Multiparous Holstein cows (n = 300) were assigned to 1 of 2 milking frequency treatments at parturition. Cows were either milked 6 times (6×) or 3 times (3×) daily to determine effects on early lactation milk yields and subsequent lactation persistency with or without use of recombinant bST (rbST). Treatments included a control group milked 3× and 3 groups milked 6× for either the first 7, 14, or 21 days in milk (DIM). Those 4 groups of cows all received rbST starting at 63 DIM. The fifth treatment group was also milked 6× for the first 21 DIM but those cows received no rbST during the entire lactation. All cows returned to 3× milking after their respective treatment periods ended. Cows milked 3× tended to produce more milk (43.2 vs. 41.5 and 41.0 ± 1.1 kg/d) during the first 9 wk of lactation compared with cows milked 6× for 7 or 21 DIM, respectively. Group milk yields after wk 9 averaged 38.3 ± 0.7 kg/d and did not differ among various groups assigned to an increased milking frequency in early lactation. Percentages of milk fat (3.8 ± 0.12%) and protein (2.9 ± 0.06%) did not differ among treatments during the first 9 wk after calving. Early lactation milk yield (41.9 ± 1.2 kg/d) did not differ between the 2 groups of cows milked 6× for 21 DIM. However, cows subsequently administered rbST (at 63 DIM) produced more milk (38.8 vs. 34.2 ± 0.9 kg/d) from wk 10 to 44. The number of cows sent to the hospital during the 305-d trial for mastitis (97), digestive disorders (14), respiratory issues (9), lameness (22), or retained placenta (16), were not affected by treatments (χ2 = 0.49). Under the conditions of this commercial dairy herd in Arizona, increasing milking frequency to 6 times daily for 7 to 21 d at the start of lactation conditions did not increase milk yield nor improve lactation persistency.  相似文献

Thromboelastography is a whole blood-based coagulation assay that can be used to investigate hypocoagulability and hypercoagulability, as seen with thromboembolic diseases and disseminated intravascular coagulation. Numerous coagulopathies due to different causes are reported in cows. The objective was to establish reference intervals for thromboelastography using the TEG 5000 (Haemonetics GmbH, Munich, Germany) with citrated whole blood samples and kaolin activation in dairy cows and to investigate possible thromboelastographic changes between cows in different lactation periods. An additional objective was to test the stability of samples for up to 100 h. Sixty blood samples from healthy Holstein-Friesian cows were examined. The samples were allocated to 3 different lactation groups (≤30 d postcalving, 31–99 d postcalving, ≥100 d postcalving). Thromboelastography was performed by using the TEG 5000 analyzer with citrated whole blood samples with kaolin activation. The calculated reference intervals were as follows: reaction time = 2.2 to 6.2 min, coagulation time = 0.8 to 2.0 min, angle α = 58.2 to 81.8°, maximum amplitude = 64.3 to 89.2 mm, and clot rigidity = 9.2 to 41.2 dyn/cm2. The 3 different lactation groups showed no significant differences in TEG parameters. No significant difference was seen in samples stored for up to 48 h at room temperature, which indicates that delays in processing samples, such as those arising during transit, are not an issue.  相似文献

In Ireland, milk is primarily produced using a spring-calving grass-based system, with the use of concentrate supplementation mainly when pasture availability and quality are reduced. In the autumn, when cows are in late lactation, reduced pasture productivity results in reduced milk yield and altered milk composition. Nitrogen utilization efficiency also reduces as lactation progresses. Concentrate supplementation has been found to increase milk production and reduce nitrogen (N) excretion, as high-N grass is usually replaced by a lower-N supplement; however, there is a paucity of information with regard to the optimum type of supplementation in late lactation. Therefore, the objective of this research is to investigate the effect of different concentrate supplementation types, based on barley or maize, on milk production, dry matter intake (DMI), rumen fermentation, and N excretion in late-lactation, spring-calving, grazing dairy cows. Thirty-six Holstein Friesian dairy cows were blocked on days in milk (185 DIM) and balanced for parity, pre-experimental milk yield, milk composition, and body condition score. Cows were randomly assigned to 1 of 3 dietary treatments in a randomized complete block design (n = 12). The 3 treatments consisted of a perennial ryegrass-based pasture-only (PO) treatment and pasture plus either of 2 supplementary concentrates, based on barley (PB) or maize (PM). The diets were fed for a 14-d acclimatization period and then for a further 63-d experimental period. Cows offered PO had a lower daily milk yield (15.1 kg) than PB (18.2 kg) or PM (16.8 kg). Similarly, PO had lower daily milk solids yield (1.46 kg) than PB or PM (1.68 and 1.53 kg, respectively). Cows offered PB had a greater milk yield and higher fat and protein yields than those offered PM. Offering PB increased total DMI (19.5 kg) compared with PO (17.7 kg), and milk response to concentrates was also greater for PB compared with PM (1.21 vs. 0.71 kg of milk per kg of concentrate). Cows offered PB had increased N in milk compared with PO. In conclusion, concentrate supplementation based on barley or maize resulted in increased milk and milk solids yield compared with offering PO. Cows offered barley had a greater response to concentrates and increased milk and milk solids yield in comparison to maize and showed increased N partitioning in milk compared with PO. A barley-based concentrate increased total DMI compared with PO.  相似文献

Modeling extended lactations for the US Holsteins is useful because a majority (>55%) of the cows in the present population produce lactations longer than 305 d. In this study, 9 empirical and mechanistic models were compared for their suitability for modeling 305-d and 999-d lactations of US Holsteins. A pooled data set of 4,266,597 test-day yields from 427,657 (305-d complete) lactation records from the AIPL-USDA database was used for model fitting. The empirical models included Wood (WD), Wilmink (WIL), Rook (RK), monophasic (MONO), diphasic (DIPH), and lactation persistency (LPM) functions; Dijkstra (DJ), Pollott (POL), and new-multiphasic (MULT) models comprised the mechanistic counterparts. Each model was separately tested on 305-d (>280 days in milk) and 999-d (>800 days in milk) lactations for cows in first parity and those in third and greater parities. All models were found to produce a significant fit for all 4 scenarios (2 parity groups and 2 lactation lengths). However, the resulting parameter estimates for the 4 scenarios were different. All models except MONO, DIPH, and LPM yielded residuals with absolute values smaller than 2 kg for the entire period of the 305-d lactations. For the extended lactations, the prediction errors were larger. However, the RK, DJ, POL, and MULT models were able to predict daily yield within a ± 3 kg range for the entire 999-d period. The POL and MULT models (having 6 and 12 parameters, respectively) produced the lowest mean square error and Bayesian information criteria values, although the differences from the other models were small. Conversely, POL and MULT were often associated with poor convergence and highly correlated, unreliable, or biologically atypical parameter estimates. Considering the computational problems of large mechanistic models and the relative predictive ability of the other models, smaller models such as RK, DJ, and WD were recommended as sufficient for modeling extended lactations unless mechanistic details on the extended curves are needed. The recommended models were also satisfactory in describing fat and protein yields of 305-d and 999-d lactations of all parities.  相似文献

The effects of lactation stage, negative energy balance (NEB), and milk fat depression (MFD) were estimated on detailed milk fat composition in primiparous Holstein-Friesian cows. One morning milk sample was collected from each of 1,933 cows from 398 commercial Dutch herds in winter 2005. Milk fat composition was measured using gas chromatography, and fat and protein percentage were measured using infrared spectrometry. Each fatty acid changed 0.5 to 1 phenotypic standard deviation over lactation, except odd-chain C5:0 to C15:0, branched-chain fatty acids, and trans-10, cis-12 conjugated linoleic acid (CLA). The greatest change was an increase from 31.2 to 33.3% (wt/wt) for C16:0 from d 80 to 150 of lactation. Energy status was estimated for each cow as the deviation from each average lactation fat-to-protein ratio (FPdev). A high FPdev (>0.12) indicated NEB. Negative energy balance was associated with an increase in C16:0 (0.696 ± 0.178) and C18:0 (0.467 ± 0.093), which suggested mobilization of body fat reserves. Furthermore, NEB was associated with a decrease in odd-chain C5:0 to C15:0 (−0.084 ± 0.020), which might reflect a reduced allocation of C3 components to milk fat synthesis. A low FPdev indicated MFD (<−0.12) and was associated with a decrease in C16:0 (−0.681 ± 0.255) and C18:0 (−0.128 ± 0.135) and an increase in total unsaturated fatty acids (0.523 ± 0.227). The study showed that both lactation stage and energy balance significantly contribute to variation in milk fat composition and alter the activity of different fatty acid pathways.  相似文献

After parturition, the start of copious milk production in dairy cows requires the closure of tight junctions (TJ) to form the blood–milk barrier and prevent paracellular transfer of blood constituents into milk [e.g., lactate dehydrogenase (LDH) and serum albumin (SA)] and vice versa [e.g., appearance of α-lactalbumin (α-LA) in blood]. Serotonin (5-HT) has been demonstrated to alter tight junction permeability in the mammary gland. The present study investigated individual differences of TJ permeability of mammary epithelium at the beginning of lactation in relation to circulating 5-HT in dairy cows. Blood and milk samples were obtained from 11 multiparous Holstein dairy cows for the first time at 4 h after parturition, at the following 5 milkings, and at the evening milkings on d 5, 8, 10, and 14 of lactation. Retrospectively, cows were split into 2 groups according to their calculated areas under the curve of serum 5-HT during the entire experimental period: a high-serum 5-HT (HSS) group (5 cows) and a low-serum 5-HT (LSS) group (6 cows). The areas under the curve of serum 5-HT concentrations over the 324-h experimental period were 62 ± 2 × 103 ng/mL in HSS and 25 ± 5 × 103 ng/mL in LSS. Plasma α-LA concentration was greater in LSS than in HSS cows at the first milking, but no difference between groups was found from the second to sixth milking. Yield of α-LA in milk was lower in HSS than in LSS during the first 6 milkings postpartum, especially in colostrum. Concentrations of α-LA, IgG1, and IgG2 in milk did not differ between groups during the entire experiment except for higher IgG observed in LSS than in HSS at the second milking and for higher IgG2 found in HSS compared with LSS on d 5. In contrast, SA concentrations and LDH activity in milk were lower in LSS compared with HSS cows during the first 6 milkings postpartum, particularly in colostrum. Milk somatic cell count was higher in HSS than in LSS throughout the study. Higher circulating 5-HT concentrations were associated with an increased transfer of the paracellularly transported SA, LDH, and somatic cell count, especially at the first milking, suggesting that 5-HT affects TJ permeability during closure of the blood–milk barrier at the onset of lactation. Furthermore, higher serum 5-HT concentrations were associated with a lower α-LA yield in milk. A consistent relationship with serum 5-HT concentrations was neither observed for the transfer of IgG2 nor the primarily transcellular transport of IgG1 during the first milkings after parturition.  相似文献

Milk production in heifers induced into lactation is lower than that of postpartum primiparous cows. A method to improve milk production in induced lactations may provide opportunities for increased profitability as well as increase our understanding of the mechanisms that regulate mammary gland development and colostrum composition. The present study was conducted to determine if dexamethasone administration at the onset of milking or age at lactation induction would affect milk production in heifers induced into lactation. Holstein heifers at 14 [n = 20; 354 ± 38 kg of body weight (BW)] and 18 mo of age (n = 20; 456 ± 30 kg of BW) were assigned randomly to dexamethasone (DEX) or control (CON) treatment groups in a 2× 2 factorial arrangement with age and dexamethasone treatment as the 2 factors. Heifers were induced into lactation with daily subcutaneous injections of estradiol-17β and progesterone (0.075 and 0.25 mg/kg of BW per d, respectively) on experimental d 1 to 7. They also received bovine somatotropin (bST) every 14 d beginning on experimental d 1. Milking began on experiment d 18 (lactation d 1). Dexamethasone (10 mg) was administered on lactation d 1 and 2 following the morning milking; CON heifers did not receive dexamethasone. Milk yield from d2 to 15 of lactation of heifers receiving DEX (7.8 kg/d) was greater than that of CON heifers (6.0 kg/d) but was similar thereafter through 305 d of lactation (18.2 kg/d). Milk production to d 11 was similar for 14- and 18-mo-old heifers but was greater for 18- (18.9 kg/d) than for 14-mo-old animals (17.4 kg/d) through 305 d in milk. Milk fat percentage increased initially and was greater in DEX (4.51%) compared with CON (3.53%) heifers until 21 d in milk. Milk protein and lactose concentrations were not affected by DEX treatment. Age at induction did not affect milk fat, protein, or lactose percentages. Mean milk IgG concentration declined from 107.4 mg/mL on d 1 to 5.0 mg/mL on d 7 of lactation, tended to be greater for 18- compared with 14-mo-old heifers, and was not different due to DEX treatment. Administration of DEX to heifers induced into lactation increased initial milk production during the first 2 wk of lactation but this effect did not persist through 305 DIM. Treatment with DEX appeared to stimulate mammary cell differentiation but did not change the rate of decline of milk IgG concentrations. Higher milk yield in 18-mo-old heifers may be due to greater mammary epithelium, higher body mass, or both.  相似文献

The objective of this experiment was to measure ruminal and lactational responses of Holstein dairy cows fed diets containing 3 different starch levels: 17.7 (low; LS), 21.0 (medium; MS), or 24.6% (high; HS). Twelve multiparous cows (118 ± 5 d in milk) were assigned randomly to dietary treatment sequence in a replicated 3 × 3 Latin square design with 3-wk periods. All diets were fed as total mixed rations and contained approximately 30.2% corn silage, 18.5% grass silage, and 5.0% chopped alfalfa hay. Dietary starch content was manipulated by increasing dry ground corn inclusion (% of dry matter) from 3.4 (LS) to 10.1 (MS) and 16.9 (HS) and decreasing inclusion of beet pulp and wheat middlings from 6.7 and 13.4 (LS) to 3.4 and 10.1 (MS) or 0 and 6.8 (HS). In vitro 6-h starch digestibility of the diet increased as nonforage sources of fiber replaced corn grain (% of dry matter; 73.6, HS; 77.3, MS; 82.5, LS) resulting in rumen-fermentable starch content by 14.6, 16.2, and 18.1% for the LS, MS, and HS diets, respectively. Diets had similar neutral detergent fiber from forage and particle size distributions. Dry matter intake, solids-corrected milk yield, and efficiency of solids-corrected milk production were unaffected by diet, averaging 26.5 ± 0.8, 40.8 ± 1.6, and 1.54 ± 0.05 kg/d, respectively. Reducing dietary starch did not affect chewing time (815 ± 23 min/d), mean ruminal pH over 24 h (6.06 ± 0.12), acetate-to-propionate ratio (2.4 ± 0.3), or microbial N synthesized in the rumen (585 ± 24 g/d). Total tract organic matter digestibility was higher for HS compared with MS and LS diets (69.2, 67.3, and 67.0%, respectively), but crude protein, neutral detergent fiber, and starch digestibilities were unaffected. As dietary starch content decreased, in vitro ruminal starch fermentability increased and, consequently, the range between HS and LS in rumen-fermentable starch (3.5 percentage units) was less than the range in starch content (6.9 percentage units). Under these conditions, dietary starch content had no measurable effect on ruminal fermentation or short-term lactational performance of high-producing Holstein dairy cows.  相似文献
