Growing interest in extracellular vesicles (EVs) has prompted the advancements of protocols for improved EV characterization. As a high-throughput, multi-parameter, and single particle technique, flow cytometry is widely used for EV characterization. The comparison of data on EV concentration, however, is hindered by the lack of standardization between different protocols and instruments. Here, we quantified EV counts of platelet-derived EVs, using two flow cytometers (Gallios and CytoFLEX LX) and nanoparticle tracking analysis (NTA). Phosphatidylserine-exposing EVs were identified by labelling with lactadherin (LA). Calibration with silica-based fluorescent beads showed detection limits of 300 nm and 150 nm for Gallios and CytoFLEX LX, respectively. Accordingly, CytoFLEX LX yielded 40-fold higher EV counts and 13-fold higher counts of LA+CD41+ EVs compared to Gallios. NTA in fluorescence mode (F-NTA) demonstrated that only 9.5% of all vesicles detected in scatter mode exposed phosphatidylserine, resulting in good agreement of LA+ EVs for CytoFLEX LX and F-NTA. Since certain functional characteristics, such as the exposure of pro-coagulant phosphatidylserine, are not equally displayed across the entire EV size range, our study highlights the necessity of indicating the size range of EVs detected with a given approach along with the EV concentration to support the comparability between different studies.  相似文献

Considering the advent of antibiotic resistance, the study of bacterial metabolic behavior stimulated by novel antimicrobial agents becomes a relevant tool to elucidate involved adaptive pathways. Profiling of volatile metabolites was performed to monitor alterations of bacterial metabolism induced by biosynthesized silver nanoparticles (bio-AgNPs). Escherichia coli, Enterococcus faecalis, Klebsiella pneumoniae and Proteus mirabilis were isolated from pressure ulcers, and their cultures were prepared in the presence/absence of bio-AgNPs at 12.5, 25 and 50 µg mL−1. Headspace solid phase microextraction associated to gas chromatography–mass spectrometry was the employed analytical platform. At the lower concentration level, the agent promoted positive modulation of products of fermentation routes and bioactive volatiles, indicating an attempt of bacteria to adapt to an ongoing suppression of cellular respiration. Augmented response of aldehydes and other possible products of lipid oxidative cleavage was noticed for increasing levels of bio-AgNPs. The greatest concentration of agent caused a reduction of 44 to 80% in the variety of compounds found in the control samples. Pathway analysis indicated overall inhibition of amino acids and fatty acids routes. The present assessment may provide a deeper understanding of molecular mechanisms of bio-AgNPs and how the metabolic response of bacteria is untangled.  相似文献

Bimetallic catalysts have been investigated as the most efficient materials to accelerate the chemical transformations at the anode in Direct Ethanol Fuel Cells. A comparative study is presented here to synthesize Ni–Cu bimetallic nanoparticles for the ethanol oxidation reaction on three conducting polymers: poly-ortho-phenylenediamine, poly-meta-phenylenediamine, and poly-para-phenylenediamine. X-ray diffraction (XRD), Scanning Electron Microscopy (SEM), Transmission Electron Microscopy (TEM), and Electrochemical Impedance Spectroscopy (EIS) were used to analyze the modified electrodes. A series of bimetallic Ni–Cu nanoparticles with tunable ratios were successfully synthesized by simply changing the concentrations of Nickel and Copper. It has been confirmed that the best Ni/Cu molar ratio was 25% in the aspect of catalytic performance. The electrocatalyst exhibited an excellent catalytic activity with an anodic current of 70.5 mA cm?2 at the lowest onset potential of 0.39 V with impressive stability. Ni4Cu1/PpPD should be considered as a good alternative to noble metal anode catalyst.  相似文献

Here, a fluoride-assisted route for the controlled in-situ synthesis of metal nanoparticles (NPs) (i.e., AgNPs, AuNPs) on polydimethylsiloxane (PDMS) is reported. The size and coverage of the NPs on the PDMS surface are modulated with time and over space during the synthetic process, leveraging the improved yield (10×) and faster kinetics (100×) of NP formation in the presence of F ions, compared to fluoride-free approaches. This enables the maskless preparation of both linear and step gradients and patterns of NPs in 1D and 2D on the PDMS surface. As an application in flexible plasmonics/photonics, continuous and step-wise spatial modulations of the plasmonic features of PDMS slabs with 1D and 2D AgNP gradients on the surface are demonstrated. An excellent spatially resolved tuning of key optical parameters, namely, optical density from zero to 5 and extinction ratio up to 100 dB, is achieved with AgNP gradients prepared in AgF solution for 12 minutes; the performance are comparable to those of commercial dielectric/interference filters. When used as a rejection filter in optical fluorescence microscopy, the AgNP-PDMS slabs are able to reject the excitation laser at 405 nm and retain the green fluorescence of microbeads (100 µm) used as test cases.  相似文献

This study investigates the effect of two different iron compounds (zero-valent iron nanoparticle: nZVI and iron oxide nanoparticles: nIO) and pH on fermentative biohydrogen production from molasses-based distillery wastewater. The nZVI and nIO of optimum particle sizes of 50 nm and 55 nm respectively were synthesized and applied for fermentative hydrogen (H2) production. The addition of nIO & nZVI at (0.7 g/L, pH: 6) resulted in the highest H2 yield, H2 production rate, H2 content and COD reduction. Moreover, the kinetic parameters of H2 production potential (P) and H2 production rate (Rm) increased to 387 mL, and 22.2 mL/h, respectively for nZVI, these values were 363 mL and 21.8 mL/h for nIO. The results obtained indicated the positive effect of nZVI and nIO addition on enhanced fermentative H2 production. The addition of nZVI & nIO resulted in 71% and 69.4% enhancement in biohydrogen production respectively.  相似文献

添加促进剂是高效制备水合物的研究热点,促进剂及其添加量对水合物生成效果至关重要。本文主要从热力学促进剂和动力学促进剂两大类型进行分析:总结了可溶水相热力学促进剂和不可溶水相热力学促进剂的浓度对水合物形成相平衡的影响;从表面活性剂、纳米粒子和相变材料等添加剂,阐述了动力学促进剂添加量对水合物生成诱导时间、储气量和生成速率等方面的影响。促进剂都存在最佳浓度的添加量,并且不同类型促进剂复配更有利于水合物生成。目前添加促进剂后水合物形成机理的研究大多从宏观现象推测,部分学者通过拉曼光谱、X射线衍射和显微观察探索分析促进剂对水合物形成的微观影响,这方面研究有待于进一步开展。添加量作为水合物形成促进剂的重要指标,研究者应得到水合物形成促进剂的最佳浓度与所研究对象的关联性。  相似文献

光降解乙醛(CH3CHO)是一种新型高效的乙醛去除方法,通常采用TiO2作为光催化剂。然而TiO2对乙醛吸附能力较弱,对产物的选择性较低,电子-空穴对重组率较高,严重限制了对乙醛的降解性能。本研究通过在TiO2上负载CuAg纳米粒子(CuAg/TiO2),成功构建了高效稳定的光催化降解乙醛催化剂,有效解决了TiO2的固有缺陷。在自然光照射下,CuAg/TiO2对乙醛的降解率高达42.49%。连续4轮全光谱光催化降解乙醛,CuAg/TiO2活性均保持在98.89%以上。进一步的机理研究表明,CuAg/TiO2中的CuAg纳米粒子在光照下产生热电子,随后热电子转移到TiO2和吸附在Ag位点上的氧中。CuAg/TiO2上生成的超氧自由基能有效地降解乙醛,从而在乙醛降解过程中表现出优异的性能。  相似文献

The purpose of this paper is to give process for preparing monodispersed silver particles with round shape having aspect ratio of 1, because the shape is suitable for preparing silver grids with fine pattern size. We found that the combination of gelatin and hydrazine gave the monodispersed silver particles with the aspect ratio of 1. Presence of the high molecular compounds is crucial probably because they are adsorbed on the surface of growing silver particles and control the uniform crystal growth. In addition, the relationship between these reaction conditions and the particle shape are discussed in detail.  相似文献
