Ganoderic acids and water-soluble polysaccharides are the main bioactive ingredients of Ganoderma lucidum and are heat sensitive. Hence, it is crucial to apply a suitable drying method for the drying of G. lucidum to minimize the loss of these bioactive ingredients. In this study, a two-stage drying method was applied to enhance the drying kinetics, as well as the retention of both ganoderic acids and water-soluble polysaccharides in dried G. lucidum fruiting bodies and slices compared to other single-stage-dried products. The results showed that two-stage drying enhanced the overall drying rate, which consequently shortened the total drying time up to 61 and 330% for fruiting bodies and slices, respectively, compared to single-stage drying methods. Furthermore, two-stage drying of Ganoderma fruiting bodies, which involves 18-h vacuum drying followed by 5-h heat pump drying, maximized the retention of ganoderic acids and water-soluble polysaccharides, with the retention of 97.90% of water-soluble polysaccharides and 4.2% more ganoderic acids than freeze-dried products. On the other hand, drying of Ganoderma slices within the shortest total drying time, which is 10-min heat pump drying followed by 50-min oven drying, also retained most of the ganoderic acids and water-soluble polysaccharides in the dried products; that is, about 80% of water-soluble polysaccharides and 11% more ganoderic acids than freeze-dried slices.  相似文献

Natural mycelial exopolysaccharide (EPS) and endopolysaccharide (ENS) extracted from bioreactor-cultivated European Ganoderma applanatum mushrooms are of potential high commercial value for both food and adjacent biopharmaceutical industries. In order to evaluate their potential toxicity for aquaculture application, both EPS (0.01–10 mg/mL) and ENS (0.01–10 mg/mL) extracts were tested for Zebrafish Embryo Toxicity (ZFET); early development effects on Zebrafish Embryos (ZE) were also analyzed between 24 and 120 h post-fertilization (HPF). Both EPS and ENS are considered non-toxic with LC50 of 1.41 mg/mL and 0.87 mg/mL respectively. Both EPS and ENS did not delay hatching and teratogenic defect towards ZE with <1.0 mg/mL, respectively. No significant changes in the ZE heart rate were detected following treatment with the two compounds tested (EPS: 0.01–10 mg/mL: 176.44 ± 0.77 beats/min and ENS: 0.01–10 mg/mL: 148.44 ± 17.75 beats/min) compared to normal ZE (120–180 beats/min). These initial findings support future pre-clinical trials in adult fish models with view to safely using EPS and ENS as potential feed supplements for supplements for development of the aquaculture industry.  相似文献

Ganoderma mushrooms (Lingzhi in Chinese) have well‐documented health benefits. Ganoderma tsugae (G. tsugae), one of the ganoderma species, has been commercially cultivated as a dietary supplement. Because G. tsugae has high antioxidant activity and because oxidative stress is often associated with cardiac injury, we hypothesized that G. tsugae protects against cardiac injury by alleviating oxidative stress. We tested the hypothesis using a work‐overload‐induced myocardial injury model created by challenging mice with isoproterenol (ISO). Remarkably, oral G. tsugae protected the mice from ISO‐induced myocardial injury. Moreover, the triterpenoid fraction of G. tsugae, composed of a mixture of nine structurally related ganoderic acids (GAs), provided cardioprotection by inhibiting the ISO‐induced expression of Fas/Fas ligand, oxidative stress, and apoptosis. The antioxidant activity of GAs was tested in cultured cardio‐myoblast H9c2 cells against the insult of H2O2. GAs dissipated the cellular reactive oxygen species imposed by H2O2 and prevented cell death. Our findings uncovered the cardioprotective activity of G. tsugae and identified GAs as the bioactive components against cardiac insults.  相似文献

Triclosan (TCS), an antimicrobial agent, is an emerging and persistent environmental pollutant that is often found as a contaminant in surface waters and sediments; hence, knowledge of its degradability is important. In this study we investigated laccase-mediated TCS transformation and detoxification, using laccase (from the fungus Ganoderma lucidum) in the presence and absence of redox mediators. Transformation products were identified using HPLC, ESI-MS and GC-MS, and transformation mechanisms were proposed. In the absence of redox mediator, 56.5% TCS removal was observed within 24 h, concomitant with formation of new products with molecular weights greater than that of TCS. These products were dimers and trimers of TCS, as confirmed by ESI-MS analysis. Among the various mediators tested, 1-hydroxybenzotriazole (HBT) and syringaldehyde (SYD) significantly enhanced TCS transformation (∼90%). The presence of these mediators resulted in products with lower molecular weights than TCS, including 2,4-dichlorophenol (2,4-DCP; confirmed by GC-MS) and dechlorinated forms of 2,4-DCP. When SYD was used as the mediator, dechlorination resulted in 2-chlorohydroquinone (2-CHQ). Bacterial growth inhibition studies revealed that laccase-mediated transformation of TCS effectively decreased its toxicity, with ultimate conversion to less toxic or nontoxic products. Our results confirmed the involvement of two mechanisms of laccase-catalyzed TCS removal: (i) oligomerization in the absence of redox mediators, and (ii) ether bond cleavage followed by dechlorination in the presence of redox mediators. These results suggest that laccase in combination with natural redox mediator systems may be a useful strategy for the detoxification and elimination of TCS from aqueous systems.  相似文献

Ganoderma is normally dried to extend its shelf life without using chemical preservative and to concentrate the medicinal value in the fruiting body. Convective hot air drying characteristics of Ganoderma tsugae Murrill were evaluated in hot air circulated oven at different drying temperatures, sizes, and air flow rates. The drying kinetics of Ganoderma tsugae in kidney shape and slices were investigated and compared at different drying conditions. The variation of effective moisture diffusivity values at decreasing moisture contents during drying was determined from the drying data. Four well-known thin-layer drying models were fitted to the experimental data and the Midilli model was found to satisfactory describe the drying characteristics of kidney-shaped Ganoderma tsugae. Ganoderma tsugae dried at 50°C with air velocity of 1.401 ms?1 showed the highest retention of crude ganoderic acid content compared to other drying conditions.  相似文献

Over the past century, a number of synthetic antimicrobial agents have been discovered and developed, but drug resistance and toxicity are still the major hindrances to gaining successful therapeutic outcomes in many instances. Herbal medicines may represent a safe and useful supplement to existing chemotherapeutic therapies for the management of infectious diseases. Ganoderma has traditionally been used to treat chronic infectious diseases, such as chronic hepatitis and bronchitis in Asia, when it is administered alone or more often in combination with chemotherapeutic agents. Preclinical (in vitro and in vivo animal) studies indicate that Ganoderma exhibits a broad spectrum of antibacterial and antiviral activities, whereas data in human beings are scanty. Polysaccharides or triterpenoids from Ganoderma showed activities against Herpes simple virus, Hepatitis B virus, HIV, and Epstein-Barr virus in vitro or in animal models. Ganoderma species also contain antibacterial constituents inhibiting gram-positive and/or gram-negative bacteria in vitro. However, it is difficult to extrapolate these findings to humans, as most of these preclinical studies were conducted under optimized conditions with the use of high doses of Ganoderma components. A double-blind, randomized, placebo-controlled clinical study indicated that treatment of hepatitis B patients with G. lucidum polysaccharides at 5400 mg/day for 12 weeks resulted in significantly decreased serum HBV DNA and hepatitis B e antigen (HbeAg) levels. The mechanisms for the antimicrobial and antiviral activities of Ganoderma are largely undefined. Currently available data do not support the use of Ganoderma as an antibiotic, but it may play an adjunct role for the management of bacterial and viral infection. 目的优化灵芝多糖脱色工艺,选取最优脱色方式。方法选取活性炭、壳聚糖、H_2O_2 3种脱色剂对灵芝多糖脱色处理,以脱色率和多糖保留率为指标,在单因素实验基础上,进行正交工艺优化。以2,2-二(4-叔辛基苯基)-1-苦肼基自由基(DPPH·)清除活性分析脱色后的灵芝多糖的抗氧化活性。结果活性炭对灵芝多糖的脱色率为67.72%,多糖保留率为72.12%;壳聚糖对灵芝多糖的脱色率为33.57%,多糖保留率63.00%;H_2O_2对灵芝多糖的脱色率为84.11%,多糖保留率为73.12%,综合考虑H_2O_2脱色效果最好,壳聚糖脱色法所得多糖的DPPH自由基清除能力最强。结论活性炭、壳聚糖、H_2O_2 3种脱色剂均可用于灵芝多糖脱色, H_2O_2效果较好,值得进一步的开发和利用。  相似文献

平板培养基上比较了冬虫夏草、韩芝及树舌的菌丝生长速度和对淀粉的利用情况,结果显示三者的生长速度分别达到1.28、0.64和0.55cm/d,且韩芝与树舌对淀粉的利用均优于冬虫夏草;混合培养结果得出菌种的最佳菌龄比为虫草:韩芝=3d:5d,最佳的接种比例为2:3;正交试验混合发酵最佳的培养基配方为(% ):葡萄糖1.0 、淀粉2.0、黄豆粉2.0、酵母粉0.5.在此条件下,菌丝生物量达到最大值1.97g/100ml.  相似文献
