The aim of this study was to estimate the hydrolytic degree and antioxidant activity of purified casein characterised by different haplotypes (αs1-, β- and k-casein) after in vitro digestion with two different enzymatic systems: pepsin from porcine gastric mucosa (EP) and a crude enzymatic extract from the edible mushroom Pleurotus eryngii. The used enzymes showed a different mode of casein catalysis with a consequent production of peptides of different antioxidant activity. The CN haplotype significantly influenced peptides production; in fact, the amino acid substitutions caused by genetic polymorphisms at the αs1-, β- and k-CN loci influenced the sites of enzymatic cleavage and therefore the produced peptides. The above is evidenced by the different antioxidant activity found in the hydrolysates depending on the used enzymatic system, the CN haplotype, and the CN haplotype × enzymatic treatment interaction. The findings of this study are a perspective for the production of specific foods that exert a biological effect in addition to the nutritional one. 相似文献

Herein we describe the synthesis of a series of new potent tachykinin NK2 receptor antagonists by the modulation of the C‐ and N‐terminal moieties of ibodutant (MEN 15596, 1 ). The N‐terminal benzo[b]thiophene ring was replaced by different substituted naphthalenes and benzofurans, while further modifications were evaluated at the C‐terminal tetrahydropyran moiety. Most compounds demonstrated a high affinity for the human NK2 receptor and high in vitro antagonist potency, indicating that a wide range of substituents at both termini can be incorporated in the molecule without detrimental effects on the interactions with the NK2 receptor. Selected compounds were tested in vivo confirming their activity as NK2 antagonists. In particular, after both iv and id administration to guinea pig, compound 61 b was able to antagonize NK2‐induced colonic contractions with a potency and duration‐of‐action fully comparable to the reference compound 1 (MEN 15596, ibodutant). 相似文献

Partitioning of long-lived minor actinides (americium and curium) from the nuclear wastes issuing the reprocessing of nuclear spent fuels, in order to transmute them into short-lived nuclides or to condition them into stable crystalline matrices, was the subject of intense research within the NEWPART research program of the European 4th Frame Work Program, FWP (1996–1999). The target waste considered was the acidic raffinate (HAR) issuing the reprocessing of the used nuclear fuels by the PUREX process. A two step separation process based on liquid-liquid extraction was designed. The first step consists in the co-separation of the mixture of trivalent actinides and lanthanides from the HAR by extraction with a malonamide extractant (DIAMEX process), while the second step concerns the actinides(III)/lanthanides(III) group separation (SANEX process). Several DIAMEX and SANEX processes were developed and successfully tested with cold, spiked and genuine high active effluents. The research carried out also included basic and fundamental works in order to better understand the relationships between the structures of the extractants and their affinities for the target metal ions. The lecture highlighted both the basic and applied aspects of the research. This work will be pursued (PARTNEW program) within the 5th FWP of the European Union during the period 2000–2003. 相似文献

BACKGROUND: The detection of pigments and colourless flavonoids in apples can provide a useful indication of fruit quality. Optical methods are preferable because they are fast and non‐destructive. In this study, a fluorescence‐based portable sensor was used in order to non‐invasively determine the content of chlorophylls, anthocyanins and flavonols in Fuji, Granny Smith and Golden Delicious apple cultivars. The aim was to define new non‐destructive optical indices of apple quality. RESULTS: The anthocyanin index (ANTH) in Fuji was higher in the sunny (i.e. sun‐exposed) side of the fruit compared to the shady side. For all cultivars, the flavonol index (FLAV) was higher in the sunny side compared with the shady side. The chlorophyll index (CHL) for the shady sides of Granny Smith and Golden Delicious was significantly higher than for the sunny sides. Fine linear regressions were found between the ANTH, FLAV and CHL indices and the actual anthocyanin, flavonol and chlorophyll concentrations, respectively, which were determined destructively on the apple peel extracts. A negative correlation was found between the apple sugar content and the chlorophyll fluorescence in the far‐red spectral band. CONCLUSION: Our results indicate that a single multiparametric fluorescence‐based sensor can provide valuable non‐destructive markers of ripening and quality in apples. The development of optical sensors for spacecraft applications requires that all components be as lightweight as possible. One method to reduce the weight of a multispectral optical system is to eliminate beamsplitting optics and multiple detectors by patterning a filter array directly onto a CCD. However, techniques commonly used in the production of these filter arrays result in decreased image resolutions. This can greatly impact the performance of sensors used for applications such as planetary probes. To address this issue, we have studied the patterning of multilayer dielectric optical coatings in a small scale, two dimensional array, which will allow development of a four color sensor with a resolution one-half that of monochromatic sensors (compared to one-fourth or less for a four color striped array). We have developed ion milling techniques for the preparation of optical filter arrays which are patterned on a scale as small as 7.5 μm, enabling each pixel of a CCD to have its own associated filter. This paper presents details of the fabrication of these multispectral arrays, and discusses problems associated with pixel-sized filters. 相似文献

The enzyme Zmp1 is a zinc‐containing peptidase that plays a critical role in the pathogenicity of Mycobacterium tuberculosis. Herein we describe the identification of a small set of Zmp1 inhibitors based on a novel 8‐hydroxyquinoline‐2‐hydroxamate scaffold. Among the synthesized compounds, N‐(benzyloxy)‐8‐hydroxyquinoline‐2‐carboxamide ( 1 c ) was found to be the most potent Zmp1 inhibitor known to date, and its binding mode was analyzed both by kinetics studies and molecular modeling, identifying critical interactions of 1 c with the zinc ion and residues in the active site. The effect of 1 c on intracellular Mycobacterium survival was assayed in J774 murine macrophages infected with M. tuberculosis H37Rv or M. bovis BCG and human monocyte‐derived macrophages infected with M. tuberculosis H37Rv. Cytotoxicity and genotoxicity were also assessed. Overall, inhibitor 1 c displays interesting in vitro antitubercular properties worthy of further investigation. 相似文献
