Primary biliary cirrhosis (PBC) is an immunologically mediated disease in which activated T lymphocytes attack and destroy epithelial cells in the small intralobular bile ducts of genetically susceptible patients. This article reviews the results of treatment of PBC with immunomodulatory agents. Results with drugs such as glucocorticoids, azathioprine, and chlorambucil have been disappointing because of either limited efficacy (azathioprine), toxicity (chlorambucil), or both (glucocorticoids). Colchicine improved tests of liver function in three prospective studies and was associated with improved survival for up to 4 years. However, survival benefits were lost at 8 years. Colchicine appears to slow the rate of progression of PBC but not to stop it. Preliminary results suggest that colchicine may have synergistic effects if used together with ursodeoxycholic acid, particularly in patients who are only partially responsive to ursodeoxycholic acid. Results with cyclosporine have been disappointing because of limited efficacy and predictable toxicity. The modest improvement in tests of liver function and survival are counterbalanced by the development of hypertension in some and worsening renal function in most. There is little beneficial effect on symptoms or histology. Results with methotrexate are promising. There are improvements in symptoms and tests of liver function that are equal to those seen with ursodeoxycholic acid and significant improvement in liver histology. Some patients, particularly those with striking inflammation and granulomas in portal triads, appear to have achieved sustained remission while on methotrexate. The effects of methotrexate are additive to those of ursodeoxycholic acid in patients whose blood tests have responded only partially to ursodeoxycholic acid. The most effective treatment of PBC will most likely use a combination of drugs such as ursodeoxycholic acid, colchicine, and methotrexate.  相似文献

We have recently developed an optical contactless method for testing the quality of solder joints during accelerated thermal cycling ageing processes.1 The method was based upon the measurement of the dynamic thermal behaviour of the joint to short bursts of Joule heating. It has proved to be efficient in revealing the formation of cracks at the lead-solder interface. We present a method to evaluate ageing at a much earlier stage in the cycling process. We have observed in earlier work,1 that before cracks appear, structural changes occur in the solder at the lead-solder interface. The thermal response of the solder joint is recorded over time to a Peltier heat perturbation produced by flowing a current pulse through the interface where structural changes occur. The key point in this method is to discriminate the Peltier effect from the Joule thermal response because both effects generate heat. The variation of the early Peltier response in the thermal cycling ageing tests is seen as a quantitative signature of the structural changes in the lead-solder interface.  相似文献

Results are presented of a detailed study of the effects of high-temperature 4-MeV neutron irradiation on the performance degradation of Si pin photodiodes together with the radiation-induced defects, observed by deep level transient spectroscopy (DLTS). It was found that the dark current increases after irradiation, while the photocurrent decreases. After irradiation, two majority electron capture levels with (E c–0.22 eV) and (E c–0.40 eV) were induced in the n-Si substrate, while one minority hole capture level with (E v+0.37 eV) was found. Additionally, the degradation of the device performance and the introduction rate of the lattice defects decreases with increasing irradiation temperature. For a 250 °C irradiation, the reduction of the reverse current is only 20% of the starting value. This result suggests that the creation and recovery of the radiation damage proceeds simultaneously at high temperatures.  相似文献

The purpose of this study was to examine individual differences in the effectiveness of learning objects in secondary school classrooms. Specifically, gender, age, grade, subject area, and computer comfort (self-efficacy) were examined in 850 students. Effectiveness was measured in terms of student attitude (learning, quality, and engagement) and student performance. No gender differences were observed between males and females with respect to student attitudes or performance. Age was significantly correlated with student attitudes and performance, however correlation coefficients were small. Grade 12 students were more positive about learning objects and performed better than grade 9 and 10 students. Science students had significantly more positive attitudes and performed better than mathematics students. Finally, students who were more comfortable about computers, appreciated learning objects more than their less confident peers, however performance was unaffected.  相似文献

Carcass properties and meat quality characteristics of 32 Belgian Blue White double-muscled bulls (DM) were compared with those of 59 bulls of the same breed with normal conformation (N). DM showed superior carcass quality as revealed in increased dressing percentage, meat production yield, conformation grade, muscle/fat and muscle/bone ratios (all P < 0·001). Longissimus dorsi (LD) shear force values, drip and cooking losses at 8 days post mortem (pm) were significantly higher (P < 0·001) for DM, whereas sarcomere lengths were not different. Calpain 1 and calpastatin levels at 1 h and 24 h pm were tremendously decreased in DM as were also cathepsin B and L levels at 1 and 8 days pm (P < 0·001). As evident from semi-quantitative SDS-PAGE, these differences were accompanied by higher titin and lower 30 kDa levels (P < 0·001) in DM. Troponin-t levels were not different, but very low. Intramuscular collagen content was significantly lower in DM (P < 0·001). This suggests that lower background toughness in DM was compensated for by reduced post-mortem proteolytic tenderization. Discrepancy with literature reports regarding tenderness of DM might be related to the extreme muscularity of the Belgian Blue White breed, compared to other DM breeds. The results also suggest that reduced protein turnover might be involved in the muscle hypertrophy phenomenon within this breed, because of likely reduced levels of calpains and cathepsins in living DM animals.  相似文献

The increasing costs associated with water supply and the disposal of wastewater has stimulated industries to seek more efficient water management systems. Mathematical modelling and simulation can be a very valuable tool for the study of the multiple alternatives available whilst assessing optimum solutions for water management in industry. This study introduces a new steady state model library able to reproduce industrial water circuits. It has been implemented in a novel software framework for the representation, simulation and optimization of industrial water networks. A water circuit representing a paper mill has been modelled and simulated showing the capability to reproduce real case studies. Alternative scenarios for the water network have also been tested to assess the capability of the models to optimize water circuits minimizing total cost.  相似文献

Lecontite, (NH4)Na(SO4).2H2O, was synthesised at room temperature in high purity compared to earlier work with a minor impurity of mascagnite, (NH4)2SO4. Rietveld refinement of the XRD results confirmed the crystal structure and unit cell dimensions as published earlier. Raman and Infrared spectroscopy, in conjunction with factor group analysis, resulted in a complex pattern of overlapping sulphate, NH and OH modes. The NH modes υ1 was observed around 2880 cm−1, υ2 around 1700 cm−1 overlapping with water OH-bending modes, υ3 around 3300 cm−1 overlapping with water OH-stretching modes around 3023, 3185 and 3422 cm−1, and υ4 around 1432, 1447 and 1462 cm−1. The sulphate group in the crystal structure displays a decrease in symmetry from Td as evidenced by the activation of the ν1 mode at 982 cm−1 and the ν2 mode around 452 cm−1 in the Infrared spectrum. The υ3 mode shows clear splitting in the infrared spectra with a strong band at 1064 cm−1 accompanied by two shoulders at 1107 and 1139 cm−1. The Raman spectra show three weak bands at 1068, 1109 and 1135 cm−1 with a shoulder at 1155 cm−1. Similar splitting was observed for the υ4 mode around 611 and 632 cm−1 in the Infrared and Raman spectra, respectively.  相似文献

The impact of different processing factors on the low-frequency (LF) noise of nMOSFETs fabricated in strained-silicon (SSi) substrates will be described. It is shown that the use of an SSi substrate can yield improved LF noise performance compared with standard Czochralski silicon material. This is demonstrated for both full-wafer and selective epitaxial SSi material. The lower 1/f noise points to an improved gate oxide quality, i.e., with a lower interface and bulk defect density, and is correlated with the low-field mobility or transconductance of the transistors. At the same time, it will be demonstrated that there exist defect-related LF noise mechanisms, which generally give rise to excess generation-recombination (GR) noise. Associated with this GR noise, a degradation of either the OFF-state leakage current or the mobility (transconductance) of the devices is observed. It is clear that noise is a sensitive parameter to local defectiveness and may be a useful tool for both materials' characterization and the analysis of processing-related device degradation mechanisms.  相似文献
