ABSTRACT: To help define the composition of commercial teas consumed in the United States, we have developed and validated an high-performance liquid chromatography (HPLC) method to analyze levels of catechins, theaflavins, and alkaloids in 77 commercial black, green, specialty (brown rice, white, oolong), and herbal teas extracted with hot water to simulate home use. The following 13 compounds were separated in a single analysis on an Inertsil ODS-3v column with acetonitrile/potassium dihydrogen phosphate as the mobile phase: (−)-epigallocatechin, (+)-catechin, (−)-epicatechin, (−)-epigallocatechin-3-gallate, (−)-gallocatechin-3-gallate, (−)-epicatechin-3-gallate, (−)-catechin-3-gallate, theaflavin, theaflavin-3-gallate, theaflavin-3'-gallate, theaflavin-3, 3'-digallate, caffeine, and theobromine. The data show that (1) extraction time from 3 min to 20 min did not significantly affect measured levels of tea compounds and (2) there was a wide variation in the composition of the tea compounds both within each tea category and among categories: black teas contained both theaflavins and catechins, green and white teas contained catechins and trace amounts of theaflavins, herbal teas contained very low amounts of all tea compounds. A statistical profile of the distribution of catechins, theaflavins, caffeine, and theobromine in the evaluated teas offers consumers a choice of teas containing high levels of health-promoting compounds.  相似文献

The VPS13 family of proteins have emerged as key players in intracellular lipid transport and human health. Humans have four different VPS13 orthologs, the dysfunction of which leads to different diseases. Yeast has a single VPS13 gene, which encodes a protein that localizes to multiple different membrane contact sites. The yeast vps13Δ mutant is pleiotropic, exhibiting defects in sporulation, protein trafficking, endoplasmic reticulum (ER)-phagy and mitochondrial function. Non-null alleles resulting from missense mutations can be useful reagents for understanding the multiple functions of a gene. The exceptionally large size of Vps13 makes the identification of key residues challenging. As a means to identify critical residues in yeast Vps13, amino acid substitution mutations from VPS13A, B, C and D, associated with human disease, were introduced at the cognate positions of yeast VPS13, some of which created separation-of-function alleles. Phenotypic analyses of these mutants have revealed that the promotion of ER-phagy is a fourth, genetically separable role of VPS13 and provide evidence that co-adaptors at the endosome mediate the activity of VPS13 in vacuolar sorting.  相似文献

We study the output-sensitive computation of all the combinations of edges and concave vertices of a simple polygon that allow an immobilizing grasp with less than four frictionless point contacts. More specifically, if n is the number of edges, and m is the number of concave vertices of the polygon, we show how to compute: in O(m4/3 log1/3 m + K) time, all K combinations that allow a form-closure grasp with two contacts; in O(n2 log4 m + K) time, all K combinations that allow a form-closure grasp with three contacts; in O(n log4 m + (nm)2/3 log2+ε m + K) time (for any constant ε > 0), all K combinations of one concave vertex and one edge that allow a grasp with one contact on the vertex and one contact on the interior of the edge, satisfying Czyzowicz's weaker conditions for immobilization; in O(n2 log3 n + K) time, all K combinations of three edges that allow a grasp with one contact on the interior of each edge, satisfying Czyzowicz's weaker conditions for immobilization.  相似文献
