The selenol group of selenocysteine is much more nucleophilic than the thiol group of cysteine. Selenocysteine residues in proteins thus offer reactive points for rapid post-translational modification. Herein, we show that selenoproteins can be expressed in high yield and purity by cell-free protein synthesis by global substitution of cysteine by selenocysteine. Complete alkylation of solvent-exposed selenocysteine residues was achieved in 10 minutes with 4-chloromethylene dipicolinic acid (4Cl-MDPA) under conditions that left cysteine residues unchanged even after overnight incubation. GdIII−GdIII distances measured by double electron–electron resonance (DEER) experiments of maltose binding protein (MBP) containing two selenocysteine residues tagged with 4Cl-MDPA-GdIII were indistinguishable from GdIII−GdIII distances measured of MBP containing cysteine reacted with 4Br-MDPA tags.  相似文献

RDFa, JSON-LD, Microdata, and Microformats allow to endow the data in HTML files with metadata tags that help software agents understand them. Unluckily, there are many HTML files that do not have any metadata tags, which has motivated many authors to work on proposals to synthesize them. But they have some problems: the authors either provide an overall picture of their designs without too many details on the techniques behind the scenes or focus on the techniques but do not describe the design of the software systems that support them; many of them cannot deal with data that are encoded using semistructured formats like forms, listings, or tables; and the few proposals that can work on tables can deal with horizontal listings only. In this article, we describe the design of a system that overcomes the previous limitations using a novel embedding approach that has proven to outperform four state-of-the-art techniques on a repository with randomly selected HTML files from 40 different sites. According to our experimental analysis, our proposal can achieve an F1 score that outperforms the others by 10.14% ; this difference was confirmed to be statistically significant at the standard confidence level.  相似文献

Large amounts of the manila clam, Ruditapes philippinarum, have been imported from Dalian (China) recently to meet the increasing demand in Korea. To ensure the quality and safety of the domestic marine product, it is necessary to be able to distinguish between domestic manila clams and clams that are imported from Dalian, China. We developed expressed sequence tags (ESTs)-derived single nucleotide polymorphism markers using 454 pyrosequencing. A total of 780,000 ESTs were assembled, and 49,540 putative SNPs were identified from 46,405 contigs. Twenty-one polymorphic SNPs from 11 primers were finally selected as diagnostic markers. Genotype analysis showed that 21 SNP markers had the opposite alleles (transitions or transversions) between domestic and imported samples, which were useful for distinguishing clam origins. The GENECLASS 2.0 program was used to estimate the ability of the markers in the discrimination between domestic and imported populations. The 21 SNP markers had 98.96% ability to discriminate domestic manila clams and 100% ability to identify those imported. These markers could be useful for discriminating between domestic and imported manila clams and can contribute to the prevention of falsified labeling of this species.  相似文献

针对模拟电路的特点和其故障诊断中存在的问题,提出了一种基于输出频谱和支持向量机的新型模拟电路故障诊断方法(FMSVM)。该方法采用多音信号作为模拟电路的激励信号,并用其输出频谱中的有限个频率点的幅值作为故障特征,采用多分类支持向量机(MSVM)进行故障模式判别,实现了模拟电路的故障诊断。实验结果表明,该故障诊断方法具有速度快、准确率高的特点,具有重要的实用价值。  相似文献

Radio‐frequency identification (RFID) technology enables the identification and tracking of objects by means of the wireless signals emitted by a tag attached to the objects of interest. Without adequate protection, however, malicious attackers can easily eavesdrop, scan or forge the information within the tag, thereby threatening the integrity of the system. Previous research has shown that the basic security requirements of RFID systems, i.e. identity authentication, information privacy and location privacy, can be satisfied using conventional cryptographic components. However, such components are expensive, and therefore conflict with the general requirement for low‐cost tag designs. Accordingly, this paper presents a low‐cost challenge‐response security protocol designated as the hidden mutual authentication protocol (HMAP) to accomplish both a mutual authentication capability between the tag and the reader and information privacy. The results show that HMAP provides an efficient means of concealing the authentication messages exchanged between the tag and the reader and is robust toward replay attacks. In addition, it is shown that HMAP is easily extended to provide complete location privacy by utilizing a hash function to generate different tag identifiers in each authentication session.
