Currently, biochemical modulation for 5-fluorouracil (5-FU) is one of the most successful chemotherapy for both colo-rectal and gastric cancer. The purpose of this study is to evaluate the significance of measuring intratumoral thymidylate synthetase (TS) and folate (FH4) levels as predictive parameters for the successful treatment. Samples were collected from 16 advanced colo-rectal and 21 advanced gastric cancer. TS and tetrahydrofolate levels in the specimens were measured by binding assay. Results showed that there were no significant difference in TS levels between the different pathologic types of carcinoma. On the other hand, well (3.94 +/- 1.75 p mol/g) and moderately (5.95 +/- 2.69 p mol/g) differentiated carcinoma showed lower FH4 levels compared to poorly differentiated carcinoma (9.58 +/- 5.27 p mol/g). In conclusion, biochemical modulation by cisplatin or leucovorin, which elevates intratumoral folate levels, is more needed for well and moderately differentiated carcinoma. Finally, measuring TS levels can also be important because two cases who responded to cisplatin/5-FU chemotherapy showed low TS levels compared to the others who had lower response.  相似文献

Phorbol esters exert a dual function in human leukemia cells, induction of differentiation and activation of integrin-mediated functions. Here we have shown that the plastic adherence of phorbol ester-treated U937 cells is mediated by expression of integrin Mac-1 (CD11b/CD18) on the cell surface and that these adherent cells exhibit anoikis (apoptosis when adherent cells are detached or adherence is inhibited). We used U937-derived clones overexpressing either antisense RNAs antisense to CD11b and CD18 mRNAs or mRNA from a truncated mutant CD11b gene. We have also shown that apoptosis in non-adherent cells or anoikis was mediated by sphingosine and that survival of adherent cells was achieved by a shift of the dynamic balance between sphingosine and sphingosine 1-phosphate toward the latter by adherence-activated sphingosine 1-kinase.  相似文献

Measurements of longitudinal bunch length of subpicosecond and picosecond electron beams have been performed by three methods with three radiation sources at the 35 MeV S-band twin liner accelerators at Nuclear Engineering Research Laboratory, University of Tokyo. The methods we adopt are the femtosecond streak camera with a nondispersive reflective optics, the coherent transition radiation (CTR) Michelson interferometer and the 10 ch polychromator that detects the spectrum of CTR and coherent diffraction radiation (CDR). The measurements by the two CTR methods were independently done with the streak camera and their results were consistent with one another. As a result, the reliability of the polychromator for the diagnostics of less than picosecond electron bunch and the usefulness of the diagnostics for the single shot measurement were verified. Furthermore, perfect nondestructive diagnostics for subpicosecond bunches was performed utilizing CDR interferometry. Then the good agreement between CDR interferometry and the streak camera was obtained.  相似文献

We have measured drift velocities and Lorentz angles for xenon gas with various types of quench gas mixtures. The measurement was motivated to help designing a new type of detector for an e+e collider experiment, where synchrotron radiation X-rays generated by electrons passing through a high magnetic field are detected to identify the electrons.  相似文献

Human–human interaction consists of various nonverbal behaviors that are often emotion-related. To establish rapport, it is essential that the listener respond to reactive emotion in a way that makes sense given the speaker's emotional state. However, human–robot interactions generally fail in this regard because most spoken dialogue systems play only a question-answer role. Aiming for natural conversation, we examine an emotion processing module that consists of a user emotion recognition function and a reactive emotion expression function for a spoken dialogue system to improve human–robot interaction. For the emotion recognition function, we propose a method that combines valence from prosody and sentiment from text by decision-level fusion, which considerably improves the performance. Moreover, this method reduces fatal recognition errors, thereby improving the user experience. For the reactive emotion expression function, the system's emotion is divided into emotion category and emotion level, which are predicted using the parameters estimated by the recognition function on the basis of distributions inferred from human–human dialogue data. As a result, the emotion processing module can recognize the user's emotion from his/her speech, and expresses a reactive emotion that matches. Evaluation with ten participants demonstrated that the system enhanced by this module is effective to conduct natural conversation.  相似文献

Spatial and temporal patterns in streamwater acidity are ecologically important, but difficult to measure in parallel. Here we present the spatial distribution of streamwater chemistry relevant to acidity from 60 stream sites distributed throughout a 67 km2 boreal catchment, sampled during a period of winter baseflow (high pH) and during a spring flood episode (low pH). Sites were grouped based on pH level and pH change from winter baseflow to spring flood. The site attributes of each pH group were then assessed in terms of both stream chemistry and subcatchment landscape characteristics. Winter baseflow pH was high throughout most of the stream network (median pH 6.4), but during the spring flood episode stream sites experienced declines in pH ranging from 0-1.6 pH units, resulting in pH ranging from 4.3-6.3. Spring flood pH was highest in larger, lower altitude catchments underlain by fine sorted sediments, and lowest in small, higher altitude catchments with a mixture of peat wetlands and forested till. Wetland-dominated headwater catchments had low but stable pH, while the spring flood pH drop was largest in a group of catchments of intermediate size which contained well-developed coniferous forest and a moderate proportion of peat wetlands. There was a trend with distance downstream of higher pH, acid neutralizing capacity (ANC) and base cation concentrations together with lower dissolved organic carbon (DOC, strongly negatively correlated with pH). This apparent scale-dependence of stream chemistry could be explained by a number of environmental factors which vary predictably with altitude, catchment area and distance downstream—most notably, a shift in surficial sediment type from unsorted till and peat wetlands to fine sorted sediments at lower altitudes in this catchment. As a result of the combination of spatial heterogeneity in landscape characteristics and scale-related processes, boreal catchments like this one can be expected to experience high spatial variability both in terms of chemistry at any given point in time, and in the change experienced during high discharge episodes. Although chemistry patterns showed associations with landscape characteristics, considerable additional variability remained, suggesting that the modeling of dynamic stream chemistry from map parameters will continue to present a challenge.  相似文献
