A vibrating reed magnetometer with high sensitivity is constructed. Using optical signal pickup, modified excitation coils, and an acoustical reference excitation system, the high sensitivity of former constructions is combined with the detection of the magnetization component perpendicular to the bias field H. The calibration procedure providing increased reliability is simplified and allows hysteresis measurements with the magnetic easy axis of the sample oriented at an arbitrary angle with respect to H. Cell suspensions of Escherichia coli as a model for bacterial populations in wastewaters were treated with ozone as a disinfectant in a continuously operated pilot plant with a plug flow reactor. Suspensions with an initial number of CFU (colony forming units) of 108 mL?1 were ozonized with ozone concentrations up to 12 mg/L. Metabolic activities and membrane functions break down with increasing ozone concentrations. The fluorochromization using 3′,6′-diacetylfluorescein (FDA) proved to be a suitable method for the detection of an alteration in permeability of the cell membranes and an inactivation of metabolic activity. By fluorescence microscopic and photometric investigations it could be clearly demonstrated that, in the case of disinfection with ozone, inactivation of the cells includes first of all a damage of the cell membranes. In contrast to the determination of the number of CFU, fluorochromization allows the detection of alteration in metabolic activities. 相似文献

The fracture assessment of structures subjected to secondary loading systems was studied by FEM-analysis of a few illustrative two-dimensional examples. The first example is an edge-cracked plate with a thermal gradient. The FEM-calculations revealed that a purely elastic analysis yielded J-values that are in most cases considerably more conservative than the ones calculated from an elastic-plastic analysis. The results were further compared with different engineering type non-linear estimates. As a second example a crack emanating from a semi-circular notch in a rectangular plate was chosen. The plate was subjected to uniaxial tension along its boundaries. It turned out that for the cases considered the J-values were fairly close to those elastically predicted. 相似文献

An inulin syrup made from Jerusalem artichoke tubers, either in its commercial form or after ultrafiltration, was freeze-dried and used as a prebiotic ingredient in the small-scale manufacture of wafer crackers. The flours used for the preparation of wafer batters were from wheat, rye or spelt wheat, or 1:1 combinations of wheat flour and rye flour or wheat flour and spelt wheat flour. Batter viscosity was strongly influenced by the selection of the flour type, but remained within technologically acceptable limits. The ultrafiltration of the inulin syrup, using a 1 kDa membrane, resulted in a significant reduction of the content of free sugars and minerals which, in turn, had a significant impact on the CIE-Lab color values of the wafer crackers. Using spelt wheat flour instead of wheat flour significantly increased wafer cracker firmness measured by penetration, as did the incorporation of ultrafiltered freeze-dried instead of native freeze-dried Jerusalem artichoke syrup. Sensory analysis revealed a significant influence of product formulation on appearance, flavor and texture of the wafer crackers. It can be concluded from quality scores, which were calculated by using weighting factors assigned to the sensory attributes, that wheat flour may be partially replaced by rye flour or spelt wheat flour without negatively affecting the sensory properties of the wafer crackers. 相似文献

Quantitative cellular in vitro nanoparticle uptake measurements are possible with a large number of different techniques, however, all have their respective restrictions. Here, we demonstrate the application of synchrotron-based X-ray fluorescence imaging (XFI) on prostate tumor cells, which have internalized differently functionalized gold nanoparticles. Total nanoparticle uptake on the order of a few hundred picograms could be conveniently observed with microsamples consisting of only a few hundreds of cells. A comparison with mass spectroscopy quantification is provided, experimental results are both supported and sensitivity limits of this XFI approach extrapolated by Monte-Carlo simulations, yielding a minimum detectable nanoparticle mass of just 5 pg. This study demonstrates the high sensitivity level of XFI, allowing non-destructive uptake measurements with very small microsamples within just seconds of irradiation time. 相似文献

The purpose of this investigation was to study the origin of the differences in paper‐to‐paper friction of linerboards based on old corrugated containers (OCC). The sheets were subjected to two extraction stages and analyzed with respect to, surface roughness, and their content of low‐molecular‐mass lipophilic compounds (LLC). Friction was measured using a friction tester based on the horizontal plane principle. The surface roughness was measured using a Perthometer profiler and the low molecular mass lipophilic constituent of the paper sheets was determined by gas chromatography‐mass spectroscopy. The sheets were imaged using environmental scanning electron microscopy (ESEM), and the relative compositions of inorganic ions on the paper surfaces were determined by energy dispersive X‐ray spectroscopy (EDS). The results showed that a high amount of LLC in the sheets lead to low friction, due to lubrication. It was also observed that large CaCO3 particles on the surface had a friction‐increasing effect, and that there was no relationship between the surface roughness and the friction. © 2002 Wiley Periodicals, Inc.
