Effects of gastrointestinal digestion models in vitro on phenolic compounds and antioxidant activity of juçara (Euterpe edulis)

This study evaluated the effects of different gastrointestinal digestion models in vitro on the bioaccessibility of phenolics and antioxidant activity in juçara frozen pulp. In the sequence, method 3 was applied to juçara fruit in three different stages of maturation (vitrin – reddish fruits, mature – purple fruits, tuíra – deep purple fruits). In the method applied, the final pH adopted was 5.0, in order to avoid interference in the assay used to determine the antioxidant activity, and BHT was used to prevent excessive oxidation in the system. In this method, higher values for antioxidant activity were obtained (3574.95–3719.10 μmol L^?1 Trolox 100 g^?1 pulp) compared with the other two methods tested (1969.14–3034.74 μmol L^?1 Trolox 100 g^?1 pulp). In relation to juçara fruit, the mature stage was found to be ideal for processing, showing generally higher values of the bioaccessibility for phenolics and antioxidant activity compared to other maturation stages.     

Radar‐based structural health monitoring of wind turbine blades: The case of damage localization

This short communication reports on a radar approach for structural health monitoring of wind turbine blades. Therefore, a bistatic frequency‐modulated continuous wave (FMCW) radar in the frequency range from 33.4 to 36.0 GHz has been developed and tested experimentally using a laboratory wind turbine demonstrator. A differential damage localization framework is presented here that exploits signal differences between measurements from the intact and the damaged structure for 3D imaging of the defect. We have achieved the localization of a 30‐mm cut in a glass fiber composite structure as well as the localization of a water pack at the backside of the specimen with a localization error of several centimeters.     

Sex difference in polybrominated diphenyl ether concentrations of walleyes

Polybrominated diphenyl ether (PBDE) concentrations were determined for mature male and mature female walleyes (Sander vitreus) sampled from the Saginaw Bay population during 2007. PBDE concentrations in prey fish caught in the Saginaw River, the primary tributary to Saginaw Bay, and in Saginaw Bay during 2005 and 2007 also were determined. Mature male and mature female walleyes averaged 70.3 ng/g and 24.8 ng/g, respectively, in ΣPBDE, which was equal to the sum of concentrations of six PBDE congeners (BDE-28, BDE-47, BDE-99, BDE-100, BDE-153, and BDE-154). This sex difference was likely due to males spending more time in the Saginaw River system than females. Prey fish captured in the Saginaw River were roughly ten times higher in ΣPBDE than those caught in Saginaw Bay. BDE-47 was the predominant congener in both walleyes and prey fish, and this congener contributed about 50%, on average, to ΣPBDE. Congener profiles differed significantly between the two sexes of walleyes. In contrast, congener profiles of the prey fish did not differ significantly between the river-caught fish and the bay-caught fish. One plausible explanation for these congener profile results was that net trophic transfer efficiencies of PBDEs to walleyes from their prey were similar for all congeners except BDE-28, and that diet composition differed between the two sexes of walleyes.     

PCB concentrations in walleyes and their prey from the Saginaw River,Lake Huron: A comparison between 1990 and 2007

Concentrations of polychlorinated biphenyls (PCBs) were measured in composited samples of walleyes Sander vitreus and their prey during 2005–2007 from Saginaw Bay, Lake Huron. There was a linear relationship between fish length and PCB concentrations in walleyes between 356 and 608 mm, but fish 680 mm had lesser concentrations than 608-mm fish. When fish 222–550 mm from 1990 were compared with those from 2007, there was a decrease of 1315 ng PCBs/g wet wt (ww). Concentrations of PCBs in gizzard shad Dorosoma cepedianum (190 ng PCBs/g ww) were three-fold less than fish collected in 1990 (516 ng PCBs/g ww). Round gobies Neogobius melanostomus collected from the Saginaw River had the greatest concentrations of PCBs (range: 200–350 ng PCBs/g ww) compared with other prey fishes (45–190 ng PCBs/g ww). Concentrations of PCBs in Saginaw River round gobies were three-fold greater than those from Saginaw Bay. Zooplankton from 1990 and 2008 contained 8.0 and 32 ng PCBs/g ww, respectively, while zebra mussels Dreissena polymorpha from 2008 contained 351 ng PCBs/g ww. Principal components analysis showed that PCB congeners differed between the largest walleyes and other fish. There are several possible explanations for lesser concentrations of PCBs observed in 2007. These include effects of dredging, changes in the food web related to round gobies, loss of alewife Alosa pseudoharengus, which was a major walleye prey item in 1990, and replacement by yellow perch Perca flavescens, or decreases in release of PCBs from sediments due to weathering, burial, or diffusion.     

Outside-in signaling in the chondrocyte. Nitric oxide disrupts fibronectin-induced assembly of a subplasmalemmal actin/rho A/focal adhesion kinase signaling complex

Elevated levels of fibronectin (Fn) in articular cartilage have been linked to the progression of both rheumatoid and osteoarthritis. In this study, we examined intracellular events which follow ligation of Fn to its receptor, the integrin alpha5beta1. In addition, we examined the regulatory influence of nitric oxide on these events, since this free radical has been implicated in cartilage degradation. Exposure of chondrocytes to Fn-coated beads resulted in the circumferential clustering of the alpha5beta1 integrin receptor, which was accompanied by the subplasmalemmal assembly of a focal activation complex comprised of F-actin, the tyrosine kinase, focal adhesion kinase (FAK), the ras related G protein rho A, as well as tyrosine-phosphorylated proteins. Treatment with exogenous nitric oxide (NO) or catabolic cytokines which induce nitric oxide synthase blocked the assembly of F-actin, FAK, rho A and tyrosine-phosphorylated proteins while not affecting the total number of beads bound per cell nor the clustering of alpha5beta1 integrin. Use of a cGMP antagonist (Rp-8-Br cGMPS) or cGMP agonist (Sp-cGMPS) either abolished or mimicked the NO effect, respectively. Adherence of chondrocytes to fibronectin enhanced proteoglycan synthesis by twofold (vs. albumin). In addition, basic fibroblast growth factor (FGF) and insulin growth factor (IGF-1) induced proteoglycan synthesis in chondrocytes adherent to Fn but not albumin suggesting a costimulatory signal transduced by alpha5betal and the FGF receptor. Both constitutive and FGF stimulated proteoglycan synthesis were completely inhibited by nitric oxide. These data indicate that the ligation of alpha5beta1 in the chondrocyte induced the intracellular assembly of an activation complex comprised of the cytoplasmic tail of alpha5beta1 integrin, actin, and the signaling molecules rho A and FAK. We show that NO inhibits the assembly of the intracellular activation complex and the synthesis of proteoglycans, but has no effect on the extracellular aggregation of alpha5beta1 integrin. These observations provide a basis by which nitric oxide can interfere with chondrocyte functions by affecting chondrocyte-matrix interactions.     

Gender difference in walleye PCB concentrations persists following remedial dredging

Eleven male walleyes (Sander vitreus) and 10 female walleyes from the Saginaw Bay (Lake Huron) population were caught during the spawning run at Dow Dam (Midland, Michigan) in the Tittabawassee River during April 1996, and individual whole-fish polychlorinated biphenyl (PCB) determinations were made. Total PCB concentrations averaged 7.95 and 3.17 mg/kg for males and females, respectively. As part of the Natural Resource Damage Assessment remediation process, contaminated sediments from the Saginaw River, the main tributary to Saginaw Bay, were removed during 2000 and 2001. Total PCB concentrations of 10 male and 10 female walleyes caught at Dow Dam during April 2007 averaged 1.58 and 0.55 mg/kg, respectively. Thus, dredging of the Saginaw River appeared to be effective in reducing PCB concentrations of Saginaw Bay adult walleyes, as both males and females decreased in PCB concentration by more than 80% between 1996 and 2007. However, the ratio of male PCB concentration to female PCB concentration did not decline between 1996 and 2007. This persistent gender difference in PCB concentrations was apparently due to a gender difference in habitat utilization coupled with a persistent spatial gradient in prey fish PCB concentrations from the Saginaw River to Lake Huron.     

Effect of compressed fluids treatment on the activity,stability and enzymatic reaction performance of β-galactosidase

The present work investigates, as a first step, the influences of pressure, exposure time and depressurisation rate on the β-galactosidase activity of permeabilized cells of Kluyveromyces marxianus CCT 7082 submitted to treatment with compressed carbon dioxide, propane and n-butane. In general, results showed that the enzyme activity changes significantly, depending on the experimental conditions investigated, allowing the selection of proper operating conditions for advantageous application of this biocatalyst in hydrolysis reactions. Further, the stability of the enzyme, after high-pressure pre-treatment, was also experimentally monitored, and the results demonstrated that the activities of this biocatalyst were always higher than those of the non-treated one. Finally, evaluation of synthesis of galactooligosaccharides (GOS), using β-galactosidase without treatment and after treatment in compressed fluids, was carried out. Results indicated promising possibilities since the use of n-butane-pre-treated enzyme led to very satisfactory reaction conversions and selectivity.
