back to the selection pressure driven bacterial immobilization mechanism in cattle rumen. CONCLUSION: This study shows that dairy manure fibrous solids have the potential to act as effective natural biomass carriers for high rate and simultaneous methanation of both solids and liquid phases of flushed dairy manure in one digester. The primary objective of this study was to evaluate the repeatability of measurements of β-hydroxybutyrate (BHB) concentrations in capillary blood obtained from minimal-invasive puncturing of different sites at the external vulva skin of dairy cows. A total of 240 multiparous cows sampled once within 21 d in milk were enrolled in the study. Capillary blood samples were obtained from the right and left labium of the external vulva skin and additionally from a site close to the first puncture. Samples were analyzed by using an electronic hand-held device [FreeStyle Precison Neo (FSP-Neo), Abbott GmbH & Co., Wiesbaden, Germany)]. Additionally, samples from a coccygeal vessel were analyzed as reference in a laboratory. Mean deviations (± standard deviation) between the 3 capillary sampling sites ranged from 0.01 ± 0.14 (left vs. right labium) to 0.02 ± 0.13 mmol/L (left or right labium vs. additional site). The BHB concentrations in capillary blood compared with the reference showed a mean deviation of 0.14 ± 0.18 mmol/L. The Spearman correlation coefficient between BHB concentrations in capillary blood and coccygeal blood was 85.4%. Mixed model analyses showed no significant effect of sampling site or squeezing the skin during sampling procedures on the differences toward the reference. Based on laboratory BHB concentrations of ≥1.2 and ≥1.4 mmol/L, respectively, the prevalence of hyperketonemia was 10.4 (n = 25) and 5.0% (n = 12). Thresholds to detect hyperketonemia with the FSP-Neo were optimized by receiver operating characteristic analyses. Based on a laboratory BHB concentration of ≥1.2 mmol/L thresholds were determined as 1.1 (left labium, 2nd puncture) and 1.0 mmol/L (right labium), with sensitivities between 94 and 99% and specificities between 89 and 97%. Based on a serum BHB concentration of 1.4 mmol/L, receiver operating characteristic analyses resulted in optimized cutoffs of 1.4 mmol/L for all sampling sites at the vulva, with sensitivities of 100% and specificities between 96 and 99%. In conclusion, vulvar capillary blood is suitable for accurate measuring of BHB concentrations by using the FSP-Neo. After adjusting the thresholds, the FSP-Neo provides very good test characteristics independent of the capillary sampling site or squeezing the skin and allows a practical and reliable screening tool for dairy cows.  相似文献

We investigated the effects of bovine somatotropin (bST) on mammary gland function and composition in the declining phase of lactation in goats. Sixteen Saanen goats, 180 +/- 11 days in milk (DIM), were divided equally into control and treated groups. The treated group received 120 mg/2 wk of slow-release bST for three cycles. Milk yield, milk composition, milk clotting measures, and plasmin-plasminogen activator activities were recorded weekly. Milk Na and K were determined in individual milk samples collected weekly during the third cycle. Blood samples were collected weekly during the second cycle and the plasma analyzed for nonesterified fatty acids (NEFA), glucose, and urea. At the end of the 6 wk, three goats from each group were slaughtered, and the udders were removed. Mammary gland weight, composition, and total DNA content were determined. The histological effects of bST on mammary tissue were investigated. The analyzed parameters included numbers of alveoli, corpora amylacea, apoptotic cells, and laminin fibronectin distribution and localization. An extensive morphological analysis on the epithelial and stromal components was performed. Milk yield was significantly higher in the treated group, fat content was not affected, but protein and nonprotein nitrogen were lower in treated goats milk. Treatment with bST did not influence milk pH but reduced coagulation time. Plasmin and plasminogen activator activities were not affected. Milk K levels were higher and the Na/K ratio was lower in treated animals. Plasma glucose, NEFA, and urea were unaffected. Mammary gland weight and total DNA were higher in treated than control animals, suggesting that with advancing lactation bST treatment maintains cells. Fat, protein, and collagen content of the mammary tissue did not differ between the groups. Treatment with bST significantly increased the number of lactating alveoli (LA) and significantly reduced the number of regressing alveoli (RA) and corpora amylacea, both within and outside the alveolar lumen. Laminin and fibronectin localization were not affected, and very few apoptotic cells were found in both treated and control samples. Our findings suggest that bST administration to dairy goats in late lactation can modulate mammary gland activity and improve lactation persistency; this is associated with maintained total mammary parenchyma weight and lactating alveoli.  相似文献

Application of transrectal real-time ultrasonography as a research tool to study bovine reproduction represents a technological breakthrough that has revolutionized our understanding of reproductive biology in cattle. The widespread adoption and use of ultrasonography for routine reproductive examinations of dairy cattle by bovine practitioners is the next contribution this technology will make to the dairy industry. Assessment of pregnancy status and fetal viability early postbreeding to identify cows that fail to conceive improves reproductive efficiency by decreasing the interval between artificial insemination services and increasing artificial insemination service rate. Early identification of cows carrying twin fetuses will allow for implementation of differential management strategies to abrogate negative effects of twinning during the periparturient period once such strategies have been developed. Ovarian and uterine pathologies not accurately detected via rectal palpation can easily be visualized by ultrasound, and appropriate therapies can be implemented. Determination of fetal sex in utero is useful when coupled with a management decision that justifies the expense of fetal sexing. Development of integrated reproductive management systems that combine ultrasound with new and existing reproductive technologies will further enhance the practical applications of ultrasonography. Development of Extension education programs to train bovine practitioners to use ultrasound for routine reproductive examinations is a critical step toward rapid implementation of this technology into the dairy industry.  相似文献
