The microbiological and physicochemical changes of industrially fermented Halkidiki and Conservolea green table olives were determined. Samples were analysed to monitor the population of lactic acid bacteria (LAB), yeasts and Enterobacteriaceae, together with changes in pH, acidity, salinity, colour, lactic acid, acetic acid and ethanol. LAB and yeast species diversity was evaluated at the beginning (1 day), middle (75 days) and final (135 days) stages of fermentation by RAPD-PCR genomic fingerprinting. Results revealed vigorous lactic acid processes as indicated by the dominance of LAB over yeasts. No Enterobacteriaceae could be detected after 30 days. Lactiplantibacillus plantarum (formerly Lactobacillus plantarum) dominated in the beginning of fermentation in both varieties. In the end, Lactiplantibacillus pentosus (formerly Lactobacillus pentosus) and Pediococcus ethanolidurans prevailed in Halkidiki and Conservolea varieties, respectively. As for yeasts, Kluyveromyces lactis/marxianus and Pichia manshurica prevailed at the onset of fermentation in Halkidiki and Conservolea varieties, whereas in the end Pichia membranifaciens dominated in both varieties.  相似文献

Recurrent protein folding motifs include various types of helical bundles formed by α-helices that supercoil around each other. While specific patterns of amino acid residues (heptad repeats) characterize the highly versatile folding motif of four-α-helical bundles, the significance of the polypeptide chain directionality is not sufficiently understood, although it determines sequence patterns, helical dipoles, and other parameters for the folding and oligomerization processes of bundles. To investigate directionality aspects in sequence-structure relationships, we reversed the amino acid sequences of two well-characterized, highly regular four-α-helical bundle proteins and studied the folding, oligomerization, and structural properties of the retro-proteins, using Circular Dichroism Spectroscopy (CD), Size Exclusion Chromatography combined with Multi-Angle Laser Light Scattering (SEC-MALS), and Small Angle X-ray Scattering (SAXS). The comparison of the parent proteins with their retro-counterparts reveals that while the α-helical character of the parents is affected to varying degrees by sequence reversal, the folding states, oligomerization propensities, structural stabilities, and shapes of the new molecules strongly depend on the characteristics of the heptad repeat patterns. The highest similarities between parent and retro-proteins are associated with the presence of uninterrupted heptad patterns in helical bundles sequences.  相似文献

In the framework of the detailed phytochemical analysis of the aerial parts of Sideritis syriaca, two novel acylflavones were isolated together with three acetylated flavone glycosides and acylated flavone glycosides. The novel acylflavones were identified as isoscutellarein 7-trans-p-coumarate and apigenin 7-,4′-bis(trans-p-coumarate). Their structures were elucidated by means of UV, 1D and 2D NMR, LC–DAD–MS and confirmed by HR-MS spectroscopy.  相似文献

Combined heat and power (CHP) plants with thermal stores may be suitable for sustainable energy production and the accommodation of fluctuating renewable energy sources. At the moment, in the UK, only a few CHP plants have thermal stores. Previous research has shown that thermal stores can improve the economics of CHP plants in the UK under the current market conditions. However, currently, it is only beneficial for CHP plants to sell their electricity to a third party, a Licensed Electricity Supplier, rather than to sell it directly to the power exchange market at prices which are much higher. If CHP plants aggregate, direct access to the power exchange market can become economically viable hence there is the possibility that thermal stores could further improve the economics of CHP plants under an aggregated electricity dispatch. This work firstly explains the conditions under which such plants could aggregate and act as a large power plant in the UK market, and secondly explores the most economic-size of gas engine and thermal store, in the case of aggregation, using energyPRO software and Excel spreadsheets. The work suggests that direct access to the power exchange market can improve the economics of the CHP plants. The highest Net Present Value (NPV), without heat dissipation, for a CHP plant exporting its electricity to the grid for a community heating load of 20 GW h, is more than £5 m, and is obtained for a 6 MW engine with a 28.2 MW h (900 m3) thermal store. The research suggests that such high electricity prices could make even larger plants more profitable than that; however, this can happen only if some of the produced heat is dissipated.  相似文献

The efficiency of the good agricultural practices (GAP) protocol AGRO 2-1 & 2-2, in advancing microbiological-quality of tomatoes and peppers, was studied in greenhouses at Ierapetra, Crete, Greece. The 240 tested vegetables-samples, produced under AGRO 2-1 & 2-2, showed satisfactory quality: Listeria monocytogenes absent per 25 g; Escherichia coli < 20 Colony Forming Units per gram (CFU/g); total coliforms 4.37–4.68 log CFU/g; aerobic plate counts 5.78–5.92 log CFU/g. Based on actual results and practices evaluation, we conclude that AGRO 2-1 & 2-2 can reduce microbial hazards for consumers and furthermore can establish practices in compliance to basic Euro-Retailer-Produce GAP (EUREPGAP) requirements.  相似文献

Well-characterized, chromosome-specific somatic cell hybrid panels are powerful tools for the analysis of the human genome. We have characterized a panel of human x hamster somatic cell hybrids retaining fragments of human chromosome 10 by fluorescence in situ hybridization and associated them to genetic markers. Most of the hybrids were generated by the radiation-reduction method, starting from a chromosome 10-specific monochromosomal hybrid, whereas some were collected from hybrids retaining chromosome 10-specific fragments as a result of spontaneous in vitro rearrangements. PCR was used to score the retention of 57 microsatellite markers evenly distributed along a well-supported framework genetic map containing 149 loci uniquely placed at 69 anchor points (odds exceeding 1,000:1), with an average spacing of 2.8 cM. As an additional resource for genomic studies involving human chromosome 10, we report the cytogenetic localization of a series of YAC and PAC clones recognized by at least one genetic marker. Somatic cell hybrids provide a powerful source of partial chromosome paints useful for detailed clinical cytogenetic and primate chromosome evolution investigations. Furthermore, correlation of the above physical, genetic, and cytogenetic data contribute to an emerging consensus map of human chromosome 10.  相似文献

Residues of the synthetic pyrethroid insecticide cypermethrin in fresh grapes of cv. Thomson Seedless and in raisins were studied. The commercial formulation Cymbush® 10% EC was applied according to the label onto the typical seedless variety and on a seed-producing clone. Samples were collected at time zero and at 15 or 21 days post-application. The raisins were produced either by submersion for 3 min in an aqueous solution of 3% potassium carbonate and 1% ethyl oleate, and dried for 15 days under direct sunlight or were dried without chemical processing. A gas chromatographic method was used for the analysis of residues. The residues in the grapes at the recommended preharvest interval (15 days) ranged from 0.23 to 0.41 mg kg -1 , whereas those in raisins ranged from 0.25 to 0.46 mg kg -1 .  相似文献

B lymphocytes are an indispensable part of the human immune system. They are the effective mediators of adaptive immunity and memory. To accomplish specificity against an antigen, and to establish the related immunologic memory, B cells differentiate through a complicated and strenuous training program that is characterized by multiple drastic genomic modifications. In order to avoid malignant transformation, these events are tightly regulated by multiple checkpoints, the vast majority of them involving bioenergetic alterations. Despite this stringent control program, B cell malignancies are amongst the top ten most common worldwide. In an effort to better understand malignant pathobiology, in this review, we summarize the metabolic swifts that govern normal B cell lymphopoiesis. We also review the existent knowledge regarding malignant metabolism as a means to unravel new research goals and/or therapeutic targets.  相似文献
