Bilayers composed of phosphatidylcholine initially resist catalysis by phospholipase A2. However, after a latency period, they become susceptible when sufficient reaction products (lysolecithin and fatty acid) accumulate in the membrane. Temperature near the main bilayer phase transition and calcium concentration modulate the effectiveness of the reaction products. The purpose of this study was to examine the individual contributions of lysolecithin and palmitic acid to the susceptibility of dipalmitoylphosphatidylcholine vesicles and to rationalize the effects of temperature and calcium. Various fluorescent probes (Prodan, Laurdan, pyrene-labeled fatty acid, and dansyl-labeled phospholipid) were used to assess changes in the ability of the reaction products to perturb the bilayer and to affect the interactions with the enzyme. Un-ionized palmitic acid decreased bilayer polarity and perturbed the membrane surface exposing some of the Prodan to bulk water. Lysolecithin increased bilayer polarity and the rate of dipolar relaxation in response to the excited states of Laurdan and Prodan. A combination of the individual contributions of each product was observed when palmitic acid and lysolecithin were present together at low calcium, and the effects of lysolecithin dominated at high calcium. Palmitic acid, but not lysolecithin, promoted the binding of phospholipase A2 to the bilayer surface in the absence of calcium. Lysolecithin reduced the ability of fatty acid to enhance binding apparently by altering the structure of fatty acid domains in the membrane. Furthermore, increased temperature and ionization of the fatty acid tended to cause segregation of bound phospholipase A2 into domains poor in phospholipid content which presumably impeded bilayer hydrolysis. In contrast, un-ionized palmitic acid and lysolecithin promoted hydrolysis by augmenting a step distal to the adsorption of enzyme to the bilayer. This kinetic response to lysolecithin was calcium-dependent. A model accounting for these varied influences of the reaction products is presented.  相似文献

This paper highlights the future perspectives of sustainable tribology by examining the economic, environmental and social impact of three tribological case studies. One case study examines the sustainability and durability of micro-CHP systems looking the tribological phenomena generated within a scroll expander system. The scroll is the main part of a specific micro-CHP system and experiences wear and cavitation damage. The tribological optimization of the scroll expander improves the sustainability of the micro-CHP unit while it has a serious economic and environmental impact to the consumers and to the society in general. Another case study is focused on friction and wear performance of lifeboat launch slipways. The causes of high friction and wear during the RNLI's lifeboat launches along an inclined slipway are investigated with a view to reducing the environmental impact due to slipway panel wear and lubricant release into the marine environment. The project encompasses the sustainable design of slipway panels using design modifications based on tribological investigations to double their lifespan, while environmental and economic impact was significantly reduced by the use of biodegradable greases and water as lubricants. The final case study involves an investigation of recycled plastic materials to replace polyurethane used on skateboard wheels, scooters and similar applications. Polyurethane (PU) is difficult to recycle. With the dwindling resources and environmental problems facing the world today, recycling for both waste reduction and resource preservation has become an increasingly important aspect of sustainability. The tribological results showed that recycled polycarbonate plastic can effectively act as a substitute to polyurethane wheels. Moreover, sustainability considerations showing the environmental benefits of the use of recycled plastics over PU include reducing the CO2 footprint by 50% and the energy consumed by 60%, among other benefits. These case studies emphasise the importance of sustainable tribology in our epoch showing that increased sustainability performance can be achieved through tribology to a significant extent in many cases, providing stability to our world and more viable long term growth to our societies.  相似文献

We are interested in building structured overlap-ping grids for geometries defined by Computer-Aided-Design (CAD) packages. Geometric information defining the boundary surfaces of a computation domain is often provided in the form of a collection of possibly hundreds of trimmed patches. The first step in building an overlapping volume grid on such a geometry is to build overlapping surface grids. A surface grid is typically built using hyperbolic grid generation; starting from a curve on the surface, a grid is grown by marching over the surface. A given hyperbolic grid will typically cover many of the underlying CAD surface patches. The fundamental operation needed for building surface grids is that of projecting a point in space onto the closest point on the CAD surface. We describe a fast and robust algorithm for performing this projection which makes use of a fairly coarse global triangulation of the CAD geometry. Before the global triangulation is constructed the connectivity of the model is determined by an edge-matching algorithm which corrects for gaps and overlaps between neighbouring patches. The thermal properties of 12 varieties of cowpea flour were studied by differential scanning calorimetry (DSC). Flour samples were prepared to a paste of 60% moisture content and sealed in standard DSC pans. Samples were scanned at a heating rate of 5 degrees C/min over a scan range of 40- 130 degrees C. Samples exhibited single major endotherms, which occurred over varied temperatures. The transition enthalpy (deltaH) ranged between 1.4 J/g and 4.7 J/g. Transition onset (T(o)) and transition peak (T(p)) temperatures ranged between 75-78 degrees C and 78-82 degrees C,respectively. All the DSC parameters measured varied significantly among the varieties. The transition enthalpy (deltaH) was the most discriminating parameter and accounted for 80% of the total variance. The major chemical components of cowpea flour, starch amylose and protein are significant predictors of deltaH. Protein denaturation appears to be a significant modification which occurs during processing of cowpea seeds to flour. The transition enthalpy deltaH could become an important functional index of cowpea flour when related to some quality parameters in products that contain the flour.  相似文献

The primary drivers for space instruments are the environment and survivability, support assets, delivery cost, and lack of accessibility. Survivability deals primarily with environments, such as launch and depressurization, extremes of temperature, high vacuum, radiation, and hypervelocity impacts. The cost to deployment is directly coupled to launch services, which are inherently expensive. These costs are nonlinear functions of the mass and volume of satellite payloads. The “user interfaces” for space-borne instruments are high-cost assets because they include mission operations centers, satellite hacking station(s), and ground networks. STUDY OBJECTIVE: To define the epidemiology, clinical manifestations, and long-term complications of respiratory viral infections in adult lung transplant recipients. DESIGN: Retrospective review of the records of 122 adult lung transplant recipients over a 5-year period at one institution. RESULTS: Ten episodes of infection with respiratory syncytial virus, parainfluenza, influenza, or adenovirus were identified. All patients presented with symptoms of respiratory tract infection. Two patients died acutely and four patients subsequently had development of obliterative bronchiolitis (OB). CONCLUSIONS: These data suggest community respiratory viral infections cause significant morbidity and mortality in lung transplant recipients. Further prospective studies are warranted to clarify the relationship between respiratory viral infection and OB and to define the optimal therapy for these viral infections.  相似文献

Abortion incidence and services in the United States, 1995-1996   总被引:1,自引:0,他引:1  
CONTEXT: In the 1980s, the number of abortion providers in the United States began to decline, and more recently, so has the number of abortions performed. Whether the decline in service providers, which was last documented in 1992, is continuing and whether this influences the availability and number of abortions is of public interest. METHODS: In 1997, the Alan Guttmacher Institute conducted its 12th survey of all known abortion providers in the United States. The number and location of abortion providers and abortions were tabulated for 1995 and 1996, and trends were calculated by comparing these data with those from earlier surveys. Limited data were also gathered on types of abortion procedures. RESULTS: Between 1992 and 1996, the number of abortions fell from 1,529,000 to 1,366,000, and the abortion rate decreased from 26 to 23 per 1,000 women aged 15-44. The number of providers fell 14%, to 2,042, with the greatest decline among hospitals and physicians' offices rather than clinics. Eighty-six percent of counties had no known abortion provider, and 32% of women aged 15-44 lived in these counties. Of the country's 320 metropolitan areas, 89 had no known abortion provider, and for an additional 12, fewer than 50 abortions each were reported. Seventy percent of abortions were performed in specialized clinics and only 7% in hospitals. In the first half of 1997, early medical abortions were being offered in about 160 facilities, virtually all of which were also providers of surgical abortions. CONCLUSIONS: While abortion services in some areas of the country have declined since 1992 and many women continue to have limited access to providers, other factors have probably had more influence on the level of abortions performed. Early medical abortion methods are too new to be a measurable factor in abortion access.  相似文献

A fixed-film continuous-flow photobioreactor was used for the removal of hydrogen sulfide from synthetic industrial wastewater and conversion of it to elemental sulfur using sixteen 150 mm length Tygon tubes as the active part of the reactor. Three different tube sizes (internal diameters (ID) 6.4, 3.2, and 1.6mm) were used to investigate the effect of tube size on reactor performance. The reactor with 1.6mm tube attained a higher sulfide loading (1451 mg/Lh) than previously reported in the literature. High bacterial concentration, light to volume ratio and intimate contact between attached bacteria and influent sulfide resulted in excellent performance of the 1.6mm tube reactor.  相似文献
