The effects of key parameters on the preparation and regeneration of protoplast from the β-carotene-producing fungus Blakeslea trispora were discussed in this paper, including the combination of various enzymes, mycelial age, digesting time and temperature, pH value, osmotic stabilizers, pretreatment, culture medium and culture method. Under the condition of mixed enzymes in osmotic stabilizer (0.6 M NaCl) combined with 2% lysozyme, 3% cellulase and 3% snailase, the highest protoplast yield, as high as 7.48×106 protoplasts/mL, was obtained when mycelial age was 60 h at pH 5.0–6.0 with digesting for 14–16 h at 28 °C. After purification of the obtained protoplasts, they were regenerated in PDA regenerative medium using bilayer plate culture method. To validate the usability of the protoplasts, a novel plasmid with green fluorescent protein (GFP) was used in transformation for easy visual observation. The results showed that the protoplasts prepared by the optimized method were active and applicable in further gene manipulation experiments. A Golden Section approach to optimization of automotive friction materials   总被引:4,自引:0,他引:4  
A Golden Section approach combined with relational grade analysis is proposed as an experimental design tool helpful in the development of new automotive friction materials. Golden Section was used to design the volume fraction of the components systematically. The changes in friction performance (friction coefficient and wear) measured using Friction Assessment and Screening Test (FAST) can be correlated with component variations by use of relational grade analysis. This approach was utilized to optimize a non-metallic friction material containing seven ingredients including two fibers (aramid and slag fiber), four fillers (Al2O3, BaSO4, graphite and nitrile rubber) and one binder (benzoxazine). The volume fraction of seven components was varied simultaneously in order to optimize the parameters of friction coefficient and wear with a minimum number of tests. Three phases were performed to find the optimal proportion of the components. The optimized friction performance was obtained after doing 19 formulation experiments.  相似文献

Hydrogenated diamond-like carbon (DLC) films were prepared by using liquid phase electrodeposition technique. The effects of the applied potential and the carbon sources on the deposition process and film structures were studied. It has been found that the organic liquids with high dielectric constants, small viscosities and the methyl group bonding to the polar group are appropriate carbon sources. The increasing of potential improves the formation of sp3 carbon during the deposition process. In a high electric field, organic molecules are polarized and reacted on the surface of the electrode, turning out DLC and other products. It is believed that the reaction follows a polarization-reaction mechanism.  相似文献

Ultra-fine polyimide fibers containing silver nanoparticles were prepared by electrospinning from poly(amic acid)/(trifluoroacetylacetonoto)silver(I) (PAA/AgTFA) solution. Thermal curing of the silver(I)-containing fibers led to cycloimidization of the poly(amic acid) into polyimide with concomitant silver(I) reduction. The polyimide–silver fibers were characterized by FT-IR, X-ray diffraction, scanning electron microscopy, and transmission electron microscopy. The average diameters of silver nanoparticles and ultra-fine PI fibers electrospun from solutions with different amounts of AgTFA were studied, and the crystal structure of silver nanoparticles was also presented.  相似文献

M41S materials are prepared by in situ assembly of inorganic precursors and organic template and can be viewed as nanocomposites of the siliceous phase and organic surfactant. Calcination of these precursors gives the M41S materials that have been used to prepare novel nanocomposite structures, in which the organic phase inside the nano-sized pores is isolated by the nano-sized inorganic pore walls. The nanocomposite structures can be formed by in situ polymerization of monomers inside the channels. Polymerization of ethylene takes place inside the nano-sized pores, producing the desired nanocomposite structure. The resulting polyethylene was found to be a mixture of crystalline and amorphous phases.  相似文献

松材线虫病害高光谱时序与敏感特征研究   总被引:2,自引:0,他引:2  
采用高光谱仪ASD FieldSpec Pro FR,连续观测了健康和发病马尾松针叶的时序高光谱,分析了松材线虫病害光谱的时序特征、敏感时期和敏感特征。结果表明:与健康马尾松相比,病害马尾松时序光谱差异较大;病害首先造成红边区域内光谱反射率减低,然后再出现红边蓝移的2阶段光谱变化规律;感染松材线虫的马尾松9月初已经出现了病态植被典型的光谱特征;近红外平台内最大的一阶微分值、红边内一阶微分的总和(SDr)与蓝边内一阶微分的总和(SDb)的比值等是指示病害发生的显著性高光谱特征。  相似文献

研究了纳米碳酸钙/N330炭黑并用比对丁苯橡胶性能的影响。结果表明,随着CaCO3/N330并用比的改变,材料的强度有了显著的提高,特别是CaCOa/N330并用比为20/30(质量份)时复合材料的拉伸强度达到了20MPa以上,同时材料具有较好的弹性和较低的硬度;两种处理剂相比,树脂酸处理的碳酸钙具有较好的性能;表面处理的纳米炭酸钙与炭黑并用,可以降低炭黑填充橡胶的动态滞后,表面处理的纳米碳酸钙的加工性能比纳米炭黑好,可以通过并用纳米碳酸钙来改善炭黑胶料的加工性能。  相似文献
