Calibration samples for the Barkhausen noise (BN) method were produced with laser processing. A planet gear wheel used for production quality control was subjected to laser irradiation to verify the BN sensor output. Different samples were found to respond similarly to the laser processing although the laser parameters needed to be adjusted for different surface qualities separately. The surface optimization for laser processing was studied with different surface qualities of samples. The ground surface was compared with a sandblasted and vibratory ground surface. The ground and sandblasted surfaces were both amenable to the laser processing whereas the vibratory grinding process created inhomogeneous surfaces for laser beam absorption. Laser processing was found to produce uniform changes in the residual stress values in two perpendicular measuring directions. The root mean square value of the BN voltage signal exhibited linear correlations with the values of the residual stress and surface hardness.  相似文献

Residual stress of case-hardened steel samples is predicted in this paper with the linear multivariable regression model. The development of the prediction model is based on the huge set of features calculated from the Barkhausen noise measurement signal among which the most suitable ones are chosen. The selection uses a genetic algorithm with leave-multiple-out cross-validation in the objective function. The original feature set contains collinear features that make the selection task even more complex. Thus a feature elimination procedure based on the successive projections algorithm is studied in this paper. Also the standard genetic algorithm is slightly modified to better serve the feature selection task. The obtained results are good showing that the proposed procedures suit well for residual stress predictions. Also the applied feature elimination procedure is applicable and can be safely used to reduce the dimensionality of the selection problem.  相似文献

A cDNA encoding a new human matrix metalloproteinase (MMP) has been cloned from RNA prepared from odontoblastic cells. The open reading frame of the cloned cDNA codes for a polypeptide of 483 amino acids and is extensively similar to the sequence of recently described porcine enamelysin, suggesting that the isolated cDNA codes for the human homologue of this enzyme. Human enamelysin (MMP-20) has a domain organization similar to other MMPs, including a signal peptide, a prodomain with the conserved motif PRCGVPD involved in maintaining enzyme latency, a catalytic domain with a Zn-binding site, and a COOH-terminal fragment similar to the sequence of hemopexin. The calculated molecular mass of human enamelysin is about 54 kDa, which is similar to that of collagenases or stromelysins. However, this human MMP lacks a series of structural features distinctive of these subfamilies of MMPs. The full-length human enamelysin cDNA has been expressed in Escherichia coli, and the purified and refolded recombinant protein is able to degrade synthetic peptides used as substrates of MMPs, confirming that human enamelysin belongs to this family of proteases. Furthermore, the recombinant human enamelysin is able to degrade amelogenin, the major protein component of the enamel matrix. On the basis of its degrading activity on amelogenin, and its highly restricted expression to dental tissues, we suggest that human enamelysin plays a central role in the process of tooth enamel formation. Finally, we have found that the human enamelysin gene (MMP-20) maps to chromosome 11q22, clustered to at least seven other members of the MMP gene family.  相似文献

Residual stresses are quantitatively predicted based on the Barkhausen noise measurement with a partial least squares regression model. The measurements are taken from two sets of case-hardened samples. The benefits of using certain feature elimination strategies prior model identification are also studied. The elimination methods applied are correlation-based elimination, uninformative variable elimination and successive projections algorithm. The results show that the best predictions are usually obtained when the successive projections algorithm is applied. The prediction accuracy of the best models found shows that partial least squares models can be successfully used for prediction of material properties based on the Barkhausen noise measurement.  相似文献

Fibroblast-type interstitial collagenase (E.C. 3.4.24.7) was associated with loosening of total hip prostheses in eight patients: there were four cemented stems and one cementless stem with the common type of loosening and two cemented stems and one cementless acetabular component with aggressive granulomatous lesions. The authors used a specific, well-characterized, heterologous, affinity-purified, polyclonal rabbit anti-human fibroblast collagenase antiserum applied in avidin-biotin-peroxidase-complex (ABC) staining. In the aggressive granulomatous type of loosening, collagenase was found in most of the fibroblast- and macrophagelike cells, including multinuclear giant cells and epithelioid cells in periprosthetic tissue. Collagenase-positive cells also were found in the periprosthetic tissue associated with common loosening. Collagenase was also found in capillary and postcapillary venule endothelial cells in the richly vascularized aggressive granulomatous tissue. Collagenase was extracted directly from the tissue samples and incubated with soluble Type I collagen. Collagen degradation products then were analyzed by sodium dodecyl sulfate (SDS)-polyacrylamide gel electrophoresis, and the three-fourths length degradation product quantitated by gel scanning densitometry. In both aggressive granulomatosis and the common type of loosening, extractable collagenase was found in tissue. No significant differences between the sample groups were detected in respect to total measurable collagenase, however. The extractable collagenase was present in a latent form that could be activated by the organomercurial procollagenase activator, phenylmercuric chloride (PMC). It is likely that interstitial collagenase contributes to rapid growth of reactive infiltrative tissue, loosening of the prosthesis associated with aggressive granulomatosis, and the periprosthetic lytic process associated with the common type of hip prosthesis loosening.  相似文献

In this study, we compared the effects of several preservation methods on the secondary phenolics of the mature leaves of purple willow (Salix purpurea L., Salicaceae) with results obtained with fresh leaf analyses. Conventional freeze-drying, in which the leaves were first frozen with liquid nitrogen and then placed in a freeze-dryer, produced substantial qualitative and quantitative changes in purple willow flavonoids and salicylates. Modified freeze-drying, in which leaves were put into a freeze-dryer without being prefrozen, gave concentrations that, for most secondary components, were comparable with those found in fresh leaves. Reducing the freeze-dryer chamber temperature hindered the decomposition of phenolics in prefrozen leaves and in leaves dried without prefreezing. Heat drying induced substantial changes in the composition of all phenolics, except for apigenin-7-glucoside. Vacuum drying at room temperature gave the highest concentrations for nearly all phenolics, while room-drying with desiccation gave results that were comparable with those obtained by fresh leaf analyses.  相似文献

The production of gear components includes numerous manufacturing operations which are carried out to ensure proper surface characteristics of components to deal with wear and fatigue. Surface shot peening is one way to increase the compressive residual stresses on the surface and thus ensure better wear and fatigue resistance. An experimental plan for shot peening was conducted to produce samples with varying surface characteristics. Residual stress profile and Barkhausen noise measurements were carried out for the samples. The objective of the study was to evaluate the interactions between the shot peening parameters studied, the residual stress profiles and the Barkhausen noise measurements. A multivariable regression analysis was applied for the task. Some remarkable correlations were found between the shot peening parameters, residual stress profile and Barkhausen noise features. The most important finding was that when the shot peening intensity was high enough, over 0.5 mmA, it dominated the shot peening coverage density parameter and thus no correlations could be gained. On the other hand, if the intensity parameter was lower than the limit of 0.5 mmA, the correlation between residual stress and Barkhausen noise measurements was remarkable. This means that the surface Barkhausen noise measurements could be used for the evaluation of the stress gradient in the shot peening process.  相似文献

In aseptic loosening of initially well inserted total hip prostheses, implant wear debris and cyclic mechanical loading lead to a foreign body type of chronic inflammatory reaction, then to osteolysis, and finally to loosening of the implant. In the present work the reactive and adaptive changes of the periprosthetic tissues and pseudojoint were characterized by analysis of the local cell proliferation. Immunohistochemical demonstration of proliferating cells was performed by application of affinity purified rabbit antihuman Ki-67 antibodies to periprosthetic tissues obtained from revision operations for loose total hip prostheses. The fibrous areas and, in particular, the cell rich, vascular areas of the interface tissue (between implant and bone) and the pseudocapsule around aseptically loosened implants contained higher numbers of proliferating cells than the tissues around well fixed implants. In addition, the pseudosynovial lining occasionally contained some Ki-67 positive proliferating cells. Somewhat surprisingly, proliferating vascular endothelial cells were relatively rare. These findings suggest that reactive (interface tissues) and adaptive (pseudojoint and capsule formed around the artificial joint) tissue changes in loosening total hip prostheses comprise proliferation of local fibroblastlike cells. It is concluded that periprosthetic tissues of the loosened total hip prosthesis represent activated mesenchymal tissue.  相似文献

Aseptic loosening is the predominant cause of total hip implant failure. It has been assumed that a layer or membrane, containing macrophages, fibroblasts and vascular endothelial cells, of synovial-like tissue develops at the implant-to-bone interface almost invariably and, with time, somehow leads to loosening of the components from the surrounding bone. These cells produce a variety of cytokines and proteolytic enzymes which stimulate bone resorption. Platelet derived growth factor (PDGF) may be one of the cytokines which stimulate bone resorption and contribute to aseptic loosening in total hip replacement (THR). Synovial-like membrane from the implant or cement-to-bone interface (n = 10) and pseudocapsule (n = 10) were obtained from ten patients operated on for aseptic loosening of THR. As a control, nine samples of connective tissues were obtained from patients who had mandibular or maxillary fractures fixed with bone implant. The avidin-biotin-peroxidase complex (ABC) method with polyclonal rabbit anti-human IgG against the A-chain and B-chain of PDGF was used for staining. ABC-alkaline phosphatase-anti-alkaline-phosphatase double staining with monoclonal mouse anti-human fibroblast IgG1 and CD68 antibodies was used to ascertain the cellular origin of PDGF. Results of the PDGF staining were quantitated by a semi-automatic VIDAS image analysis system. The PDGF-A and PDGF-B chain containing cells were found in all periprosthetic tissues, in particular in macrophages with phagocytosed particulate debris, but to some extent also in fibroblasts and in endothelial cells. The numbers of PDGF-A and PDGF-B chain positive cells per mm 2 in synovial-like interface membrane (1881 +/- 486 and 1877 +/- 214) and pseudocapsule (1786 +/- 236 and 1676 +/- 152) were higher (P < 0.01) around loose THR than in control tissue (821 +/- 112 and 467 +/- 150), respectively. The results of the present study suggest that PDGF is preferably expressed by macrophages, which to an increased extent produce it in the synovial-like interface membrane and pseudocapsular synovial-like membrane. Because of its role in bone resorption, it may well play a role in periprosthetic bone loss and aseptic loosening and deserves more detailed study as a mediator and potential target in the modulation or prevention of loosening of THR.  相似文献
