Tumor cell aggregation is critical for cell survival following the loss of extracellular matrix attachment and dissemination. However, the underlying mechanotransduction of clustering solitary tumor cells is poorly understood, especially in non-small cell lung cancers (NSCLC). Here, we examined whether cell surface protrusions played an important role in facilitating the physical contact between floating cells detached from a substrate. We employed poly-2-hydroxyethyl methacrylate-based 3D culture methods to mimic in vivo tumor cell cluster formation. The suprastructural analysis of human NSCLC A549 cell spheroids showed that finger-like protrusions clung together via the actin cytoskeleton. Time-lapse holotomography demonstrated that the finger-like protrusions of free-floating cells in 3D culture displayed exploratory coalescence. Global gene expression analysis demonstrated that the genes in the organic hydroxyl transport were particularly enriched in the A549 cell spheroids. Particularly, the knockdown of the water channel aquaporin 3 gene (AQP3) impaired multicellular aggregate formation in 3D culture through the rearrangement of the actomyosin cytoskeleton. Moreover, the cells with reduced levels of AQP3 decreased their transmigration. Overall, these data indicate that cell detachment-upregulated AQP3 contributes to cell surface protrusions through actomyosin cytoskeleton remodeling, causing the aggressive aggregation of free-floating cells dependent on the property of the substratum and collective metastasis.  相似文献

Many astonishing biological collective behaviors exist in nature, and artificial microrobotic swarms have been developed by emulating these scenarios. However, these microswarms typically have single structures and lack the adaptability that many biological swarms exhibit to thrive in complex environments. Inspired by viscoelastic fire ant aggregations and using a combination of experiment and simulation, a strategy to trigger ferrofluid droplets into forming microswarms exhibiting both liquid-like and solid-like behaviors is reported. By spatiotemporally programming an applied magnetic field, microswarms can be liquefied to implement reversible elongation with a high aspect ratio and solidified into entireties to perform overturning and bending behaviors. It is demonstrated that reconfigurability enables the microswarm to be a mobile dexterous micromanipulator, acting not only as a soft “octopus arm” to explore a confined environment and grasp a targeted object but also adaptively navigate multiple terrains, such as uneven surfaces, curved grooves, complex mazes, high steps, narrow channels, and even wide gaps. This microrobotic swarm can reconfigure both shapes and tasks based on the demands of the environment, presenting novel solutions for a variety of applications.  相似文献

Dairy cows have a daily pattern of feed intake which influences ruminal fermentation and nutrient absorption. Milk synthesis also exhibits a daily rhythm and is altered by the timing of feed availability. Nutrients can regulate physiological rhythms, but it is unclear which specific nutrients affect the rhythms of milk synthesis in the cow. The objective of this study was to determine the effect of the timing of acetate infusion on the daily rhythms of feed intake, milk synthesis, milk fatty acids, plasma insulin and metabolites, and core body temperature. Ten lactating ruminally cannulated Holstein cows (127 ± 24.6 d in milk; mean ± standard deviation) were arranged in a 3 × 3 Latin square design. Treatments were ruminal infusions of 600 g/d of acetate either continuously throughout the day (CON) or over 8 h/d during the day (day treatment, DT; 0900 to 1700 h) or the night (night treatment, NT; 2100 to 0500 h). Experimental periods were 14 d with a 7-d washout between periods. Cows were milked every 6 h during the final 7 d of each experimental period to determine the daily pattern of milk synthesis. Blood samples were taken to represent every 4 h across the day and plasma glucose, insulin, β-hydroxybutyrate, urea nitrogen, and acetate concentration were measured. An intravaginal temperature logger was used to measure core body temperature. Data were analyzed with cosinor-based rhythmometry to test the fit of a cosine function with a period of 24 h and to determine the acrophase (time at peak) and amplitude (peak to mean) of each rhythm. Milk yield fit a daily rhythm for all treatments and DT and NT phase-delayed the rhythm and DT increased the robustness of the rhythm. Milk protein concentration fit a daily rhythm for all treatments and DT increased robustness, whereas NT phase-delayed the rhythm. Plasma acetate concentration also fit a daily rhythm in all treatments. Plasma acetate peaked at ~1600 h in CON and DT and at 0053 h in NT, reflecting the timing of treatment infusions. There was a daily rhythm in plasma β-hydroxybutyrate that reflected the plasma acetate rhythm. Core body temperature fit a rhythm for all treatments, but the amplitude of the rhythm was smaller than previously observed. In conclusion, the timing of acetate infusion influences peripheral rhythms of milk synthesis and plasma metabolites.  相似文献

The creative city approach is going through a redefinition after the 2008 global financial crisis. In the specific case of South-European cities, in the context of austerity and cuts in public investment, creativity is becoming a strategy for achieving maximum social benefit and improvement of the built environment with minimum economic expenditure. This paper looks at this redefinition of creativity through the case study of Seville, in southern Spain. Through research methods that include video-recorded testimonies of the actors involved, mapping at the online platform “Laboratorio Q”, and public engagement activities, this paper explores how the civic society, professional, and public authorities have reinvented how to produce collective spaces. The paper concludes that bottom-up creative processes for producing collective spaces have become more visible since the 2008 crisis, when architects, planners, public authorities and policy-makers have been “learning” from them.  相似文献

Animal groups in nature often display an enhanced collective information-processing capacity. It has been speculated that natural selection will tune this response to be optimal, ensuring that the group is reactive while also being robust to noise. Here, we show that this is unlikely to be the case. By using a simple model of decision-making in a dynamic environment, we find that when individuals behave rationally and are subject to selection based on their accuracy, optimality of collective decision-making is not attained. Instead, individuals overly rely on social information and evolve to be too readily influenced by their neighbours. This is due to a classic evolutionary conflict between individual and collective interest. The result is a sub-optimal system that is poised on the cusp of total unresponsiveness. Individuals in the evolved group exhibit delayed reactions to changes in the environment, before responding with rapid, socially reinforced transitions, reminiscent of familiar human and animal social systems (markets, stampedes, fashions, etc.). Our results demonstrate that behaviour of this type may not be pathological, but instead could represent an evolutionary attractor for such collective systems.  相似文献

Reflective judgement is crucial for medical-related practitioners in dealing with controversial issues. However, the conformity phenomenon is likely to occur and interfere with reflective judgement learning during interactive activities. Effective strategies are required to moderate the conformity behaviour tendency (CBT) and improve reflective judgement performance (RJP). This study demonstrates two significant results: (a) Compared with the guided self-reflection learning strategy, the online collective reflection (OCR) learning strategy effectively weakened the learners' general CBT while dealing with professional controversial issues; and (b) a significantly negative correlation between the RJP achieved and the change of CBT in online environment was detected in the OCR group. The implications and potential applications in higher education were discussed. Further studies are needed to confirm the long-term effects and the extending application to other professional studies.  相似文献

In this work, gas–liquid distributions in an industrial centrifugal pump operated at various steady state conditions have been quantitatively determined. Therefore, high-resolution gamma-ray computed tomography (HireCT) has been applied, operated in time-averaging rotation-synchronized CT scanning mode. Detailed studies have been performed on a hydraulic test facility providing authentic operating conditions for industrial centrifugal pumps. The gas phase distribution in the centrifugal pump has been studied for swirling gas–liquid two-phase flow as an inlet flow regime and at defined inlet gas volumetric flow rates between 0% and 5%. In this way, the influence of the inlet flow boundary conditions on the performance of the centrifugal pump as well as the gas holdup distribution within the impeller region could be successfully determined.  相似文献

为全面掌握某钢厂帘线钢在非稳态浇注工况下的洁净度水平,为生产制定合理的头尾坯长度以及进行产品分级管理提供依据,对非稳态浇注和稳态浇注工况下的连铸坯分别取样,对比分析其洁净度差异,并通过水力学试验模拟分析了中间包充包过程中的渣-钢界面行为,进而解释了造成铸坯洁净度差异的原因。研究结果表明,非稳态浇注工况下帘线钢铸坯的洁净度明显低于稳态工况,其中T.O质量分数是稳态浇注工况的1.76倍,N质量分数是稳态浇注工况的1.23倍;两种工况下的显微夹杂物分别为52.89个/mm2和26.1个/mm2,大型夹杂物分别是11.49 mg/10 kg钢、4.36 mg/10 kg钢。充包时钢液裸露、二次氧化和卷渣是造成非稳态浇注时铸坯洁净度差的主要原因。  相似文献
