Expansion microscopy combined with single-molecule localization microscopy (ExSMLM) has a potential for approaching molecular resolution. However, ExSMLM faces multiple challenges such as loss of fluorophores and proteins during polymerization, digestion or denaturation, and an increase in linkage error arising from the distance between the fluorophore and the target molecule. Here, we introduce a trifunctional streptavidin to link the target, fluorophore and gel matrix via a biotinylizable peptide tag. The resultant ExSMLM images of vimentin filaments demonstrated high labeling efficiency and a minimal linkage error of ∼5 nm. Our ExSMLM provides a simple and practical means for fluorescence imaging with molecular resolution.  相似文献

A large-scale high-precision scan stage is important equipment in the industrial productions of micro-fabrication such as flat panel display (FPD) lithography systems. Designing controllers for multi-input multi-output (MIMO) systems is time-consuming and needs experience because of the interaction between each axis and many controller tuning parameters. The aim of this study is to develop a peak filter design method based on frequency response data to reduce repetitive disturbance. This data-based approach does not use the model and only uses the frequency response data of the controlled system and the disturbance spectrum calculated from the scanning error data (Contribution 1). The peak filter is designed by convex optimization and satisfies robust stability conditions for six-degree-of-freedom systems (Contribution 2). The control performance of the designed peak filter is experimentally demonstrated with an industrial MIMO large-scale high-precision scan stage in reducing the scanning error of the main stroke of the translation along the x-axis (Contribution 3).  相似文献

One of the most challenging issues in radio received signal strength (RSS)-based localization systems is the generation and distribution of a radio map with a coordinate system linked with spatial information in a large indoor space. This study proposes a novel spatial-tagged radio-mapping system (SRS) that effectively combines the heterogeneous properties of LiDAR and mobile phones to simultaneously perform both spatial and radio mappings. The SRS consists of synchronization, localization, and map building processes, and enables real-time spatial and radio mapping. In the synchronization process, the distance range, motion data, and radio signals obtained through the LiDAR and mobile phone are collected in nodal units according to the sensing time. In the localization process, a feature variance filter is used to control the number of features generated from LiDAR and estimate the positions at which the nodes are generated in real time according to the motion data and radio signals. In map building, the estimated positions of the nodes are used to extract spatial and radio maps by using a unified location coordinate system. To ensure mobility, the SRS is manufactured in the form of a backpack supporting LiDAR and a mobile phone; the usefulness of the system is experimentally verified. The experiments are performed in a large indoor shopping mall with a complex structure. The experimental results demonstrated that a common coordinate system could be used to build spatial and radio maps with high accuracy and efficiency in real time. In addition, the field applicability of the SRS to location-based services is experimentally verified by applying the constructed radio map to well-known fingerprinting algorithms using the heterogeneous mobile phones.  相似文献

Coke deposition has been considered to be one of the most important reasons hindering the stability of the catalyst during CH4/CO2 reforming. In this study, after the addition of P123 (PEG-PPG-PEG triblock copolymer), Ni2+ can be well-dispersed on the mesoporous molecular sieve MCM-41. And then, the catalysts were prepared by using N2 radio frequency (RF) discharge plasma for different treatment times to reduce the size of Ni particles, improve the anti-coking performance, and thereby improve the stability of the catalyst. The results showed that the catalyst NM-P123-PN2h exhibits superior catalytic properties in the CH4/CO2 reforming. The initial conversions of CH4 and CO2 were 90.80% and 89.60% at 750 °C, respectively. The catalyst NM-P123-PN2h showed highly coke resistance with less carbon deposition (1.12%) at 750 °C after 10 h of continuous reaction, while the carbon deposition of the catalyst NM-C was 37.32%. Compared with the traditional calcination method, the catalyst prepared by plasma treatment has a smaller particle size and better dispersibility of nickel. In particular, the nickel particle size of the catalyst NM-C was 8.37 nm, however, that of the catalyst NM-P123-PN2h was only 1.70 nm, and the nickel particle size was reduced by 5 times. Therefore, it can be concluded that the catalyst prepared under the combined action of P123 and RF plasma-treated can effectively improve the coke resistance of the catalyst and the stability of the CH4/CO2 reforming.  相似文献

KDP/DKDP倍频晶体是惯性约束聚变系统中的关键元件,其主截面方向与晶体相位匹配角、晶体吸收系数紧密相关。为了实现KDP/DKDP倍频晶体主截面方向的高精度定位,本文提出一种光强测量间接定位方法。通过激光器结合稳功率仪及半波片输出稳定线偏振光,同时旋转相互正交的起偏器与检偏器可获得晶体的最佳消光位置即为主截面方向。推导了该测量系统光强的琼斯矩阵模型,给出了光强与起偏器、检偏器角度间的关系表达式。采用最小二乘方法拟合经过起偏器、倍频晶体及检偏器的光强变化曲线,从而可精确定位倍频晶体主截面的方向。通过计算机仿真模拟和实验验证了该方法的正确性和可行性。实验表明,该方法定位的重复测量精度优于0.02°,满足惯性约束聚变系统中KDP/DKDP倍频晶体主截面的定位控制精度要求。  相似文献

随着基于位置服务(Location Based Services,LBS)的发展与智能移动设备的普及,室内定位算法与系统受到了广泛研究与关注。为提高室内定位精度、增强系统鲁棒性,提出了基于多边限定的fingerprint定位方法。基于Wi-Fi RSSI(Received Signal Strength Indication)信号处理建立离线fingerprint数据库;通过对拟合距离-RSSI函数分析,提出了多边限定的方法确定一个最佳参考点(Reference Point,RP)集合,缩小在线定位阶段的搜索范围。在此基础上,再利用fingerprint定位方法进行定位。此外,实现了基于提出方法的室内定位系统原型用于算法性能评估。通过大量真实场景实验分析、验证了相较于传统fingerprint方法,基于多边限定的fingerprint定位方法能有效提高室内定位精度,增强系统鲁棒性。  相似文献

This paper presents a hierarchical framework for managing the sampling distribution of a particle filter (PF) that estimates the global positions of mobile robots in a large‐scale area. The key concept is to gradually improve the accuracy of the global localization by fusing sensor information with different characteristics. The sensor observations are the received signal strength indications (RSSIs) of Wi‐Fi devices as network facilities and the range of a laser scanner. First, the RSSI data used for determining certain global areas within which the robot is located are represented as RSSI bins. In addition, the results of the RSSI bins contain the uncertainty of localization, which is utilized for calculating the optimal sampling size of the PF to cover the regions of the RSSI bins. The range data are then used to estimate the precise position of the robot in the regions of the RSSI bins using the core process of the PF. The experimental results demonstrate superior performance compared with other approaches in terms of the success rate of the global localization and the amount of computation for managing the optimal sampling size.  相似文献

Frequency band selection (FBS) in rotating machinery fault diagnosis aims to recognize frequency band location including a fault transient out of a full band spectrum, and thus fault diagnosis can suppress noise influence from other frequency components. Impulsiveness and cyclostationarity have been recently recognized as two distinctive signatures of a transient. Thus, many studies have focused on developing quantification metrics of the two signatures and using them as indicators to guide FBS. However, most previous studies almost ignore another aspect of FBS, i.e. health reference, which significantly affect FBS performance. To address this issue, this paper investigates importance of a health reference and recognize it as the third critical aspect in FBS. With help of the health reference, the frequency band where the fault transient exists could be located. A novel approach based on classification is proposed to integrate all three aspects (impulsiveness, cyclostationarity, and health reference) for FBS. Classification accuracy is developed as a novel indicator to select the most sensitive frequency band for rotating machinery fault diagnosis. The proposed method (coined by accugram) has been validated on benchmark and experiment datasets. Comparison results show its effectiveness and robustness over conventional envelope analysis, the kurtogram, and the infogram.  相似文献

This paper presents a digital constant frequency sliding mode control (SMC) law for interleaved DC–DC converters. Constant switching frequency and interleaving are achieved by dynamically adjusting the hysteresis of the control signals generating comparators. The interleaving method neither imposes constraints on the number of required phases to obtain a specific output voltage nor uses quasi-SMC. Hence, the control scheme achieves high flexibility, robustness, and performance. Furthermore, a sliding mode observer (SMO) for reconstructing the inductor currents is proposed. Experimental results for a two-phase buck converter are reported. The control concept accomplishes an improved dynamic performance in comparison with quasi-SMC.  相似文献
