Carbon monoxide (CO)—gaseous or released by CO-RMs—both possess antiplatelet properties; however, it remains uncertain whether the mechanisms involved are the same. Here, we characterise the involvement of soluble guanylate cyclase (sGC) in the effects of CO—delivered by gaseous CO–saturated buffer (COG) and generated by CORM-A1—on platelet aggregation and energy metabolism, as well as on vasodilatation in aorta, using light transmission aggregometry, Seahorse XFe technique, and wire myography, respectively. ODQ completely prevented the inhibitory effect of COG on platelet aggregation, but did not modify antiplatelet effect of CORM-A1. In turn, COG did not affect, whereas CORM-A1 substantially inhibited energy metabolism in platelets. Even though activation of sGC by BAY 41-2272 or BAY 58-2667 inhibited significantly platelet aggregation, their effects on energy metabolism in platelets were absent or weak and could not contribute to antiplatelet effects of sGC activation. In contrast, vasodilatation of murine aortic rings, induced either by COG or CORM-A1, was dependent on sGC. We conclude that the source (COG vs. CORM-A1) and kinetics (rapid vs. slow) of CO delivery represent key determinants of the mechanism of antiplatelet action of CO, involving either impairment of energy metabolism or activation of sGG. 相似文献

目前,测定柴油十六烷值的试验机价格较高,现有十六烷指数预测精度低,为满足炼油厂生产柴油在线调合的需要,迫切需要建立预测精度高的柴油十六烷值预测模型。基于450个具有代表性的柴油样本,建立了柴油理化性质、烃族组成与十六烷值数据库;进而采用逐步回归分析方法,应用统计产品和服务解决方案(SPSS)软件,建立了基于柴油理化性质的十六烷值预测模型和基于柴油烃族组成的十六烷值预测模型。采用F检验、T检验、残差分析验证了上述模型的有效性,并通过计算均方根误差,比较了上述两个模型的精度,结果表明,两种预测模型均有效,基于理化性质模型的预测精度优于基于烃族组成的模型。 相似文献

为了提高黄浆水的附加值和利用率,缓解排放黄浆水带来的环境污染,本文对黄浆水中的可溶性固形物进行絮凝处理。在单因素实验基础上结合响应面法对黄浆水中可溶性固形物的絮凝工艺进行优化,并且对絮凝前后黄浆水进行了蛋白质和总糖含量测定、生化需氧量和化学需氧量测定以及代谢组学测定。实验结果表明,絮凝沉降黄浆水可溶性固形物最佳工艺参数为:壳聚糖添加量为0.54 mg/mL,海藻酸钠添加量为0.24 mg/mL,pH为4.4,温度为47 ℃,此时黄浆水可溶性固形物沉降率为(46.26%±0.38%);蛋白质含量下降40.68%、总糖含量下降8.41%;生化需氧量和化学需氧量去除率分别为43%、40.95%。代谢组学实验结果表明:黄浆水絮凝前后有66种化合物含量存在显著变化。本研究结果表明絮凝能有效的降低黄浆水中可溶性固形物的含量,为黄浆水的进一步开发利用提供一定的参考。 相似文献

The provision of pasture and outdoor access for dairy cattle differs around the globe. For example, in Ireland, New Zealand, and Australia, dairy farms are largely pasture based, whereas dairy farms in the United States and Canada are largely confinement based. There is a high level of public support for pasture and outdoor access for dairy cows, and the available evidence shows that dairy cattle are highly motivated to access pasture, especially at night. The decision as to whether to provide outdoor access is typically made by farmers, but little is known about dairy farmers' perspectives on this topic. We investigated perspectives of Western Canadian dairy farmers on outdoor access, as well as how they believe different stakeholders (i.e., the dairy industry, the dairy cows, and the general public) regard outdoor access for dairy cows. Data were collected via (1) 11 focus group discussions with a total of 50 Western Canadian dairy farmers, and (2) semi-structured individual interviews with an additional 6 dairy farmers of Hutterite colonies. Data were analyzed using template analysis. Although most participants in this study did not provide outdoor access on their farms, or only provided outdoor access to certain cow groups, participants generally mentioned that they enjoyed seeing cows on pasture or outdoors. However, participants shared that the Canadian supply management system (including processors) required a consistent flow of production, which was thought to be easier and more economically realized with indoor housing of lactating cows. Participants believed that pasture or outdoor access for dairy cows was desired by the public. Some participants believed that dairy cows prefer to spend time outside under favorable weather conditions, but others felt that cows preferred to stay indoors in modern, ventilated freestall barns. The results of this study describe the perspectives of dairy farmers regarding the views of dairy industry stakeholders as they relate to outdoor access, helping to inform conversations around the provision of outdoor access for dairy cattle. 相似文献

为改善红花籽粕可溶性膳食纤维的部分理化性质和其吸附特性,以红花籽粕为原料,分别考察碱-酶法、酶-高温蒸煮法、碱-高温蒸煮法3种不同改性方式对其可溶性膳食纤维(SDF)的持水力、膨胀力、持油力等部分理化性质及对葡萄糖、阳离子、胆固醇和亚硝酸根离子吸附能力的影响。结果表明,碱-高温蒸煮法优于其他两种方法,碱-高温蒸煮法改性的红花籽粕SDF的持水力、膨胀力和持油力最佳,分别为5.58 g/g、3.98 mL/g和4.38 g/g;对葡萄糖吸附能力为16.08 mmol/g,在1% NaOH添加量为1~4 mL时,阳离子吸附效果最佳;在pH为2和7时,对胆固醇吸附能力分别为7.68 mg/g、10.14 mg/g,对亚硝酸盐吸附能力为56.43 μg/g、30.53 μg/g。 相似文献

以小南海岩溶水为研究对象,通过分析水化学和氢氧同位素数据,确定岩溶水中主要物质组分来源及补给径流特征,对岩溶水的合理开发利用及生态环境保护具有重要意义。结果显示:所有水样均为弱碱性、低矿化度水,水化学类型以HCO3-Ca型为主;岩溶水溶质组分受控于岩石风化作用,主要来源于碳酸盐岩类和部分硫酸盐岩类矿物溶解,此外还受人类活动影响;吴家坪组、阳新组、仙女洞组岩溶水以流经灰岩含水层为主,大冶组岩溶水流经灰岩和白云岩含水层。氢氧同位素特征指示岩溶水均源于不同海拔大气降水的混合补给,降水入渗补给前,岩溶水受到的蒸发作用强于地表水。水化学特征和氘过量参数揭示小南海岩溶水系统径流条件好,溶滤作用较强,双龙洞岩溶水系统径流条件好,溶滤作用较弱。 相似文献

Accessing aldehydes from carboxylate moieties is often a challenging task. In this regard, carboxylate reductases (CARs) are promising catalysts provided by nature that are able to accomplish this task in just one step, avoiding over-reduction to the alcohol product. However, the heterologous expression of CARs can be quite difficult due to the excessive formation of insoluble protein, thus hindering further characterization and application of the enzyme. Here, the heterologous production of the carboxylate reductase from Nocardia otitidiscaviarum (NoCAR) was optimized by a combination of i) optimized cultivation conditions, ii) post-translational modification with a phosphopantetheinyl transferase and iii) selection of an appropriate expression strain. Especially, the selection of Escherichia coli tuner cells as host had a strong effect on the final 110-fold increase in the specific activity of NoCAR. This highly active NoCAR was used to reduce sodium benzoate to benzaldehyde, and it was successfully assembled with an in vitro regeneration of ATP and NADPH, being capable of reducing about 30 mM sodium benzoate with high selectivity in only 2 h of reaction. 相似文献

采用超声预处理-柠檬酸辅助亚临界水提法从小麦麸皮(以下简称“麦麸”)中提取水溶性膳食纤维。通过单因素实验考察超声预处理功率、柠檬酸/麦麸液固比、亚临界水提取温度和时间对麦麸水溶性膳食纤维得率的影响,在此基础上,采用响应面优化法,对提取工艺参数进行优化。结果表明,最佳提取条件为:超声预处理功率195 W,柠檬酸/麦麸液固比39:1 mL/g,亚临界水提取温度和时间分别为179 ℃和30 min。此时,麦麸水溶性膳食纤维的得率为41.00% ± 0.29%。因此,该方法能够提高麦麸水溶性膳食纤维的得率,且具有提取时间短、绿色、环保等优点,为工业化生产麦麸水溶性膳食纤维提供技术参考。 相似文献
