Temperature is one of the critical factors affecting gene expression in bacteria. Despite the general interest in the link between bacterial phenotypes and environmental temperature, little is known about temperature-dependent gene expression in plant pathogenic Pectobacterium atrosepticum, a causative agent of potato blackleg and tuber soft rot worldwide. In this study, twenty-nine P. atrosepticum SCRI1043 thermoregulated genes were identified using Tn5-based transposon mutagenesis coupled with an inducible promotorless gusA gene as a reporter. From the pool of 29 genes, 14 were up-regulated at 18 °C, whereas 15 other genes were up-regulated at 28 °C. Among the thermoregulated loci, genes involved in primary bacterial metabolism, membrane-related proteins, fitness-corresponding factors, and several hypothetical proteins were found. The Tn5 mutants were tested for their pathogenicity in planta and for features that are likely to remain important for the pathogen to succeed in the (plant) environment. Five Tn5 mutants expressed visible phenotypes differentiating these mutants from the phenotype of the SCRI1043 wild-type strain. The gene disruptions in the Tn5 transposon mutants caused alterations in bacterial generation time, ability to form a biofilm, production of lipopolysaccharides, and virulence on potato tuber slices. The consequences of environmental temperature on the ability of P. atrosepticum to cause disease symptoms in potato are discussed. 相似文献

The St. Clair-Detroit River System contains a world-class Great Lakes muskellunge (Esox masquinongy) fishery that has avoided the declines observed in many Great Lakes muskellunge populations. Muskellunge are an upper trophic level predator, and therefore a naturally low-density species. Limited fishery-independent data exist on which to base management decisions. To remedy this, we initiated an acoustic telemetry study in May of 2016, in collaboration with the Great Lakes Acoustic Telemetry Observation System. Our objective was to describe patterns of movement of muskellunge in this large and open system to better understand their spatial ecology. We acoustically tagged 133 muskellunge in the Detroit River and Lake St. Clair, and movements of 58 fish that passed our data quality control screens were analyzed. We utilized mixed modelling to assess the effects of sex, length, release location, and season on daily movement rates. We found that movement rates only differed among seasons, with highest movement rates occurring in the fall and lowest movement rates in the winter. Muskellunge tagged at different locations exhibited distinct residency patterns, and fish frequently crossed jurisdictional and waterbody boundaries. Ultimately our study highlights the scope and patterns of muskellunge movement in a large, unimpounded system and demonstrates that management of these fish would benefit from consideration of their full distribution covering multiple management jurisdictions. 相似文献

Mycenarubin C, a previously unknown red pyrroloquinoline alkaloid, was isolated from fruiting bodies of the mushroom Mycena rosea and its structure was elucidated mainly by NMR spectroscopy and mass spectrometry. Unlike mycenarubin A, the major pyrroloquinoline alkaloid in fruiting bodies of M. rosea, mycenarubin C, contains an eight-membered ring with an additional C1 unit that is hitherto unprecedented for pyrroloquinoline alkaloids known in nature. Incubation of mycenarubin A with an excess of formaldehyde revealed that mycenarubin C was generated nearly quantitatively from mycenarubin A. An investigation into the formaldehyde content of fresh fruiting bodies of M. rosea showed the presence of considerable amounts of formaldehyde, with values of 5 μg per gram of fresh weight in fresh fruiting bodies. Although mycenarubin C did not show bioactivity against selected bacteria and fungi, formaldehyde inhibits the growth of the mycoparasite Spinellus fusiger at concentrations present in fruiting bodies of M. rosea. Therefore, formaldehyde might play an ecological role in the chemical defence of M. rosea against S. fusiger. In turn, S. fusiger produces gallic acid—presumably to detoxify formaldehyde by reaction of this aldehyde with amino acids and gallic acid to Mannich adducts. 相似文献

The aim of this study is to develop an industrial ecosystem whereby wastes/products from a Post-combustion CO2 Capture (PCC) plant are utilised in a hydrogen biorefinery. Subsequently, five hydrogen biorefinery models are developed that use PCC's model amine i.e. monoethanolamine (MEA) as a nitrogen source during microbial hydrogen production and CO2 as a process chemical. Technical evaluations of the five case models are carried out to identify the ones that maximise value by multiproduct generation from biomass and fulfil total/partial parasitic energy demand. The case meeting these criteria, produces 3.1t of succinylated lignin adhesive, 4.9t of dry compost and 2744 kWh of electricity from 10t (dry) of sawdust feedstock, daily. Its daily power and heat duties stand at 3906 kWh and 52.1 GJ respectively. Simulations also demonstrate biohydrogen's potential as an energy storage vector for peak/backup power with an annual 1001.4 MWh of power storage capacity from 10t/d feedstock. 相似文献

Information systems, like biological systems, are susceptible to external perturbations. Similar to flora and fauna in a biome, species of data can be classified within a dataphora. While entropic properties and data geometries can be used to describe local species of data within a dataphora, they are not designed to describe the global properties of an information system or evaluate its stability. Ecologists have used Information Theories to describe macro-level properties of biological ecosystems and statistical tools to evaluate biological systems. This research leverages an ecological perspective to model information systems as a living system. Our findings support the theory of dataphoric ascendancy with Wikipedia having a Diversity Index value of 0.68, within the range of 0.65 and 0.80 that indicates a balanced state. We further support our findings with additional evaluations of other ecosystems including the predicted collapse of the information service known as the Digital Universe. This research allows for an information system’s stability to be (a) characterized and (b) predicted using ecological measures specific to the diversity of data within the ecosystem. 相似文献
