In the present study a synthetic glycolipid system is presented that can be readily incorporated into phospholipid micelles and that allows the study of cell-surface-exposed carbohydrate units by high-resolution NMR techniques. Here, we present an efficient route for the synthesis of glycolipid compounds that contain mannose, mannobiose, or mannotriose coupled either directly to an alkyl chain or through a poly(ethylene glycol) linker. Furthermore, we have validated our model system by measuring the binding of cyanovirin N (CV-N), a cyanobacterial protein that binds with nanomolar affinity to the terminal arms of high-mannose structures of the HIV surface-envelope glycoprotein gp120, to glycolipids the carbohydrate portions of which comprise the corresponding high-mannose moieties. From the results of chemical-shift mapping with uniformly (15)N-labelled CV-N, we conclude that binding to the protein occurs at sites similar to those involved in binding the nonconjugated carbohydrates. We characterized the insertion of the glycolipids into dodecylphosphocholine (DPC) micelles by measuring translational diffusion, and we observed that the diffusion constants of the glycolipids were very similar to those of the DPC micelles themselves, but significantly deviated from those of the free glycolipids. We also present experimental proof that the glycolipids remain inserted in the micelles while binding to CV-N. Finally, by addition of a ligand that had a higher affinity to CV-N but which was not attached did not couple to a lipid anchor, CV-N could be released from the glycolipid and, hence, from the micelle-associated state.  相似文献

The efficiency of a biologically activated carbon system for treating wastewater polluted with petroleum products was examined and the effects of process parameters on its efficacy were evaluated. In each experiment 17 alkylated and 19 non-alkylated polycyclic aromatic hydrocarbons (PAHs) and total petroleum hydrocarbons (TPHs, C10–C40) were extracted using semipermeable membrane devices from wastewater before and after treatment. The acquired data during experiments were analyzed using principal component analysis (PCA). The treatment system robustly removed dissolved PAHs across the studied ranges of the process parameters, providing overall removal efficiencies of 96.9–99.7% for the sum of 36 PAHs. However, the major contributor to their removal was sorption rather than biodegradation, and despite the general efficiency of the process there was up to a 9-fold range in the sums of quantified PAHs in the effluents between experiments. Combinations of long process contact time (24 h) with high temperature (24 °C) and moderate oxygen concentration (6–7 mg O2 L−1) resulted in good removal of bioavailable PAHs. The removal of TPHs was more dependent on biological activities during the wastewater treatment, and consequently more dependent on the process parameters. In addition, small but significant proportions of PAHs were volatilized and released during the wastewater treatment.  相似文献

The concept of sustainable energy development cannot be separated from the understanding of additional positive socio-economic effect of sustainable energy projects (energy efficiency measures, use of renewable energy sources). Implementation of sustainable energy projects has positive impact on security of energy supply, provides financial economies and improved comfort and has multiplier effect for new jobs, involving small and medium size enterprises. Therefore integration of sustainable energy projects into regional development process may create external positive effect concerning increased energy security and other regional development goals (reduction of unemployment, reduction of environmental impact, etc.). The examples of EU-15 countries implementing their sustainable energy development and climate change mitigation policies on local level can be successfully applied in Lithuania. The aim of the article is to discuss methodological problems related with integration of sustainable energy projects into regional development procedures and provide guidelines, ensuring that energy elements may compile integral uniformity in terms of regional goals.  相似文献

Deoxynivalenol (DON) together with two acetylated derivatives, 3-acetyldeoxynivalenol (3-ADON) and 15-acetyldeoxynivalenol (15-ADON) occurs in cereal grains and their products. Co-occurrence of DON and acetylated derivatives in cereal grain is detected worldwide. Until now, DON and its derivatives have been considered equally toxic by health authorities. In this study, we analysed 103 samples of spring wheat grain, originating from the fields of different production systems in Lithuania, for the co-occurrence of type-B trichothecenes (DON, 3-ADON, 15-ADON). The samples were classified according to the production system—organic, sustainable and intensive. Mycotoxin levels in the spring wheat grain samples were determined by the HPLC method with UV detection. The type-B trichothecenes were found to be present at higher concentrations in the grain from the intensive production system. Eighty-one percent of the spring wheat grain samples from the intensive production system were co-contaminated with a combination of DON+3-ADON+15-ADON, 1% with DON+3-ADON. Additionally, DON+15-ADON and DON were found in 5% and 10% of the tested samples, respectively. Two percent of the samples were free from mycotoxins. In the grain samples from the sustainable production system, DON and a combination of DON+3-ADON showed a higher incidence – 47% and 23%, respectively. The samples with a combination of DON+3-ADON+15-ADON accounted for 18%. Completely different results were obtained from the analyses of organic grain samples. A large number of the organic spring wheat grain samples were contaminated with DON+3-ADON (55%) or DON (36%). The combination of DON+3-ADON+15-ADON was not present, while DON+15-ADON was present in 9% of the samples tested. The production systems did not lead to significant differences in mycotoxin levels, although a trend toward higher incidence and higher contamination was observed for the samples from the intensive and sustainable production systems.  相似文献

Nowadays, there is a growing interest in natural, minimally processed, nutritional and healthy foods. Sprouted seeds can be offered as natural nutritive products. Regrettably, existing seed decontamination technologies are limited and have specific disadvantages. 5-aminolevulinic acid (5-ALA) as a novel and effective tool for wheat decontamination from microfungi is proposed in this work. Inhibition of wheat with 5-ALA revealed a drastically suppressed development of microfungi. Studies of wheat germination characteristics showed that 5-ALA stimulates the growth of wheat seedlings and roots without impairing the vigor of germination and the viability of seeds. 5-ALA also induces either marginal or significant activities of antioxidant enzymes which can be associated with enhanced cellular capacity to detoxify reactive oxygen species. The results indicate that 5-ALA application may be an effective, environmentally friendly and inexpensive technology to be used in producing sprouts for human consumption.  相似文献
