Development and monitoring of an integrated bioprocess for production of a potential malaria vaccine with Pichia pastoris

An engineered variant of the yeast Pichia pastoris was used to produce the potential malaria vaccine D1M1. This paper describes the implementation of an expanded bed adsorption chromatography step into a sequential cultivation process of the yeast in order to link protein expression, cell release, and product capture of the secreted protein in a fully automated plant. The process is operated with a semi-continuous strategy for an integrated production. Process improvements have been achieved using design of experiments. Resulting purities of product achieved up to 87% with recoveries of 51% in a single downstream operation. Furthermore, a multivariate analysis of historical batch data was used to develop a golden batch model for process monitoring.

Preschool Competency in Context: An Investigation of the Unique Contribution of Child Competencies to Early Academic Success.

The present concurrent study combined developmental and ecological considerations to examine the unique contribution of multiple preschool competencies to an indicator of early academic success. Participants included 195 Head Start children from 32 classrooms representative of a large, urban Head Start program. Dimensional (variable-centered) analyses revealed 3 distinct classroom competency dimensions (i.e., General Classroom Competencies, Specific Approaches to Learning, and Interpersonal Classroom Behavioral Problems). The first 2 of these dimensions were found to be uniquely associated with early academic success. Findings from typological (person-centered) analyses supported the dimensional findings. Typological analyses revealed 7 profiles of classroom competency distinguished by high scores on the dimensions of General Competencies and Approaches to Learning, and these profiles were found to relate differentially to the indicator of early academic success. Gasification of Biogenic Solid Fuels

As a result of shrinking fossil fuels, biomass as a regenerative energy source gains importance. To realize biomass projects it is essential to investigate in convenient thermal procedures. On this evidence an analysis and evaluation of diverse gasification technologies with different boundary conditions and diverse biomasses are indispensable. Form and kind of the biomass as well as the type of the gasification plant cause different compositions of the product gas. The gasifiers show advantages and disadvantages concerning the biomass and the produced gas quality, depending on reactor type, kind of heat supply, gasification medium, and the pressure ratio in the reactor. As the ideal gasifier for different biomass is presently not available, it will be shown, which biomass is suitable for fixed bed or fluidized bed gasifiers.

以H 2软性填料作为氧化亚铁硫杆菌的固定化载体 ,构成了固定床生物反应器。在一定操作条件下 ,考察了固定床生物反应器对Fe2 +的氧化速率 ,最大可达 7.6 7g/ (L·h)。并对固定床生物反应器运行中在载体表面形成的络合物进行了研究 ,X射线衍射证明此络合物为黄钾铁矾 [KFe3(SO4 ) 2 (OH) 6]。
