Physalis angulata L (Solanaceae) is a medicinal plant from North of Brazil, whose different extracts and infusions are commonly used in the popular medicine for the treatment of malaria, asthma, hepatitis, dermatitis and rheumatism. However, the genotoxic effects of P. angulata on human cells is not well known. The main purpose of the present study was to evaluate the in vitro genotoxic effects of aqueous extract of P. angulata using the comet assay and the micronucleus assay in human lymphocytes provided from 6 healthy donors. Treatments with P. angulata extracts were performed in vitro in order to access the extent of DNA damage. The comet assay has shown that treatments with P. angulata at 0.5, 1.0, 2.0, 3.0 and 6.0 microg/mL in culture medium were genotoxic. Lymphocytes treated with P. angulata at the concentrations of 3.0 and 6.0 microg/mL in culture medium showed a statistically significant increase in the frequency of micronucleus (p<0.05), however, the cytokinesis blocked proliferation index (CBPI) was not decreased after P. angulata treatment. In conclusion, the present work demonstrated the genotoxic effects of P. angulata extract on human lymphocytes in vitro.  相似文献

Activated calpain I immunoreactivity (76 kDa band) was detected in membranes prepared from rat brain hippocampal slices using a polyclonal antiserum raised against an N-terminus peptide of the cleaved subunit of calpain I. While basal levels of activated calpain I were stable over incubation times, 1 nM dopamine (DA) produced an initial 32% increase (5 min) in the 76 kDa protein followed by a 53% decrease in this band at 20 min of incubation. The DA-induced changes in activated calpain I immunoreactivity were blocked by the calpain inhibitor peptide, N-acetyl-Leu-Leu-norleucinal(100 microM) or by EGTA. Basal levels of the 76 kDa band were not affected by the calpain inhibitor. These changes in activated calpain I, elicited by DA, are in accord with the DA-induced decreases in the levels of the calpain substrate, gamma PKC (Yurko-Mauro and Friedman; J Cell Biochem [Abstr] 180:80, 1994; J Neurochem 65: 1622-1630, 1995) and suggest that DA activates this Ca(++)-dependent protease in its regulation of neuronal signal transduction.  相似文献

To define conditions promoting inherited infection by Lyme disease spirochetes in Ixodes ticks, we variously infected ticks with Borrelia afzelii and examined their progenies by dark-field microscopy, immunofluorescence, PCR, and serial passage. No episode of inherited infection was evident, regardless of instar or gender infected or frequency of exposure. We suggest that these spirochetes rarely, if ever, are inherited by vector ticks.  相似文献

Electrophoretic and chromatographic techniques were used to determine water‐soluble peptide profiles aiming to identify the adulteration of buffalo milk mozzarella cheese by the addition of cow's milk. Thus, cheeses were produced with contents of cow's milk varying from 0% to 100%, and the peptides extracted after production and after 20 days of refrigeration. Polyacrylamide gel electrophoresis under denaturing conditions in the presence of sodium dodecyl sulphate (SDS‐PAGE) identified a potential peptide marker of exclusively bovine origin with a size of about 21 kDa for the addition of cow's milk above 30%. Reverse‐phase high‐performance liquid chromatography (RP‐HPLC) indicated the existence of two potential peptides present in higher concentrations in buffalo milk and one exclusive for cow's milk, the latter making it possible to estimate the addition of cow's milk to buffalo milk. Six commercial brands of buffalo mozzarella cheese were evaluated, and indications of adulteration found in four of them.  相似文献

The aim of this study was to evaluate tubular dentin sealer penetration, comparing different final irrigation protocols using a conventional needle (CONV), EndoActivator system (EAS), EndoVac system (EVS), and ultrasound (PUI). Initially, fifty‐two first maxillary molars with a single canal in the palatal root, without abrupt curvatures, resorptive processes, or previous endodontic treatment were selected for this study. Then, the crowns were sectioned to obtain palatal roots 15 mm in length. The root canals were prepared with the ProTaper Universal System and irrigated with 5% NaOCl. Afterwards, the specimens were divided into four groups (n. 13), according to the final irrigation protocol: CONV, EAS, EVS, and PUI. After filling, slices at 3 mm and 5 mm from the apex were obtained for analysis by confocal laser scanning microscopy. Two‐way comparisons between the groups and the levels were performed with Games Howell's test (p < .05). Tubular dentin sealer penetration was higher at 5 mm compared with 3 mm from the apex (p < .05). The EAS group showed a higher percentage of tubular dentin sealer penetration, compared with the CONV group, at both levels. At 3 mm, there was no statistically significant difference among EAS, EVS, and PUI; however, these groups showed better performance, compared with the CONV group. At 5 mm, there was no statistically significant difference between the EAS and EVS groups, but both showed higher sealer penetration than the PUI group (p < .05). The EAS and EVS groups achieved better degrees of tubular dentin sealer penetration, compared with the other groups.  相似文献

In recent years, several papers on machining processes have focused on the use of artificial neural networks for modeling surface roughness. Even in such a specific niche of engineering literature, the papers differ considerably in terms of how they define network architectures and validate results, as well as in their training algorithms, error measures, and the like. Furthermore, a perusal of the individual papers leaves a researcher without a clear, sweeping view of what the field’s cutting edge is. Hence, this work reviews a number of these papers, providing a summary and analysis of the findings. Based on recommendations made by scholars of neurocomputing and statistics, the review includes a set of comparison criteria as well as assesses how the research findings were validated. This work also identifies trends in the literature and highlights their main differences. Ultimately, this work points to underexplored issues for future research and shows ways to improve how the results are validated.  相似文献

Systematic annealing at temperatures between 1300 °C and 1380 °C was applied to sheets of INCOLOY MA-956, an oxide-dispersion strengthened (ODS), mechanically alloyed, iron-base steel containing (in mass percent) 20.8Cr, 5.0A1, 0.5Y2O3, and 0.5Ti. The billets, comprised of hot isostatically pressed (“hipped”), mechanically alloyed powder, were hot- and cold-rolled to produce a 0.5-mm-thick sheet with a strong (100)«110» deformation texture. Light and transmission electron microscopy established that recrystallization initiated by nucleation at the sheet centerline. Initial rapid growth of the centerline-nucleated grains, designated stage I, resulted in plate-shaped grains oriented parallel to the rolling plane at the sheet centerline. Subsequent growth, designated stage II, was developed by planar growth fronts through the sheet thickness at a slower rate. The final product was a very coarse grain structure, sometimes with only a single grain through the sheet thickness. The recrystallization kinetics were typified by an incubation time, a temperature dependance characterized by an activation energy of 506 kJ/mole, and a decreasing rate of boundary migration with increasing time at temperature. The microstructural evolution is discussed in terms of the influences of deformation texture, residual stress, dislocation substructure, and oxide dispersion on the recrystallization process.  相似文献

The pulse radiolysis technique has been employed in the investigation of the dismutation of superoxide radicals, O?2 and HO2, in the presence of superoxide dismutase in aqueous solutions. The decay of superoxide radicals in the presence of the enzyme was found to be first order in both enzyme and superoxide concentrations. An apparent second order reaction rate constant was found to be about 2 × 109 M?1 sec?1, decreasing slightly as the pH is increased from 5 to 9.5. A mechanism which accounts for all our observations is proposed. It includes two steps: (1) formation of a product (EO?2 or E?) from one enzyme (E) molecule and one O?2 radical ion; (2) regeneration of E by a reaction of this product with an additional O?2 ion radical. The reaction rate constants k = (1.4 ± 0.2) × 109 and k = (1.9 ± 0.6) × 109 M?1 sec?1 were measured at pH = 7 in an oxygenated 0.16 M sodium formate solution.  相似文献
