采用间接法定量,建立了HPLC-ELSD外标法和GC-FID内标法测定中长链甘油三酯含量的方法,并对两种方法进行比较。结果表明:两种方法在各自的质量浓度范围内均具有良好的线性关系,HPLC-ELSD外标法测定辛癸酸甘油酯和大豆油的平均加标回收率分别为101.0%和99.5%,RSD分别为1.58%和1.62%,得到酯交换产物中中长链甘油三酯含量为72.8%±0.80%;GC-FID内标法测定辛癸酸甘油酯和大豆油的平均加标回收率分别为99.4%和99.6%,RSD分别为1.24%和2.03%,得到酯交换产物中中长链甘油三酯含量为72.5%±0.84%。说明两种分析方法均具有良好的精密度和重复性。HPLC-ELSD外标法的分析时间为43 min,GC-FID内标法分析时间仅需12.7 min。 相似文献

建立HPLC-ELSD法测定了30批银杏叶中4种银杏内酯的含量,并对所得数据进行分析研究.色谱条件为:phenonoonon C18色谱柱(250 mm×4.6 mm,5 μm),35%(体积分数)甲醇为流动相,1.000 mL/min的流速,ELSD检测器漂移管温度为60 ℃,氮气压力为1.72×105 Pa.得出红土栽培,未嫁接,树龄6~7 a的银杏叶中银杏内酯含量较高;4种银杏内酯总量集中在0.3%~0.5%(质量分数).该方法准确,重现性好,适用于银杏叶中4种银杏内酯的含量测定;银杏内酯的含量受生长环境影响差别较大,影响因素有待深入研究. 相似文献

为了解酱香型白酒酿造过程中产多元醇酵母的种类和功能,以酱香型白酒大曲和酒醅中分离的酵母为研究对象,采用薄层层析法和高效液相色谱-蒸发光散射检测器法(HPLC-ELSD)对具有产多元醇功能的酵母进行筛选和多元醇定性、定量分析。实验结果显示,从142株酵母菌中筛选出5株具有产多元醇功能的酵母菌娄德酵母(Lodderomyces elongisporus) FBKL2.0073、库得毕赤酵母(Pichia kudriavzevii) FBKL2.0008、Trichosporon coremiiforme FBKL2.0310、Trichosporon coremiiforme FBKL2.0307和Trichosporonoides sp. FBKL2.0315,其中前4株菌都能产D-阿拉伯糖醇,L. elongisporus FBKL2.0073菌株产多元醇的总量最多,为(9.87±0.85) g/L。 相似文献

该研究采用高效液相色谱-蒸发光散射检测(HPLC-ELSD)法测定新疆药桑叶中1-脱氧野尻霉素(1-DNJ)的含量,色谱柱为Waters XBridgeTM HILIC(2.1 mm×100 mm, 5 μm),乙腈-水(85∶15, V/V)为流动相,流速0.2 mL/min。结果表明,1-DNJ在1.2~12.0 μg/mL质量浓度范围内线性关系良好,平均回收率为98.25%,相对标准偏差(RSD)为1.16%。该方法快捷简便、结果可靠,可用于药桑叶中1-脱氧野尻霉素含量的分析检测。 相似文献

以木薯茎杆栽培的食用菌为试材,对海藻糖的提取方法和高效液相色谱-蒸发光散射(high performanceliquid chromatography-evaporative light scattering detector,HPLC-ELSD)检测方法进行优化,分析不同比例木薯茎秆栽培3 种食用菌(黑木耳、平菇、榆黄蘑)海藻糖含量。结果表明,食用菌中的海藻糖在80 ℃水浴60 min条件下可有效提取,最高含量达到190.5 mg/g;利用优化后色谱条件分析标准品和样品中海藻糖,其分离效果均良好。并分析黑木耳、平菇和榆黄蘑中海藻糖含量,发现当栽培基质的木薯茎秆比例为31.2%时,3 种食用菌海藻糖含量较高,分别为61.5、250.3 mg/g和15.0 mg/g。结果表明,HPLC-ELSD可分析3 种食用菌中海藻糖含量,方法准确度高、重复性好、样品稳定性较好;3 种食用菌海藻糖含量与培养基质中木薯茎秆比例有关,合适的木薯茎秆比例可提高食用菌中海藻糖含量。 相似文献

An effective HPLC method to analyse platycosides from the balloon flower root was developed using ELSD. The optimum resolution of the platycosides was achieved on an ODS column with gradient elution of eluent A, 30 mM ammonium acetate buffer (pH 4.81): methanol: acetonitrile = 75:5:20 (v/v/v), and B, 69:5:26 (v/v/v). Amongst 18 platycosides, platycoside E showed the highest content, followed by polygalacin D2 and 3″-O-acetylplatyconic acid A. The sum of these three compounds was recommended for quality control of balloon flower root for medicinal purposes. The samples could be clustered into groups based on platycoside content. Group I, characterised by a high concentration of platycosides, was located near the west coast of Korea, whereas group II, characterised by a low concentration of platycosides, was located inland or in mountainous area. Monitoring the consumption of sugars during fermentation is a key to optimizing product formation and maintaining a healthy environment for microorganisms. Difficulty arises in the availability of a rapid, inexpensive and sensitive method for the detection of sugars because fermentation media are a complex mix of nutrients, cell debris, waste and target products. A method involving reaction-based UV-Vis spectrophotometry for the quantitative determination of xylose as the target sugar was developed. Factors affecting xylose concentration measurements such as hydrochloric acid concentration, heating time and the amount of Fe3+ catalyst were investigated. A continuous scan revealed the working wavelength to be 671 nm. The effect of other components in the fermentation broth was found to be negligible. Absorbance shows a linear relationship with xylose concentration within a range of 0.1-0.5 g/L. Xylose concentrations from fermentation samples obtained at specific time intervals (0-168 h) were determined with the method and compared with YSI 2700, an enzyme electrode, HPLC-ELSD method, currently a common technique for measuring xylose and GC aldononitrile sugar derivatization method. Dilution is necessary for comparable xylose concentrations with YSI 2700 and HPLC-ELSD. Xylose concentration measurements obtained with the UV-Vis spectrophotometric method although quantitatively comparable to HPLC-ELSD xylose measurements were easily and conveniently obtained compared to YSI 2700, HPLC-ELSD and GC derivatization methods. 相似文献

利用从酱香型大曲中和酒醅筛选出的德巴利酵母和Trichosporon coremiiforme提高酱香型白酒中醇甜风格特征。将FBKL2.0130(德巴利酵母)和FBKL2.0310(Trichosporon coremiiforme)添加到下沙堆积后酒醅中模拟窖池发酵,考察不同菌株添加量对酒醅中多元醇含量的影响,采用高效液相-蒸发光散射检测器对酒醅中多元醇含量进行测定,对酒醅中微生物生物量和理化指标进行分析。结果表明,发酵后酒醅中细菌和酵母数量为102g-1,未发现霉菌存在;接种量增加,会加快对原料中还原糖的利用;添加功能性菌株FBKL2.0130和FBKL2.0310均能使酒醅中多元醇含量增加,菌株FBKL2.0130接种量10%时,多元醇含量最高,为1.418 mg/g;菌株FBKL2.0310接种量12%时,多元醇含量最高,为1.461 mg/g。 相似文献
