Growing interest in extracellular vesicles (EVs) has prompted the advancements of protocols for improved EV characterization. As a high-throughput, multi-parameter, and single particle technique, flow cytometry is widely used for EV characterization. The comparison of data on EV concentration, however, is hindered by the lack of standardization between different protocols and instruments. Here, we quantified EV counts of platelet-derived EVs, using two flow cytometers (Gallios and CytoFLEX LX) and nanoparticle tracking analysis (NTA). Phosphatidylserine-exposing EVs were identified by labelling with lactadherin (LA). Calibration with silica-based fluorescent beads showed detection limits of 300 nm and 150 nm for Gallios and CytoFLEX LX, respectively. Accordingly, CytoFLEX LX yielded 40-fold higher EV counts and 13-fold higher counts of LA+CD41+ EVs compared to Gallios. NTA in fluorescence mode (F-NTA) demonstrated that only 9.5% of all vesicles detected in scatter mode exposed phosphatidylserine, resulting in good agreement of LA+ EVs for CytoFLEX LX and F-NTA. Since certain functional characteristics, such as the exposure of pro-coagulant phosphatidylserine, are not equally displayed across the entire EV size range, our study highlights the necessity of indicating the size range of EVs detected with a given approach along with the EV concentration to support the comparability between different studies.  相似文献

采用有限元模拟和实验研究了挤压钛合金弯曲管件。通过实验验证了工件的形状和尺寸精度,并通过有限元模拟分析了工艺参数对挤出过程中变形体的平均压应力分布情况和挤出弯管件的曲率半径的影响规律。结果表明:有限元模拟中,弯管件的曲率半径误差为6.03%,弯管直径误差为3.82%;在靠近定径带处,平均压应力呈非均匀分布;在焊合腔内,靠近细分流孔区域的平均压应力小于靠近粗分流孔区域的平均压应力,平均压应力的大小顺序在通过粗、细分流孔前后相反;在模具结构固定不变时,弯管件的曲率半径随挤压速度的减小而增大,不随挤压温度的变化而变化。  相似文献

Liquid-phase adsorption has hardly been established in micro-flow, although this constitutes an industrially vital method for product separation. A micro-flow UV-photo isomerization process converts cis-cyclooctene partly into trans-cyclooctene, leaving an isomeric mixture. Trans-cyclooctene adsorption and thus separation was achieved in a fixed-bed micro-flow reactor, packed with AgNO3/SiO2 powder, while the cis-isomer stays in the flow. The closed-loop recycling-flow has been presented as systemic approach to enrich the trans-cyclooctene from its cis-isomer. In-flow adsorption in recycling-mode has hardly been reported so that a full theoretical study has been conducted. This insight is used to evaluate three process design options to reach an optimum yield of trans-cyclooctene. These differ firstly in the variation of the individual residence times in the reactor and separator, the additional process option of refreshing the adsorption column under use, and the periodicity of the recycle flow.  相似文献

凌庄子水厂蓄水池进水口处有一保水堰,为非标准薄壁堰,不能使用已有堰流公式对其过流量进行准确计算。为了得出较为精确的过流流量,按照重力相似准则制作几何比尺为1∶5的模型进行试验研究。在已有自由出流公式的基础上,对自由出流流量系数进行修正并对淹没情况下流量变化过程进行研究。对该非标准堰自由出流流量系数的实测值与经验值进行分析比较,发现堰板槽降低了实际自由出流过流能力。淹没出流的流量系数主要与下游尾水位有关,试验中形成的淹没式堰流受实际堰型尺寸影响,下游尾水位和堰上水位近似相等,不完全适用已有淹没出流流量公式,通过试验给出了修正淹没系数随h/p的变化关系。结果表明利用堰前、堰后水位初步计算过流流量是可行的,可为该工程提供参考,也可为实际工程中非标准矩形堰的流量计算提供思路。  相似文献

Recently, researchers have devoted more attention to supercapacitors (SCs) to integrate with batteries in energy storage systems (ESSs) for vehicle applications. In this study, we attempted to characterize the use of SCs in the ESS for a PEM fuel cell vehicle equipped with an alternator to maximize the performance of regenerative braking. We applied lithium-ion batteries (LIBs) and SCs as energy storage devices to examine their effect on ESS. Then we used a hysteresis brake to apply controllable braking force on the flywheel to form hybrid braking (HB) and made efforts to study its behavior to suggest a braking control strategy. We also ran the whole system over the rotational speed to cover the range of driving speed. At last, we sized the SCs for the most commonly used fuel cell electric vehicle (FCEV) in Korea, i.e., Hyundai NEXO, based on the results obtained from the above study by alternator efficiencies.  相似文献

Subsurface hydrogen (H2) storage in geological formations is of growing interest for decarbonization. However, there is a knowledge gap in understanding the multiphase flow involved in this process, which can have a significant impact on the recovery performance of H2. Therefore, a full-compositional modeling study was conducted to analyze potential issues and to understand the fundamental hydrodynamic mechanisms of H2 storage. We performed a range of 2D vertical simulations at the decametre scale with a very fine cell size (0.1 m) to observe the detailed flow behaviour of H2 with carbon dioxide (CO2) as cushion gas in various flow regimes. Issues such as viscous instability, capillary bypassing, gas trapping and gravity segregation are analysed here. To generalize our calculations, we have validated and applied the scaling theory in the context of subsurface H2 storage. Since this study is focused on the hydrodynamic behaviour, three dimensionless groups, including aspect factor, capillary/viscous ratio and gravity/viscous ratio were identified to correlate recovery performance between various scales in a fixed heterogeneous system. It was found that H2 could infiltrate the cushion gas in the proximity of the injectors, meaning that CO2 is not displaced away from the injectors in a piston-like fashion. As a result, the purity of the back produced H2 is much degraded, particularly in a viscous-dominated scenario. On the other hand, the injected H2 mostly accumulates at the top forming a highly restricted mixing zone with CO2 in the gravity-dominated case. The recovery performance is therefore much improved in this case. Although the gas distribution can be significantly altered by capillary forces leading to bypassed zones, the recovery performance of H2 is hardly influenced. This is because the back-produced H2 recovery is not dependent on the sweep efficiency of the gas. H2 can be back produced following the same paths which were formed during injection.  相似文献

A series of novel branched sulfonated polyimide (bSPI-x) membranes with 8% branched degree are developed for application in vanadium redox flow battery (VRFB). The sulfonation degrees of bSPI-x membranes are precisely regulated for obtaining excellent comprehensive performance. Among all bSPI-x membranes, the bSPI-50 membrane shows strong vanadium permeability resistance, which is as 8 times as that of commercial Nafion 212 membrane. At the same time, the bSPI-50 membrane has remarkable proton selectivity, which is four times as high as that of Nafion 212 membrane. The bSPI-50 membrane possesses slower self-discharge speed than Nafion 212 membrane. Furthermore, the bSPI-50 membrane achieves stable VRFB efficiencies during 200-time charge-discharge cycles at 120–180 mA cm?2. Simultaneously, the bSPI-50 membrane exhibits excellent capacity retention compared with Nafion 212 membrane. All results imply that the bSPI-50 membrane possesses good application prospect as a promising alternative separator of VRFB.  相似文献

Oblivious polynomial evaluation (OPE) is a two-party protocol that allows a receiver, R to learn an evaluation f(α), of a sender, S's polynomial f(x), whilst keeping both α and f(x) private. This protocol has attracted a lot of attention recently, as it has wide ranging applications in the field of cryptography. In this article we review some of these applications and, additionally, take an in-depth look at the special case of information theoretic OPE. Specifically, we provide a current and critical review of the existing information theoretic OPE protocols in the literature. We divide these protocols into two distinct cases (three-party and distributed OPE) allowing for the easy distinction and classification of future information theoretic OPE protocols. In addition to this work, we also develop several modifications and extensions to existing schemes, resulting in increased security, flexibility and efficiency. Lastly, we also identify a security flaw in a previously published OPE scheme.  相似文献

为缓解我国水、能源和粮食资源紧张问题,促进资源可持续利用,构建水-能源-粮食系统,利用耦合协调度模型对我国的30个省(自治区、直辖市)进行测算,并利用空间杜宾模型分析主要影响因素。结果表明:2003—2017年,我国能源、粮食评价[JP]指数高于水资源评价指数,系统综合评价指数逐年递增;大部分省份耦合协调度处于初级协调水平且呈现逐年上升的态势,个别省份耦合协调度濒临失调;耦合协调度空间自相关性较强,虽有明显波动,但是呈现逐年加强的态势;影响耦合协调度的主要因素有从业人口数、固定资产投资额、人均生产总值、人口总数、[JP]文盲人口占比、工业污染排放、城镇化。  相似文献
