Fertilization of the sea urchin egg is accompanied by a sequence of structural changes in the egg cortex that include exocytosis, endocytosis, and microvillar growth. This architectural reorganization is coordinated by two intracellular signals: a rapid, transient rise in cyto-solic free calcium and a slower, longer lasting increase in cytoplasmic pH. In this report we provide ultrastructural views of these events in quick-frozen eggs and discuss their relationship to the calcium and pH signals.  相似文献

Although self-help/mutual-help groups, like Alcoholics Anonymous, are important factors in the recovery of many chemically dependent individuals, the large dropout rates from such groups and the awareness that many who drop out do not return to dependent behavior raises interesting questions about other paths to sustained abstinence and recovery. Current emphasis on treatment outcome studies provides a means for researching factors in both those who recover with and without the assistance of self-help/mutual-help groups. The authors suggest some questions which might be investigated.  相似文献

McCloskey and Zaragoza (1985a) have recently argued that all item-specific retroactive interference (RI) can be attributed to response bias or demand characteristics. These sources of RI were eliminated in the present studies by a modified recognition test that excluded the interpolated item. Nature pictures were shown; some were followed by related pictures (experimental), and others were followed by dissimilar pictures (control). When forced to choose between two related pictures (the original and a new picture), the original picture was chosen more often for control than for experimental items (Experiments 1 and 2). Paired-associate studies were cited that have found RI in analogous matching recognition tests. These findings contradict McCloskey and Zaragoza's conclusion and must be attributed to some other cause of item-specific RI, such as trace alteration (e.g., Loftus, 1979). Experiments 3 and 4 showed that most items must be accessible in order to detect RI in the forced-choice modified test. However, even with sensitive measures, RI has not always been found in modified recognition. Suggestions are offered to explain the discrepant results. We present a structural study of two Nd3+/MgO codoped LiNbO3 waveguides fabricated by two different methods: ion-implantation and proton-exchange. Rutherford backscattering spectrometry in combination with channelling was used to study the influence of the waveguide fabrication method on the possible modification of the LiNbO3 crystal structure within the waveguiding layer. The results show that the ion-implanted waveguide mainly maintains the same properties as the virgin crystal, apart from a deterioration of a thin surface layer, whilst the proton-exchanged waveguide exhibits a quite different behaviour, although its surface is less damaged.  相似文献

The results of a parametric study of torsional coupling effects in the response of asymmetric buildings to a grouped selection of 45 strong motion earthquakes from Europe, North America, the Middle East and Southern Pacific are presented. The period dependency of the key response parameters is studied in relation to the site dependent criteria employed by certain earthquake-resistant design codes for specifying the lateral shear forces to be resisted by the structural frames and/or walls. Unlike lateral forces, the torsional response is relatively insensitive to the ration of peak ground accelaration to velocity, which has been used in previous studies as a measure of the effect on earthquake ground motions of site soil conditions and epicentral distance. The results show that the effect of torsional coupling on edge displacement response is more pronounced in stiff, short period structures. However, the dynamic shear and torque responses are relatively insensitive to variations in structural period. Codified design provisions for torsional effects specified in the United States, Canadian and European seismic building regulations are assessed, along with alternative proposals, in relation to the analytical results. It is concluded that in some cases special provisions are needed for short period asymmetric systems, a feature which has been neglected by the codes and inadequately accounted for in previous proposals.  相似文献

Whole cell voltage- and current-clamp recordings were obtained from mesencephalic trigeminal sensory (Mes 5) neurons identified visually in thin brain stem slices of neonatal rats with the use of infrared video microscopy. These cells exhibited accommodation in spike discharge responses to depolarizing current injection protocols whose duration differed as a function of holding potential (-50 vs. -65 mV). Several spikes were elicited before the membrane response accommodated from -50 mV, whereas from -65 mV only single action potentials were evoked. In response to similar protocols, application of the K+ channel blocker 4-aminopyridine (4-AP) (50 microM to 2 mM) caused sustained repetitive spiking whereas tetraethylammonium (TEA) (10-30 mM) did not cause repetitive spiking. In voltage clamp, 4-AP application (100 microM) revealed a sustained outward current (I4-AP) that was active between -60 and -30 mV. I4-AP was responsible for suppressing sustained repetitive spiking behavior, producing accommodation under normal circumstances. TEA application in voltage clamp revealed a sustained outward current evoked positive to -40 mV. Two transient outward currents (TOCs) were identified by prepulse protocols typically used to characterize A-type currents: a 4-AP-insensitive fast TOC, and a slow TOC (ITOC-S) sensitive to 4-AP (> 500 microM). A Ca(2+)-dependent outward current that activated positive to -30 mV was also characterized. A mathematical model of a Mes 5 neuron was assembled from our voltage-clamp records to simulate the dynamic interaction of outward currents during membrane excitation. We conclude that in Mes 5 neurons, the 4-AP-sensitive currents ITOC-S and I4-AP determine the duration of spike trains. In particular, the noninactivating I4-AP determines whether cells exhibit sustained repetitive discharge or accommodate in response to depolarizing current. Neurotransmitter modulation of this current or modulation of the resting membrane potential could modify the output properties of Mes 5 neurons, and therefore the properties of these currents must be incorporated into our current understanding of how these cells contribute to shaping oral-motor pattern generation.  相似文献

Acrosomal integrity of bovine spermatozoa was evaluated using naphthol yellow S-erythrosine B stain with bright-field microscopy, .2% glutaraldehyde fixation with Nomarski optics, and live wet smears with Nomarski optics to compare the evaluation techniques. Four ejaculates per two Holstein bulls were enriched through a Percoll gradient, capacitated with heparin, and acrosome-reacted with lysophosphatidylcholine to prepare spermatozoa for in vitro fertilization. Spermatozoa samples were taken after each preparatory step, and percentage of intact acrosomes was evaluated with the stain, .2% glutaraldehyde, and live wet smear. Acrosomal integrity decreased with each preparatory step across all techniques. The decrease in integrity from heparin to lysophosphatidylcholine indicated that all methods detected the acrosome reaction. Technique means across preparation steps showed no differences. Correlations between microscopy techniques were high. The live wet smear used no fixation methods. Both the naphthol yellow S-erythrosine B stain and the .2% glutaraldehyde techniques employed fixation steps that may have introduced artifacts that could influence the data. The live wet smear evaluated by Nomarski optics allowed a comparable evaluation of the acrosome-reacted bovine spermatozoa. Comparing brightfield with differential interference contrast microscopy for detecting the acrosome reaction in bovine spermatozoa showed that both methods were equally accurate.  相似文献

Molecular electrostatic potential (MEP) maps of certain 3-methoxy flavone derivatives having different anti-picornavirus activities have been studied. Geometries of the molecules were optimised and charge distributions computed using the AM1 molecular orbital method. Hybridization displacement charges (HDC) were combined with the L?wdin charge distributions to compute the MEP maps. Reliability of the method of computing MEP maps was tested by studying certain other molecules for which ab initio MEP results are available. The anti-picornavirus activities of the flavones have been shown to be related with negative MEP values in two regions, one near the 3-methoxy group and another in a diagonally opposite region near the substituent attached to the C7 atom of the molecules.  相似文献
