Pycnodysostosis, a rare autosomal recessive skeletal dysplasia, is caused by a deficiency of cathepsin K. Patients have impaired bone resorption in the presence of normal or increased numbers of multinucleated, but dysfunctional, osteoclasts. Cathepsin K degrades collagen type I and generates N-telopeptide (NTX) and the C-telopeptide (CTX) that can be quantified. Levels of these telopeptides are increased in lactating women and are associated with increased bone resorption. Nothing is known about the consequences of cathepsin K deficiency in lactating women. Here we present for the first time normalized blood and CTX measurements in a patient with pycnodysostosis, exclusively related to the lactation period. In vitro studies using osteoclasts derived from blood monocytes during lactation and after weaning further show consistent bone resorption before and after lactation. Increased expression of cathepsins L and S in osteoclasts derived from the lactating patient suggests that other proteinases could compensate for the lack of cathepsin K during the lactation period of pycnodysostosis patients.  相似文献

We investigated the differences in the effect of intramammarily infused endotoxin for cows in early (EL) and late (LL) lactation. In this crossover study, nine cows were challenged twice with 100 microg of intramammarily infused Escherichia coli 0111:B4 LPS, each cow serving its own control. Systemic and local signs were recorded throughout the experiment to assess the severity of each cow's response. Daily milk yield and indicators of inflammation in blood and milk were also recorded. The response was significantly more severe in the EL period. Before the challenge, milk yield and serum free fatty acid concentration were higher and serum urea concentration lower in EL compared with LL cows. No significant differences were present in other parameters. After the challenge, milk SCC and leukocyte function, measured as chemiluminescence, increased, more markedly in EL. Blood neutrophil content varied from neutropenia to neutrophilia. Simultaneously with neutropenia, the function of neutrophils increased in LL but decreased in EL. Serum cortisol peaked 4 h after challenge in both groups. In conclusion, endotoxin challenge resulted in more severe response in EL. During that time cows also had impaired neutrophil function postchallenge.  相似文献

The southeastern United States is characterized as humid subtropical and is subject to extended periods of high ambient temperature and relative humidity. Because the primary nonevaporative means of cooling for the cow (radiation, conduction, convection) become less effective with rising ambient temperature, the cow becomes increasingly reliant upon evaporative cooling in the form of sweating and panting. High relative humidity compromises evaporative cooling, so that under hot, humid conditions common to the Southeast in summer the dairy cow cannot dissipate sufficient body heat to prevent a rise in body temperature. Increasing air temperature, temperature-humidity index and rising rectal temperature above critical thresholds are related to decreased dry matter intake (DMI) and milk yield and to reduced efficiency of milk yield. Modifications including shade, barns which enhance passive ventilation, and the addition of fans and sprinklers increase body heat loss, lowering body temperature and improving DMI. New technologies including tunnel ventilation are being investigated to determine if they offer cooling advantages. Genetic selection for heat tolerance may be possible, but continued selection for greater performance in the absence of consideration for heat tolerance will result in greater susceptibility to heat stress. The nutritional needs of the cow change during heat stress, and ration reformulation to account for decreased DMI, the need to increase nutrient density, changing nutrient requirements, avoiding nutrient excesses and maintenance of normal rumen function is necessary. Maintaining cow performance in hot, humid climatic conditions in the future will likely require improved cooling capability, continued advances in nutritional formulation, and the need for genetic advancement which includes selection for heat tolerance or the identification of genetic traits which enhance heat tolerance.  相似文献

The ability of liver to respond to changes in nutrient availability is essential for the maintenance of metabolic homeostasis. Autophagy encompasses mechanisms of cell survival, including capturing, degrading, and recycling of intracellular proteins and organelles in lysosomes. During negative nutrient status, autophagy provides substrates to sustain cellular metabolism and hence, tissue function. Severe negative energy balance in dairy cows is associated with fatty liver. The aim of this study was to investigate the hepatic autophagy status in dairy cows with severe fatty liver and to determine associations with biomarkers of liver function and inflammation. Liver and blood samples were collected from multiparous cows diagnosed as clinically healthy (n = 15) or with severe fatty liver (n = 15) at 3 to 9 d in milk. Liver tissue was biopsied by needle puncture, and serum samples were collected on 3 consecutive days via jugular venipuncture. Concentrations of free fatty acids and β-hydroxybutyrate were greater in cows with severe fatty liver. Milk production, dry matter intake, and concentration of glucose were all lower in cows with severe fatty liver. Activities of serum aspartate aminotransferase, alanine aminotransferase, glutamate dehydrogenase, and γ-glutamyl transferase were all greater in cows with severe fatty liver. Serum concentrations of haptoglobin and serum amyloid A were also markedly greater in cows with severe fatty liver. The mRNA expression of autophagosome formation-related gene ULK1 was lower in the liver of dairy cows with severe fatty liver. However, the expression of other autophagosome formation-related genes, beclin 1 (BECN1), phosphatidylinositol 3-kinase catalytic subunit type 3 (PIK3C3), autophagy-related gene (ATG) 3, ATG5, and ATG12, did not differ. More important, ubiquitinated proteins, protein expression of sequestosome-1 (SQSTM1, also called p62), and microtubule-associated protein 1 light chain 3 (MAP1LC3, also called LC3)-II was greater in cows with severe fatty liver. Transmission electron microscopy revealed an increased number of autophagosomes in the liver of dairy cows with severe fatty liver. Taken together, these results indicate that excessive lipid infiltration of the liver impairs autophagic activity that may lead to cellular damage and inflammation.  相似文献

Thromboelastography is a whole blood-based coagulation assay that can be used to investigate hypocoagulability and hypercoagulability, as seen with thromboembolic diseases and disseminated intravascular coagulation. Numerous coagulopathies due to different causes are reported in cows. The objective was to establish reference intervals for thromboelastography using the TEG 5000 (Haemonetics GmbH, Munich, Germany) with citrated whole blood samples and kaolin activation in dairy cows and to investigate possible thromboelastographic changes between cows in different lactation periods. An additional objective was to test the stability of samples for up to 100 h. Sixty blood samples from healthy Holstein-Friesian cows were examined. The samples were allocated to 3 different lactation groups (≤30 d postcalving, 31–99 d postcalving, ≥100 d postcalving). Thromboelastography was performed by using the TEG 5000 analyzer with citrated whole blood samples with kaolin activation. The calculated reference intervals were as follows: reaction time = 2.2 to 6.2 min, coagulation time = 0.8 to 2.0 min, angle α = 58.2 to 81.8°, maximum amplitude = 64.3 to 89.2 mm, and clot rigidity = 9.2 to 41.2 dyn/cm2. The 3 different lactation groups showed no significant differences in TEG parameters. No significant difference was seen in samples stored for up to 48 h at room temperature, which indicates that delays in processing samples, such as those arising during transit, are not an issue.  相似文献

Methylmercury (MeHg) is one of the most dangerous toxic pollutants spread throughout the earth. Chronic MeHg intoxication by contaminated food ingestion is the most common threat to human health, including impairment to the developing fetus. The present study aims at investigating the effects of maternal exposure to MeHg during gestation and lactation on the spinal cord of offspring. Pregnant rats received oral doses of MeHg (40 μg/kg/day) over a period of 42 days (21 gestation and 21 lactation). Control animals received the vehicle only. Total mercury concentration was measured in blood samples from offspring collected at the 41st postnatal day. Counting of motor neurons and immunoreactivity for myelin basic protein (MBP) were assessed in the spinal cords in both control and MeHg-intoxicated animals. Our results showed that MeHg promoted an increase in blood Hg levels. In addition, it caused a reduction in the number of spinal cord motor neurons as well as decreased MBP immunoreactivity in the cervical, thoracic and lumbar segments. Our present findings suggest that MeHg intoxication during rat pregnancy and lactation is associated with a pattern of motor neuron degeneration and downregulation of myelin basic protein in different segments of a developing spinal cord. Further studies are needed to establish the effect of MeHg intoxication in both young and adult rats.  相似文献

In the present experiment, we tested the hypothesis that in does kidding in summer, melatonin administration during dry period is galactopoietic for the subsequent lactation and results in improved growth of their suckling kids. Twenty-five multiparous pregnant creole does were enrolled into a randomized complete block design during their dry period in the 49 d prepartum, and under natural long photoperiods around the summer solstice, pregnant does either received 2 subcutaneous ear implants (18 mg) of melatonin (MEL, n = 10) or served as nonimplanted controls (CONT, n = 15). During the first 14 wk of subsequent lactation (suckling and milking periods), MEL does yielded more milk than CONT does. Throughout subsequent lactation, milk composition was not affected by treatment. In MEL does, peripheral triiodothyronine levels peaked at 2 wk of lactation, remaining higher than in CONT does. The mean daily weight gain was higher in MEL compared with CONT kids and was also higher in males than females, and for males, was positively correlated with milk yield. The current data support our hypothesis that melatonin during the prepartum period is galactopoietic in suckling does.  相似文献

The current energy requirements system used in the United Kingdom for lactating dairy cows utilizes key parameters such as metabolizable energy intake (MEI) at maintenance (MEm), the efficiency of utilization of MEI for 1) maintenance, 2) milk production (kl), 3) growth (kg), and the efficiency of utilization of body stores for milk production (kt). Traditionally, these have been determined using linear regression methods to analyze energy balance data from calorimetry experiments. Many studies have highlighted a number of concerns over current energy feeding systems particularly in relation to these key parameters, and the linear models used for analyzing. Therefore, a database containing 652 dairy cow observations was assembled from calorimetry studies in the United Kingdom. Five functions for analyzing energy balance data were considered: straight line, two diminishing returns functions, (the Mitscherlich and the rectangular hyperbola), and two sigmoidal functions (the logistic and the Gompertz). Meta-analysis of the data was conducted to estimate kg and kt. Values of 0.83 to 0.86 and 0.66 to 0.69 were obtained for kg and kt using all the functions (with standard errors of 0.028 and 0.027), respectively, which were considerably different from previous reports of 0.60 to 0.75 for kg and 0.82 to 0.84 for kt. Using the estimated values of kg and kt, the data were corrected to allow for body tissue changes. Based on the definition of kl as the derivative of the ratio of milk energy derived from MEI to MEI directed towards milk production, MEm and kl were determined. Meta-analysis of the pooled data showed that the average kl ranged from 0.50 to 0.58 and MEm ranged between 0.34 and 0.64 MJ/kg of BW0.75 per day. Although the constrained Mitscherlich fitted the data as good as the straight line, more observations at high energy intakes (above 2.4 MJ/kg of BW0.75 per day) are required to determine conclusively whether milk energy is related to MEI linearly or not.  相似文献

Several functions were used to model the fixed part of the lactation curve and genetic parameters of milk test-day records to estimate using French Holstein data. Parametric curves (Legendre polynomials, Ali-Schaeffer curve, Wilmink curve), fixed classes curves (5-d classes), and regression splines were tested. The latter were appealing because they adjusted the data well, were relatively insensitive to outliers, were flexible, and resulted in smooth curves without requiring the estimation of a large number of parameters. Genetic parameters were estimated with an Average Information REML algorithm where the average information matrix and the first derivatives of the likelihood functions were pooled over 10 samples. This approach made it possible to handle larger data sets. The residual variance was modeled as a quadratic function of days in milk. Quartic Legendre polynomials were used to estimate (co)variances of random effects. The estimates were within the range of most other studies. The greatest genetic variance was in the middle of the lactation while residual and permanent environmental variances mostly decreased during the lactation. The resulting heritability ranged from 0.15 to 0.40. The genetic correlation between the extreme parts of the lactation was 0.35 but genetic correlations were higher than 0.90 for a large part of the lactation. The use of the pooling approach resulted in smaller standard errors for the genetic parameters when compared to those obtained with a single sample.  相似文献
