Isolation and preliminary characterization of Pichia pinus mutants insensitive to glucose repression
A new method for the isolation of glucose repression-insensitive mutants in the methylotrophic yeast Pichia pinus was developed. The method is based on screening of small suspension samples derived from 2-deoxyglucose-resistant colonies for alcohol oxidase activity. Alcohol oxidase activity was evaluated by determination of formaldehyde excreted by cells. Mutants with glucose non-repressible alcohol oxidase and catalase synthesis were obtained. All mutants grew poorly on D -xylose compared to the wild type, whereas growth on L -arabinose was similar to the wild type. Changes in the glucose transport system were suggested to be responsible for altered growth characteristics and defective glucose repression. 相似文献

Individual yeast cells can be successfully isolated and recultured on plates with a new isolation method making use of optical trapping with infrared laser light. The cells can be selected on morphological criteria by high resolution microscopy. The isolation device is constructed from two coverslips separated by spacers, in which selected cells are transferred to a plastic capillary, using the optical trap. To test the procedure, selection experiments were done with a mixture of two Saccharomyces cerevisiae strains, distinguishable both in fluorescence microscopy and on agar plates. These experiments showed that only selected cells were isolated, and close to 100% of the isolated stationary-phase cells formed colonies on agar plates, indicating a high recovery. A lower recovery was obtained with exponential-phase cells, possibly because of a higher sensitivity to laser irradiation. Applications for this method may include the isolation of mutants with altered morphology and the isolation of subpopulations of yeast cultures, for their separate investigation or for the initiation of pure cultures. 相似文献

In this paper, we develop a futures trading simulation system to determine how speculative behavior affects the futures mar ket. A eonfigurable client is designed to simulate traders, and users can define trade strategies using different programming languages. A lightweight server is designed to handle large scale and highly eoncmTent access requests from clients. HBase is chosen as the database to grantee sealability of the system. As HBase only supports single row transaction, a transaction support mechanism is developed to improve data consistency for HBase. The HBase transaction support mecha nism supports multirow and multitaMe by using two phase commit protocol. The experiments indicate that our system shows high efficiency in the face of the large scale and high concurrency access request, and the read/write performance loss of HBase introduced by the transaction support mecha nisms is also acceptable. 相似文献

Controlled systems can be subjected to faults that may affect the performance of the system, and unable its objectives to be achieved. Fault detection and isolation algorithms are then used to study these faults. The bond graph tool can be used for modeling purposes and then its structural, and causal properties can be exploited for automatic generation of analytical redundancy relations (ARRs) through a procedure named causality inversion method, which are used for diagnosis applications. These ARRs are mathematical constraints that are used to verify the coherence between the process measurements and the system model. This paper proposes an extension of the causality inversion method by different versions of the same ARR. The goal is to increase the number of isolable faults. Moreover, structural conditions are given in order to avoid the generation of redundant ARRs. A combination of two conventional junction isolation structures is used to produce a device, which significantly improves the blocking of minority carriers injected into the substrate of a power IC due to switching of an inductive load. Simulation results show that the connection scheme employed greatly enhances the efficiency of the structures. A substrate current reduction of up to four orders of magnitude compared to conventional junction isolation structures is achieved.
