The human follicle-stimulating hormone receptor (FSH-R) consists of two distinct domains of >330 amino acids, the N-terminal extracellular exodomain and membrane-associated endodomain. The exodomain alone binds hormone with high affinity, whereas the endodomain is the site of receptor activation. Coordination of these two domains is essential for successful hormone action but little is known about their functional and structural relationship. In this communication, we report that exoloop 3 of FSH-R constrains follicle-stimulating hormone binding to the exodomain. When the FSH-R exodomain was prepared by truncating its endodomain, the hormone binding affinity of the exodomain was slightly improved, compared with the wild type receptor. The binding affinity was further improved by >3-fold when the exodomain was attached to the membrane-associated domain of CD8. These results suggest that the FSH-R endodomain attenuates hormone binding at the exodomain. As a first step to test this hypothesis, the 11 amino acids except Ala589 of exoloop 3 were individually substituted with Ala. Ala substitution for Leu583 or Ile584 improved the hormone binding affinity by 4-6-fold while totally abolishing cAMP induction, indicating an inverse relationship. The Ala substitution for Lys580 or Pro582 had a similar trend but to a lesser extent. This significant improvement in the binding affinity suggests that the four residues at the N-terminal region of exoloop 3 interact with the exodomain and constrain the hormone binding in the wild type receptor. This effect is specific since substitutions for other than the 4 residues did not improve the hormone binding affinity. Computer modeling shows that the 4 residues can be positioned on one side of exoloop 3. This result and the apparent inverse relationship of hormone binding and cAMP induction suggest that these two essential functions may work against each other. Therefore, hormone binding might be compromised to preserve cAMP inducibility while maintaining a reasonably high, but below maximum, binding affinity.  相似文献

The effects of outdoor ageing and of accelerated UV exposure on the grade of acrylonitrile butadiene styrene copolymer (ABS) used by UK manufacturers of motorcycle helmets were compared. Although the surface layer becomes embrittled the helmets still survive an impact test after conditioning at -20° C because the cracks are arrested before they penetrate the full thickness of the shell. A 4 mm thick rubber-toughened polycarbonate, used in helmets from continental suppliers, shows a transition from tough to brittle crack propagation behaviour at -15° C. For 4 mm thick ABS this transition had not occurred at the lowest test temperature of -20° C, but if 6 mm sheet is tested the transition is at -15° C. Temperature increases in the 40 sec delay between conditioning a helmet at -20° C and impact testing are shown to be significant.  相似文献

The wave propagation model investigated herein is based on the known fact that material discontinuities affect the propagation of elastic waves in solids. The change in certain material characteristics, such as a local change in stiffness or inertia caused by a crack or the presence of material damage, will affect the propagation of transmitted elastic waves and will modify the received signal. Wave frequencies associated with the highest detection sensitivity depend, among other things, on the type of structure, the type of material, and the type of damage. This paper presents a method of wave propagation, which can be further used to detect small delaminations in beam-like structures. Bulk single crystals of InxGa1-xAs (0.01 <x < 0.12) were successfully grown by the Liquid Encapsulated Czochraski (LEC) technique. These crystals are of high quality and can provide tunable substrate material for epitaxial application, since the lattice constant may be adjusted by varying the composition. Nominally undoped crystals were semi-insulating as grown ifx is less than about 0.05. Semi-insulating crystals with higher In composition can be obtained by chromium doping during crystal growth. The asgrown crystals were studied by 4.2 K photoluminescence spectroscopy (PL) and thermally stimulated current spectroscopy (TSC). Results from PL measurements indicate good crystal quality and impurity control. TSC spectra reveal several bulk traps in undoped and Cr doped semi-insulating samples. The activation energies of these traps decrease as In composition is increased. The amount of change in activation energy is about one third of the bandgap shrinkage due to increased In concentration.  相似文献

An Escherichia coli O157:H7 subtyping method based on PCR amplification of variable DNA sequences between the repetitive element IS3 was developed. Template DNA was prepared by boiling cells in Chelex. Two separate IS3 PCR amplifications were performed for each isolate: one with a single primer (primer IS3A) and one with two primers (primers IS3A and IS3B). The IS3 PCR subtyping method was applied to 35 epidemiologically related and unrelated E. coli O157:H7 isolates that had been previously characterized by pulsed-field gel electrophoresis (PFGE). PFGE identified 25 different subtypes (difference of one or more bands). PCR with single primer IS3A and primer pair IS3A-IS3B identified 6 and 14 different subtypes, respectively. By combining the results of the two PCR amplifications, 15 different IS3 PCR subtypes were identified. While not as sensitive as PFGE, IS3 PCR subtyping grouped all outbreak-related isolates. IS3 PCR banding patterns were reproducible between amplifications and between subcultures. IS3 PCR could serve as a simple, rapid screening method for the identification of unrelated E. coli O157:H7 isolates.  相似文献

A system is described that controls the dyeing process so as to produce a package within a final unlevelness tolerance in the minimum time. This is achieved by incorporating into the control algorithm a mathematical model capable of predicting the unlevelness in a dyed package from the current state of the system. The use of such a general strategy allows production of dyeings of acceptable quality in minimum time and requires little intervention from the machine operator.  相似文献

The putative role of the nuclear nucleoside triphosphatase (NTPase) is to provide energy to the nuclear pore complex for poly A(+) mRNA export. Previous work has demonstrated that liver nuclear NTPase activity is greater in 6 month old corpulent (cp/cp) female JCR:LA rats, a hyperlipidemic rat model, compared to lean (+/?) animals. This increase appeared to be related to increases in nuclear membrane cholesterol content. The current study extended these initial data to compare NTPase activity as a function of age and sex in isolated JCR:LA-cp rat liver nuclei, to further test the hypothesis that nuclear membrane cholesterol may modulate NTPase activity. NTPase activity was increased in cp/cp female animals compared to +/? females at all ages studied, with Vmax values increased by 60-176%. Membrane integrity of cp/cp female nuclei was reduced compared to +/? female nuclei. Nuclear membrane cholesterol levels increased linearly with age by 50, 150 and 250% in 3, 6 and 9 month old cp/cp females over leans. In contrast, nuclei from cp/cp males exhibited only minor, isolated changes in NTPase activity. Furthermore, there were no significant changes in nuclear cholesterol content or membrane integrity in the less hyperlipidemic male animals at any age. These data suggest that altered lipid metabolism may lead to changes in nuclear membrane structure, which in turn may alter NTPase activity and functioning of the nuclear pore complex.  相似文献

Wound infections retard healing rates and can lengthen the time a patient is required to spend in hospital. Obviously it is desirable to minimize or remove pathogenic organisms to allow wound healing to progress optimally. At present there are a large range of topical applications available in the form of antibiotics or antiseptics, but the toxic effects of these agents may outweight the benefits. In this study two conventional antiseptic agents were examined together with a range of phosphate-based glasses which were designed to release metal ions into an aqueous environment. As a control a series of standard metal ion solutions were prepared and examined in the same test system. The metal ions examined are considered important in the wound healing environment either for their potential bacteriostatic effects or indeed their potential for increasing cellular activity. This part of the study examined the effects of these agents on mammalian cells in vitro, using a standard MTT assay to assess the cytotoxicity or activity of cultured mouse fibroblasts. It was found that chlorhexidene, a commonly used antiseptic agent, was toxic at low levels. The controlled release glasses, in most cases, increased the rate of cell activity before reaching toxic levels. This effect was also found with the standard metal solutions.  相似文献
