Dendritic cells (DCs) can be divided by lineage into myeloid dendritic cells (mDCs) and plasmacytoid dendritic cells (pDCs). They both are present in mucosal tissues and regulate the immune response by secreting chemokines and cytokines. Inflammatory bowel diseases (IBDs) are characterized by a leaky intestinal barrier and the consequent translocation of bacterial lipopolysaccharide (LPS) to the basolateral side. This results in DCs activation, but the response of pDCs is still poorly characterized. In the present study, we compared mDCs and pDCs responses to LPS administration. We present a broad panel of DCs secreted factors, including cytokines, chemokines, and growth factors. Our recent studies demonstrated the anti-inflammatory effects of quercetin administration, but to date, there is no evidence about quercetin’s effects on pDCs. The results of the present study demonstrate that pDCs can respond to LPS and that quercetin exposure modulates soluble factors release through the same molecular pathway used by mDCs (Slpi, Hmox1, and AP-1).  相似文献

Among the rights in the Bill of Rights contained in the South African Constitution is the right to access adequate housing. This right is bound up with other rights in the Constitution, including the right to have human dignity respected, and the right to, inter alia water and health care. The right to adequate housing is also included in several international human rights declarations including the Universal Declaration of Human Rights by the United Nations. However, the right to access adequate housing is more than just shelter but includes a number of other elements such as security of tenure, and access to basic services and facilities. Housing must be affordable and accessible. It must be safe and habitable and be culturally acceptable. Moreover, adequate housing is well located with respect to economic and other opportunities. The provision of housing and basic services has been the focus of the ANC-led government since 1994, and a major portion of municipal resources has been dedicated to providing water and sanitation to particularly the poorest households, while over 4.3 million state-subsidized housing opportunities have been delivered. whereas the scale of delivery has been impressive, has it really impacted on the lives of people in remote communities? Research conducted in the Joe Morolong Municipality in the Northern Cape Province regarding the realization of the right to access adequate housing reveals that the right to adequate housing has at best, only been partially realized. In addition to interviews with office bearers and municipal officials, 120 households were interviewed (of which half were from self-built houses and half were residing in “RDP” houses) regarding their interpretation and experience of the right to access adequate housing in the municipality. The study reveals that the structures do not meet the criteria for adequate housing and the provision of water and sanitation remains a challenge. Access to health and education facilities is poor, due to the low densities and vast distances between settlements.  相似文献

More than 160 arginine analogues modified on the C-terminus via either an amide bond or a heterocyclic moiety (1,2,4-oxadiazole, 1,3,4-oxadiazole and 1,2,4-triazole) were prepared as potential inhibitors of NO synthases (NOS). A methodology involving formation of a thiocitrulline intermediate linked through its side-chain on a solid support followed by modification of its carboxylate group was developed. Finally, the side-chain thiourea group was either let unchanged, S-alkylated (Me, Et) or guanidinylated (Me, Et) to yield respectively after TFA treatment the corresponding thiocitrulline, S-Me/Et-isothiocitrulline and N-Me/Et-arginine substrate analogues. They all were tested against three recombinant NOS isoforms. Several compounds containing a S-Et- or a S-Me-Itc moiety and mainly belonging to both the dipeptide-like and 1,2,4-oxadiazole series were shown to inhibit nNOS and iNOS with IC50 in the 1–50 μM range. Spectral studies confirmed that these new compounds interacted at the heme active site. The more active compounds were found to inhibit intra-cellular iNOS expressed in RAW264.7 and INS-1 cells with similar efficiency than the reference compounds L-NIL and SEIT.  相似文献

For the first time, South Africa has a single national piece of legislation, the Spatial Planning and Land Use Management Act, 2013, that creates an overarching framework for spatial planning, policy and land use management for the entire country, including rural and informal settlements. Spatial plans linked to zoning schemes are at the heart of this planning system. However, zoning as a land use management tool has been intensely criticised as being exclusionary and socially, economically and environmentally unsustainable. Given that the purpose of the new legislation is to create equitable and sustainable development, this paper questions whether zoning is indeed the most suitable land use management tool in South Africa. The paper briefly outlines the requirements of the new act and then evaluates the usefulness of various forms of land use management such as land use zoning, performance zoning, form-based control and discretionary systems in the South African context. Drawing on the strengths of some of the alternative land use management systems, proposals are made for a more suitable land use management system for South Africa that could have applicability in other countries with a similar colonial history.  相似文献

The aim of the present study was to evaluate the effect of a prolonged administration of phytoestrogens on sheep growth rate, female reproductive response, lamb carcass conformation and meat quality. To investigate these effects, two groups of Comisana sheep (24 females and 20 males, initial average live weights of 25.0 and 29.9 kg, respectively) were fed on subterranean clover (SC, phytoestrogen content = 0.8 mg/g of DM) and Italian ryegrass (C, non-oestrogenic control diet) for about one year. Feedstuffs were offered ad libitum and supplemented with maize grain and sunflower meal to maintain an adequate and similar energy and protein intake. The results demonstrated that the prolonged administration of the selected subterranean clover cultivars, with low formononetin content (lower than 10% of total isoflavones on dry basis), did not affect ewe reproduction but induced a significant improvement in animal weigh gain and, in males, good carcass and meat characteristics.  相似文献

In Haiti, a novel approach to nutritional surveillance was developed on the basis of a low-cost, simple-to-repeat set of household surveys in all nine administrative departments using sentinel community sites. This system allows each department to independently conduct follow-up surveys as needed. The results of the first round of surveys conducted in 1995 show lower malnutrition rates in typical food-deficient departments and high levels of malnutrition in several food-surplus areas. Further analyses underscore the importance of variables related to child-care practices and of care-enabling factors such as household food security, health environment, and caregivers' time and education. These findings challenge the traditional thinking among a majority of Haitian policy makers who look at the malnutrition problem solely from the perspective of local food production.  相似文献

The perspective of downscaling organic electrochemical transistors (OECTs) in the nanorange is approached by depositing poly(3,4‐ethylenedioxythiophene) polystyrene sulfonate (PEDOT:PSS) on electrodes with a nanogap designed and fabricated by electromigration induced break junction (EIBJ) technique. The electrical response of the fabricated devices is obtained by acquiring transfer characteristics in order to clarify the specific main characteristics of OECTs with sub‐micrometer‐sized active channels (nanogap‐OECTs). On the basis of their electrical response to different scan times, the nanogap‐OECT shows a maximum transconductance unaffected upon changing scan times in the time window from 1 s to 100 µs, meaning that fast varying signals can be easily acquired with unchanged amplifying performance. Hence, the scaling down of the channel size to the nanometer scale leads to a geometrical paradigm that minimizes effects on device response due to the cationic diffusion into the polymeric channel. A comprehensive study of these features is carried out by an electrochemical impedance spectroscopy (EIS) study, complemented by a quantitative analysis made by equivalent circuits. The propagation of a redox front into the polymer bulk due to ionic diffusion also known as the “intercalation pseudocapacitance” is identified as a limiting factor for the transduction dynamics.  相似文献
