为了适应发酵蔬菜生产企业菌种保藏,本试验探讨乳酸菌培养基质的灭菌时间,并在单因素试验基础上,采用响应面法优化改良MRS培养基配方。结果表明,乳酸菌培养基质白菜帮颗粒灭菌时间为9 min,改良MRS培养基最佳配方为:白菜帮颗粒25 g/L、蛋白胨9 g/L、牛肉膏8 g/L、葡萄糖14 g/L、柠檬酸三铵2 g/L、酵母膏5 g/L、乙酸钠2 g/L、K2HPO4 2 g/L、MgSO4·7H2O 0.58 g/L、MnSO4·4H2O 0.25 g/L。在优化培养基条件下,副干酪乳杆菌的菌数为1.19×1010 CFU/mL。1~4 ℃冷藏可保藏3~4年,-16~-20 ℃冷冻可保藏4~5年。此保藏方法操作简单、成本低,菌种质量稳定可靠,适合蔬菜发酵企业保藏生产用乳酸菌菌种。  相似文献

Well-sintered CaO granules with hydration resistance are very important for the manufacturing and application of CaO-containing refractories. CaO granules with sol-processed metal oxide protective layers were prepared by granulation and surface treatment. The obtained results indicated that the usage of nano sol greatly improved the hydration resistance of CaO granules, especially when the CaO granules were treated by alumina sol. The promoting effect of nano sols in the sintering processes improved the density of CaO granules. Nanoparticles reacted with lime at lower temperature and a new phase was formed uniformly in the treated layer, which promoted densification of the treated layer. The grain size of CaO granules in the treated layer was increased obviously with the promotion of sintering. Moreover, nanoparticles were favorable for the formation of direct bonding between CaO grains by decreasing the dihedral angle.  相似文献

Improving the hydration resistance of CaO aggregates is the key to successful application of lime-based refractories in metallurgical industry. Additive Zr(OH)4 and Al(OH)3 were introduced in the preparation of CaO granules using granulation equipment and calcination method in this study. The results showed that the hydration resistance of CaO granules was improved significantly, especially for granules with 0.6 wt.% Zr(OH)4 and 0.9 wt.% Al(OH)3, respectively. The shell of CaO granules was relatively dense after calcination and the volume of open pores of CaO granules decreased from 3.56 × 10−2 to 1.80 × 10−2 cm3/g when additive was introduced. Zr(OH)4 and Al(OH)3 have the opposite effect on the closed porosity of CaO granules, the closed porosity of CaO granules was decreased with Zr(OH)4 addition, but increased with Al(OH)3 addition.  相似文献

用X射线微层析成像(X-μCT)技术无损研究了TATB造型颗粒经单、双向钢模温压(UWDC和BWDC)法和软模温等静压(IWSC)法成形的高聚物粘结炸药(PBX)三维微细结构特征。结果表明,单向、双向压法压成的PBX内部存在初始细小裂纹和一些残余孔隙。初始损伤尺度范围从百微米至毫米。在相同温度(85℃)和压力(90 MPa)下,UWDC和BWDC法可产生初始细观损伤,但IWSC法能避免初始细观损伤。  相似文献

A combination of two‐dimensional (2D) and three‐dimensional (3D) analyses of tissue volume ultrastructure acquired by serial block face scanning electron microscopy can greatly shorten the time required to obtain quantitative information from big data sets that contain many billions of voxels. Thus, to analyse the number of organelles of a specific type, or the total volume enclosed by a population of organelles within a cell, it is possible to estimate the number density or volume fraction of that organelle using a stereological approach to analyse randomly selected 2D block face views through the cells, and to combine such estimates with precise measurement of 3D cell volumes by delineating the plasma membrane in successive block face images. The validity of such an approach can be easily tested since the entire 3D tissue volume is available in the serial block face scanning electron microscopy data set. We have applied this hybrid 3D/2D technique to determine the number of secretory granules in the endocrine α and β cells of mouse pancreatic islets of Langerhans, and have been able to estimate the total insulin content of a β cell.  相似文献

ABSTRACT

Improving the hydration resistance of CaO particle in manufacturing and application of free CaO-containing materials has practical significance. In this study, CaO granules was made from Ca(OH)2 particles, which were fabricated by the granulation method. The results showed that the hydration resistance of the CaO granules which was prepared under 1700?r?min?1 was the best, the CaO granules was sintered well in calcination process, the shell of CaO granules was relatively dense, which improves the hydration resistance of CaO granules, and the rate of hydration weight increment was 0.58% after placed in the air for 20 days under a temperature of 10–14°C and a relative humidity of 57–81%.  相似文献

In our previous study, we prepared the granules by embedding artemisinin into alginate-chitosan using microcapsule technology. Thesgranules can release artemisinin sustainably and have a strong inhibitory effect on the growth of both single Microcystis aeruginosa and mixealgae. To safely and effectively use artemisinin sustained-release granules to control algal blooms, the ecotoxicity was studied by assessing theacute and chronic toxicity to Daphnia magna(D. magna) and Danio rerio(D. rerio), along with their antioxidant activities. The results showethat the 48-h median effective concentration(EC50) of pure artemisinin to D. magna was 24.54 mg/L and the 96-h median lethal concentratio(LC50) of pure artemisinin to D. rerio was 68.08 mg/L. Both values were classified as intermediate toxicity according to the Organization foEconomic Co-operation and Development(OECD). The optimal algae inhibitory concentration of artemisinin sustained-release granules(1 g/Lhad low acute toxicity to both D. magna and D. rerio. The sustained-release granules had higher chronic toxicity to D. magna than to D. reriPartial indices of D. magna were inhibited by granules when the concentrations were larger than 0.1 g/L. Low granule concentration had ainductive effect on antioxidant enzyme activities in D. magna and D. rerio. With the increase of the exposure concentration and time, the enzymactivity presented a trend of first increasing and then decreasing, and the overall changes were significant. The change trend and range of enzymactivity indicated that the granules could cause serious oxidative stress to D. magna and D. rerio, and the changes were consistent with the resultof toxicity experimentation.  相似文献

The experimental die apparatus of the solid granules medium forming on sheet metal was designed and manufactured.Typical parts,such as conical,parabolic,cylindrical and square-box-shaped components,were successfully trial-produced as well.According to the analysis of the changing trends of the cross-section shape and the wall thickness during the process,it can be found that the shape of the free deformation zone of the sheet metal,which is the most critical thinning area,can be described as an approximately spherical cap.According to this forming feature,back pressure deep drawing technology with solid granules medium on sheet metal was proposed to restrain drastic thinning at the bottom of the part through the joint friction effect of solid granules medium,the back pressure tringle and the sheet metal.Therefore,the deep drawing limit of the sheet metal is significantly improved.In order to fabricate thin-walled rotary parts with great drawing ratio and complex cross-sections,a finite element model based on the material property test of the solid granules medium was established to optimize the scheme of the back pressure deep drawing.The effects on the forming performance of sheet metal from back pressure load and the approach of blank holding control were analyzed through this model.  相似文献

Partitioning the universe of discourse and determining intervals containing useful temporal information and coming with better interpretability are critical for forecasting in fuzzy time series. In the existing literature, researchers seldom consider the effect of time variable when they partition the universe of discourse. As a result, and there is a lack of interpretability of the resulting temporal intervals. In this paper, we take the temporal information into account to partition the universe of discourse into intervals with unequal length. As a result, the performance improves forecasting quality. First, time variable is involved in partitioning the universe through Gath–Geva clustering-based time series segmentation and obtain the prototypes of data, then determine suitable intervals according to the prototypes by means of information granules. An effective method of partitioning and determining intervals is proposed. We show that these intervals carry well-defined semantics. To verify the effectiveness of the approach, we apply the proposed method to forecast enrollment of students of Alabama University and the Taiwan Stock Exchange Capitalization Weighted Stock Index. The experimental results show that the partitioning with temporal information can greatly improve accuracy of forecasting. Furthermore, the proposed method is not sensitive to its parameters.  相似文献
