1,25-Dihydroxycholecalciferol, the hormonally active vitamin D3 metabolite, is known to exhibit therapeutic effects against breast cancer, mainly by lowering the expression of estrogen receptors and aromatase activity. Previously, the safety of the vitamin D active metabolite (24R)-1,24-dihydroxycholecalciferol (PRI-2191) and 1,25(OH)2D3 analog PRI-2205 was tested, and the in vitro activity of these analogs against different cancer cell lines was studied. We determined the effect of the two vitamin D compounds on anastrozole (An) activity against breast cancer based on antiproliferative activity, ELISA, flow cytometry, enzyme inhibition potency, PCR, and xenograft study. Both the vitamin D active metabolite and synthetic analog regulated the growth of not only estrogen receptor-positive cells (T47D and MCF-7, in vitro and in vivo), but also hormone-independent cancer cells such as SKBR-3 (HER-2-positive) and MDA-MB-231 (triple-negative), despite their relatively low VDR expression. Combined with An, PRI-2191 and PRI-2205 significantly inhibited the tumor growth of MCF-7 cells. Potentiation of the antitumor activity in combined treatment of MCF-7 tumor-bearing mice is related to the reduced activity of aromatase by both An (enzyme inhibition) and vitamin D compounds (switched off/decreased aromatase gene expression, decreased expression of other genes related to estrogen signaling) and by regulation of the expression of the estrogen receptor ERα and VDR.  相似文献

针对模拟电路健康管理的特点,提出了一种基于PSO优化多核RVM的模拟电路故障预测方法。利用参数分析得到电路的输出频域响应作为特征,计算其与电路无故障标准响应的欧氏距离来表征电路元件健康值,将多个核函数线性组合,并用PSO优化多核RVM参数后的模型实现对各个时间点元件的健康值变化轨迹进行预测。仿真结果表明,该方法在小样本情况下,预测效果优于单一核函数的RVM模型,适用于健康管理中实时预测,具有较好的实用性。  相似文献

为了在嵌入式控制系统中充分利用超声电机特有功能,对超声电机的驱动原理进行了研究,在分析了超声电机的驱动控制原理的基础上,提出了一种基于单片机控制超声电机的软硬件控制实现方法。利用单片机输出数字量作为控制信号,通过串行输入并行输出芯片扩展后,输出到超声电机驱动器,实现超声电机启动/停止/正反转的功能;通过扩展单片机数模转换,将单片机控制超声电机速度的数字信号转换为电压信号,实现超声电机的速度控制功能;利用运行脉冲宽度控制实现超声电机的位置控制功能,并将光电编码器实现超声电机的反馈,最后通过试验验证了单片机系统实现超声电机驱动设计。  相似文献

A 4th-order Butterworth class-AB current-mode low-pass filter is proposed, based on second-generation Current Conveyors (CCII). Class-AB operation allows high-power efficiency and driving large loads with small quiescent currents. The CCII topology uses the class-AB output buffer with error amplifiers: this topology is known to be sensitive to mismatch errors, which cause offsets in the error amplifiers, affecting the biasing current of the stage. This problem is solved via a control loop, which compensates the effect of mismatches. The technique is shown to be effective in Monte Carlo simulations with process variations and mismatches. Simulations have been carried out in 40 nm CMOS technology. The proposed filter achieves good power efficiency, thanks to the class-AB architecture, and good dynamic range, thanks to the closed-loop output buffer. A cut-off frequency of 6 MHz, with 184 μW of total quiescent power consumption, is achieved, with a THD of -55 dB and a SNR of 49 dB.  相似文献

Analog integrated circuit design has as integral parts both analytical reasoning and numerical validation in the process from topology construction to sizing. Given a circuit topology, different circuit sizing results can be obtained from different processes of sizing inference. Sizing methods by simulation-based numerical searching have been a continuously studied subject. However, almost all approaches in this category require an overwhelming number of circuit simulations to arrive at an optimized sizing result. On the other hand, many published manual sizing methods by using the conventional device equations also require repeated SPICE simulations to correct the equation-based sizing results. This paper proposes a systematic gm/ID-based initial sizing method specifically customized for designing multiple-stage operational amplifiers (Op Amps). A main feature of the proposal is to use circuit-level design equations as constraints on the gm/ID table lookup method to substantially reduce the uncertainty in the sizing calculations. As a result, a significant amount of SPICE based correction work can be reduced to complete an initial sizing. The proposed sizing procedure includes a few regular sizing rules customized to the configuration of multi-stage Op Amps. We validate the proposed sizing method by application to several multi-stage Op Amp examples with a capacitive load or Miller compensation. Simulations have justified that the produced initial sizing results can achieve most of the prespecified design targets.  相似文献

Isobutane alkylation with 2-butene to produce high-quality gasoline was catalyzed by Nmethylacetamide(NMA)-AlCl_3 based ionic liquid(IL) analog with a NMA/AlCl3 molar ratio of 0.75 and CuCl modification,which was marked as CuCl-modified 0.75 NMA-1.0 AlCl_3.The long-term experiment was carried out in the autoclave operated in continuous mode to investigate the distribution of alkylate under different experimental nodes.The result indicated that the long-term alkylation was divided into three stages:rising,stable,and descending regions.C8 selectivity and molar ratio of trimethylpentanes(TMPs) to dimethylhexanes(DMHs) reached the highest level in the stable region,and research octane number(RON) of alkylate was as high as 97.Anionic Al species([AlbCl_7]~-,[A1 CuC15]-) and cationic Al species([AlCl_2 L]~+) from IL analog as two active Lewis acidic species played a catalytic role in the long-term alkylation,whereas the neutral Al species did not participate into the alkylation.Moreover,the structure of CuCl-modified 0.75 NMA-1.0 AlCl_3 was destroyed after the deactivation,and CuCl was enriched in the CD_2 Cl_2-insoluble substance,resulting in a decreasing TMP/DMH ratio.The catalytic lifetime of IL analog was similar with CuCl-modified 0.55 Et_3 NHCl-1.0 AlCl_3 IL,but IL analog had a lower cost.  相似文献

The delivery of siRNAs to selectively target cells poses a great challenge in RNAi-based cancer therapy. The lack of suitable cell-targeting methods seriously restricts the advance in delivering siRNAs to extrahepatic tissues. Based on prostate-specific membrane antigen (PSMA)-targeting ligands, we have synthesized a series of lysine-urea-glutamate (KUE)-siRNA conjugates and verified their effective cell uptake and gene silencing properties in prostate cancers. The results indicated that the KUE-siRNA conjugates could selectively enter PSMA+ LNCaP cells, eventually down-regulating STAT3 expression. Based on post-synthesis modification and receptor-mediated endocytosis, this strategy of constructing ligand-siRNA conjugates might provide a general method of siRNA delivery for cell-targeted gene silencing.  相似文献

通过动电位极化曲线法,在几种含不同氯离子浓度的典型碱性水溶液中研究了模拟古代灰口铸铁、白口铸铁的电化学行为。结果表明,在不同碱性溶液中同种铸铁试样对氯离子浓度的钝化/活化敏感性不同;不同材质的铸铁试样在相同溶液中对氯离子的敏感性也不相同。并且,对于同种溶质的碱性溶液,铸铁对氯离子的敏感性则随着溶液的pH值增加而降低。这一结果将为铁质文物脱盐除氯的终点判定提供重要的依据。  相似文献
