The objective of the study was to evaluate the relationship between curing light intensity and (1) linear post-gel polymerization contraction strain, and (2) degree of conversion of a dental composite. Cylindrical specimens of a dental resin composite were cured from a distance of 7 mm for 40 s at four attenuated light intensities (71%, 49%, and 34% of control intensity and for 20 s at 71% plus 20 s at 100% intensity). A group cured at full intensity served as a control. Degree of conversion (DC) was measured at the top and bottom and linear contraction strain was measured at the bottom of the composite samples. DC at the sample top was significantly different (P < 0.05) between all groups except the 71% and 49% intensity groups. At the sample bottom, DC resulting from the two highest intensities (71% and 100%) were not significantly different from each other (P > 0.05). All other groups were significantly different from each other (P < 0.05). DC for the sample cured at two light intensities was not significantly different from those cured at the lower intensity or higher intensity for 40 s (P > 0.05). The sample cured with two intensities showed a 21.8% reduction from the contraction strain predicted by a light energy density calculation. Application of light at less than the maximum intensity of the curing light resulted in significant reduction of polymerization contraction strain without significantly affecting the degree of conversion.  相似文献

Effects of condensed tannins (CT), either via extract or plant-bound, and saponin extract on ruminal biohydrogenation of α-linolenic acid (ALA) were investigated in vitro. Grass-clover hay served as basal diet (control). The control hay was supplemented with extracts contributing either CT from Acacia mearnsii [7.9% of dietary dry matter (DM)] or saponins from Yucca schidigera (1.1% of DM). The fourth treatment consisted of dried sainfoin (Onobrychis viciifolia), a CT-containing forage legume, in an amount also providing 7.9% CT in dietary DM. All diets were supplemented with linseed oil at a level contributing 60% of total dietary ALA in all treatments. Diets were incubated for 10 d (n = 4) in the rumen simulation technique system, using the last 5 d for statistical evaluation. Fatty acids were analyzed in feed, feed residues, incubation fluid, and its effluent. Data were subjected to ANOVA considering diet and experimental run as main effects. Both CT treatments reduced ruminal fiber and crude protein degradation, and lowered incubation fluid ammonia concentration. Only the CT extract suppressed methane formation and shifted microbial populations toward bacteria at cost of protozoa. The saponin extract remained without clear effects on fermentation characteristics except for increased protozoal counts. The extent of ALA biohydrogenation was 20% less with the CT plant, but this probably resulted from reduced organic matter degradability rather than from an inhibition of biohydrogenation. After incubation analysis of incubation fluid effluent and feed residues showed a considerable proportion of the 3 biohydrogenation intermediates, cis-9, trans-11, cis-15 C18:3, trans-11, cis-15 C18:2, and trans-11 C18:1, which did not occur in the initial feeds. Only the CT-extract diet led to a different profile in the effluent compared with the control diet with trans-11 C18:1 being considerably increased at cost of C18:0. This could have been achieved by suppressing protozoa and enhancing the bacterial population, thus removing potential microbes involved in biohydrogenation and increasing competition between bacteria involved in biohydrogenation and others. The elevation of trans-11 C18:1 as the precursor of cis-9, trans-11 conjugated linoleic acid formed in body tissue and mammary gland is probably favorable from a human health point of view.  相似文献

Zearalenone (ZON) is one of the worldwide most common mycotoxins and exhibits estrogenic activity in the range of natural steroid estrogens such as 17β-estradiol (E2). The occurrence of ZON has been reported in drainage water, soil, wastewater effluents and rivers, but its ecotoxicological effects on fish have hardly been investigated.In this study the estrogenic potency of the ZON was compared to E2 in a recombinant yeast estrogen screen (rYES) and the effects of waterborne ZON exposure on reproduction, physiology and morphology of zebrafish (Danio rerio) were investigated in a 42-day reproduction experiment. E2 as well as ZON evoked a sigmoid concentration-response curve in the rYES with a mean EC50 of 2 and 500 μg/L, respectively, resulting in an E2:ZON EC50 ratio of 1:250. Exposure to ZON for 21 days reduced relative spawning frequency at 1000 and 3200 ng/L to 38.9 and 37.6%, respectively, and relative fecundity at 100, 320, 1000 and 3200 ng/L to 74.2, 41.7, 43.8 and 16.7%, respectively, in relation to the 21-day pre-exposure period. A 4.4 and 8.1 fold induction of plasma vitellogenin (VTG) was observed in male zebrafish at 1000 and 3200 ng/L ZON, respectively. Exposure to ZON did not affect fertility, hatch, embryo survival and gonad morphology of zebrafish.The results of this study demonstrate that although ZON possesses a moderate estrogenic potency in vitro, it exhibits a comparably strong effect on induction of VTG and reproduction in vivo. This indicates that ZON might contribute to the overall estrogenic activity in the environment and could therefore pose a risk for wild fish in their natural habitat.  相似文献

Folding of cytochrome c from its low pH guanidine hydrochloride (Gdn-HCl) denatured state revealed a new intermediate, a five-coordinate high spin species with a water molecule coordinated to the heme. Incorporation of this five-coordinated intermediate into the previously reported ligand exchange model can quantitatively account for the observed folding kinetics. In this new model, unfolded cytochrome c is converted to its native structure through an obligatory folding intermediate, the histidine-water coordination state, whereas the five-coordinate state and a bis-histidine state are off-pathway intermediates. When the concentration of Gdn-HCl in the refolding solution was increased, an acceleration of the conversion from the bis-histidine coordinated state to the histidine-water coordinated state was observed, demonstrating that the reaction requires unfolding of the mis-organized polypeptide structure associated with the bis-histidine state.  相似文献

We have previously shown that 1,25-dihydroxyvitamin D3 (1,25(OH)2D3) plays a major role in growth zone chondrocyte (GC) differentiation and that this effect is mediated by protein kinase C (PKC). The aim of the present study was to identify the signal transduction pathway used by 1,25(OH)2D3 to stimulate PKC activation. Confluent, fourth passage GC cells from costochondral cartilage were used to evaluate the mechanism of PKC activation. Treatment of GC cultures with 1,25(OH)2D3 elicited a dose-dependent increase in both inositol-1,4,5-trisphosphate and diacylglycerol (DAG) production, suggesting a role for phospholipase C and potentially for phospholipase D. Addition of dioctanoylglycerol to plasma membranes isolated from GCs increased PKC activity. Neither pertussis toxin nor choleratoxin had an inhibitory effect on PKC activity in control or 1,25(OH)2D3-treated GCs, indicating that neither Gi nor Gs proteins were involved. Phospholipase A2 inhibitors, quinacrine, OEPC (selective for secretory phospholipase A2), and AACOCF3 (selective for cytosolic phospholipase A2), and the cyclooxygenase inhibitor indomethacin decreased PKC activity, while the phospholipase A2 activators melittin and mastoparan increased PKC activity in GC cultures. Arachidonic acid and prostaglandin E2, two downstream products of phospholipase A2 action, also increased PKC activity. These results indicate that 1,25(OH)2D3-dependent stimulation of PKC activity is regulated by two distinct phospholipase-dependent mechanisms: production of DAG, primarily via phospholipase C and production of arachidonic acid via phospholipase A2.  相似文献

Since E. W. Dijkstra's work on the mutual interdependencies of co-operating sequential processes there has been much discussion on various synchronizing mechanisms. This discussion usually leads to abstract definitions of synchronizing primitives. However, their realization proceeds differently in various environments. Length of critical regions, number of processors, and dispatching strategy have to be taken into account. In this paper is presented a family of synchronizing operations, the members of which are all based on the same fundamentals. BACKGROUND: Two experiments were conducted to compare the effects of white and red clover on the fatty acid composition of milk fat from cows fed ryegrass‐based diets. In experiment 1, fresh ryegrass was mixed with white or red clover (60/40, on a dry matter (DM) basis). Experiment 2 involved similar mixed diets in ensiled form, and one ryegrass‐silage diet without the addition of clover. RESULTS: Total DM intake, milk yield and milk fat content were not affected by dietary treatments. Feeding freshly cut white versus red clover supplemented diets resulted in a higher proportion of n‐3 fatty acids, especially α‐linolenic acid, in milk fat. Addition of any clover silage to ryegrass silage increased the proportions of n‐3 fatty acids in milk fat, and reduced the proportions of conjugated linoleic acids (CLA), including C18:2 c9t11, and C18:1cis isomers. The n‐6/n‐3 ratio was elevated compared to the diet not supplemented with clover. CONCLUSION: White clover seemed to be slightly superior to red clover supplementation, but the effects of clover supplementation as such were generally larger than that of clover type. However, a higher concentration of n‐3 fatty acids with clover supplementation coincided with a lower concentration of CLA in the milk fat. Copyright © 2007 Society of Chemical Industry  相似文献
