ABSTRACT: Carotenoids are C-40 tetraterpenoid compounds with potential health beneficial effects. Major dietary sources include a variety of fruits and vegetables. Rapid screening methods are therefore desired, but their accuracy varies depending on the carotenoid profile and the matrix of the plant food. In the present study, 3 different methods were compared, all based on a rapid extraction protocol and spectrophotometric measurements to determine the total amount carotenoids present in fruits and vegetables (n = 28), either with or without chlorophyll. Published methods (a) Lichtenthaler and (b) Hornero-Méndez and Mínguez-Mosquera were compared with a newly developed method (method c) based on the average molar absorption coefficient (135310 Lcm−1mol−1) and wavelength (450 nm in acetone), for the 5 predominant carotenoid species (beta-carotene, zeaxanthin, lycopene, lutein, beta-cryptoxanthin) in the investigated foods. All results were compared to HPLC (method d). To avoid overestimating carotenoid concentrations due to chlorophyll A and B presence, the effect of saponification was studied for all methods. Overall, saponification led to significant carotenoid losses (12.6 ± 0.9%). Methods a, b, c, and d yielded 5.1 ± 0.4 mg/100 g, 4.6 ± 0.5 mg/100 g, 4.3 ± 0.5 mg/100 g, and 4.2 ± 0.5 mg/100 g total carotenoids, respectively, with method a leading to significant higher mean concentrations compared to all other methods (P < 0.001, Bonferroni) with methods b and c being not significantly different and highly correlated compared to HPLC (> r = 0.95). Similar results were found when stratifying for chlorophyll content and fruits compared with vegetables, however, accuracy varied for individual fruits, highlighting the limitation to use the same method for all plant foods. Practical Application: This study presents a comparison of various rapid spectrophotometric measurements to determine total carotenoid content in various fruits and vegetables and could aid in the selection of the appropriate method for individual plant foods with different carotenoid profile and matrices.  相似文献

Several dietary and host related factors potentially influencing carotenoid (beta-carotene, lutein and zeaxanthin) bioaccessibility from spinach, including different concentrations of sodium, calcium and magnesium, were systematically investigated by means of an in vitro digestion model. Bioaccessibility was highest when milk (4% fat) and lowest when skimmed milk or more complex food matrices such as sausage were added to the meal. Micellarisation significantly depended on the presence and concentration of bile salts and pancreatin (p < 0.001, Bonferroni) but was unaffected by pepsin. Micellarisation significantly decreased to 61.4 ± 3.0% of control (p < 0.001, Dunnett’s) at high cholesterol (114 mg/test meal) but not at similar stigmasterol concentrations. Calcium and magnesium ?13.8 mM individually inhibited micelle formation ( >40% on average), presumably due to the generation of insoluble soaps with fatty acids and bile salts. Increased sodium concentrations (280 and 460 mM) altered carotenoid micellarisation patterns, favoring beta-carotene isomers (p < 0.001, Bonferroni) but decreasing lutein and zeaxanthin (p < 0.001 and p < 0.05, respectively, Bonferroni). This study suggests that minerals may impact carotenoid bioavailability.  相似文献

Austenitic stainless steels are widely used in medical and food industries because of their excellent corrosion resistance. However, they suffer from weak wear resistance due to their low hardness. To improve this, plasma nitriding processes have been successfully applied to austenitic stainless steels, thereby forming a thin and very hard diffusion layer, the so‐called S‐phase. In the present study, the austenitic stainless steels AISI 304L and AISI 316L with different microstructures and surface modifications were used to examine the influence of the steel microstructure on the plasma nitriding behavior and corrosion properties. In a first step, solution annealed steel plates were cold‐rolled with 38% deformation degree. Then, the samples were prepared with three kinds of mechanical surface treatments. The specimens were plasma nitrided for 360 min in a H2–N2 atmosphere at 420 °C. X‐ray diffraction measurements confirmed the presence of the S‐phase at the sample surface, austenite and body centered cubic (bcc)‐iron. The specimens were comprehensively characterized by means of optical microscopy, scanning electron microscopy, glow discharge optical emission spectroscopy, X‐ray diffraction, surface roughness and nano‐indentation measurements to provide the formulation of dependencies between microstructure and nitriding behavior. The corrosion behavior was examined by potentio‐dynamic polarization measurements in 0.05 M and 0.5 M sulfuric acid and by salt spray testing.  相似文献

Dendritic flux penetration is a well known phenomenon in high temperature superconductors. We investigated the conditions under which this instability can be observed in different materials. We found three qualitatively different behaviors: dendrite development above a “critical field” and up to a certain temperature, above a “critical field” with no measurable temperature dependence, or no dendrite development at all.  相似文献

Recent efforts to increase public awareness of child abuse may result in an increased number of inappropriate reports of suspected child abuse. The authors believe that digital hair strangulation should be included among the conditions that may be confused with child abuse. Digital hair strangulation (toe tourniquet syndrome) occurs primarily in infants and is characterized by a constricting band of foreign material that becomes tightly wrapped around a digit or digits (most often the toes). The consensus in the medical literature is that this condition is not the result of intentional injury. As no reference to the toe tourniquet syndrome exists in the child abuse literature, it was hypothesized that child welfare workers would be more likely than physicians and public health nurses to misinterpret this condition as resulting from intentional injury. A survey was conducted to test this hypothesis. Professionals from the fields of medicine, nursing, and child welfare were provided with a history and photographic findings of a child with a typical case of the toe tourniquet syndrome. Participants were surveyed regarding their interpretation of the described injuries. More than 50% of all respondents indicated that they would report this case as suspected abuse. Child welfare workers responded that the injuries were suggestive of abuse (83%), significantly more often than public health nurses did (45%), (chi 2 = 4.55, p = .03).(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献
