The viability and β‐galactosidase activity of four Lactobacillus strains in milk drink containing gums during 28 days of refrigerated storage at 4 °C were assessed. The population of Lactobacillus rhamnosus GGB101 and Lactobacillus rhamnosus GGB103 were maintained, whereas the population of Lactobacillus reuteri DSM20016 and Lactobacillus reuteri SD2112 significantly decreased. The recommended level of 6 log CFU g?1 was exceeded for all tested trains throughout storage. The highest viable number of Lactobacillus rhamnosus GGB103 (8.76 ± 0.03 log CFU mL?1) was obtained in the product containing carrageenan–maltodextrin. The addition of guar–locust bean–carrageenan led to 20‐fold increase in the level of β‐galactosidase activity for L. rhamnosus GGB101 (1208 ± 2.12 Miller units mL?1) compared to the control (61 ± 2.83 Miller units mL?1). Our results suggested that gums could be added to milk to improve viability and enhance β‐galactosidase activity of Lactobacillus. 相似文献

In submerged cultures performed in chemically defined fermentation medium containing glucose and glutamate, the growth and production of water‐soluble red pigments and citrinin by the filamentous fungus Monascus ruber were studied under various carbon/nitrogen (C/N) ratios. The specific production of the red pigments was optimal at a glucose/glutamate ratio of about 10 and then steadily decreased at higher C/N ratio. In contrast, the production of the mycotoxin increased with increased C/N with an optimum in the range of 30–45. In a fed‐batch mode, it was also found that the production of pigments was not favoured in fed‐batch mode by feeding the medium with glucose while keeping the C/N ratio lower than 10. This low production likely resulted from concurrent high accumulation of L‐malic acid that was reported to inhibit this production. In contrast, this mode of cultivation was rather favourable for the production of the mycotoxin. 相似文献

A cell vertex finite volume method for the solution of steady compressible turbulent flow problems on unstructured hybrid
meshes of tetrahedra, prisms, pyramids and hexahedra is described. These hybrid meshes are constructed by firstly discretising
the computational domain using tetrahedral elements and then by merging certain tetrahedra. A one equation turbulence model
is employed and the solution of the steady flow equations is obtained by explicit relaxation. The solution process is accelerated
by the addition of a multigrid method, in which the coarse meshes are generated by agglomeration, and by parallelisation.
The approach is shown to be effective for the simulation of a number of 3D flows of current practical interest.

ABSTRACT: The objective of this work was to observe the microstructure of feta cheese using confocal scanning laser microscopy. Low fat cheese and cheese containing capsule-producing cultures were made. The protein network was observed using the reflectance mode of the confocal microscope. Fat was stained by Nile red dye diluted with whey. More even distribution of fat with a larger number of smaller globules was observed in cheese made with noncapsule-forming culture compared to that made with the capsule-forming culture. The levels of mRNA expression of three UDP-N-acetyl-alpha-D-galactosamine:polypeptide GalNAc N-acetylgalactosaminyltransferases (GalNAc-transferases) were quantified for human adenocarcinoma cell lines from pancreas, colon, stomach, and breast. Two of the GalNAc-transferases, GalNAc-T1 and GalNAc-T2, were expressed constitutively and at low levels in most or all cell lines examined. A third GalNAc-transferase, GalNAc-T3, was differentially expressed. Well-differentiated adenocarcinoma cell lines expressed high levels and moderately differentiated cell lines expressed lower levels of GalNAc-T3. Cell lines classified as poorly differentiated failed to express GalNAc-T3 mRNA at levels that could be detected by Northern blot analysis. Differential expression of the GalNAc-T3 protein was confirmed in these cell lines by Western blotting. We propose that glycosylation in tumor cell lines may be regulated in part by differential expression of GalNAc-transferases, and we suggest that GalNAc-T3 gene expression may be a molecular indicator of differentiated adenocarcinoma. 相似文献

One of the frequently discussed applications of artificial intelligence in motion control is the replacement of a standard proportional plus integral (PI) speed controller with a fuzzy logic (FL) speed controller. Regardless of all the work, it appears that a thorough comparison of the drive behavior under PI and FL speed control is still missing. This paper attempts to fill in this gap, by providing an in-depth comparison of operation of a vector-controlled permanent-magnet synchronous motor, using at first an experimental rig. Speed responses, obtained under PI and FL speed control, are recorded and compared for a variety of operating conditions. The transients studied include response to a large step speed command from standstill with nominal inertia and an increased inertia, response to small step speed reference change, and response to step load torque application. The transient behavior is examined for various initial speed settings, so that a thorough comparison is enabled. Experimental results are further supplemented with a set of simulation results, obtained using a different permanent-magnet machine and a different FL controller. Better generalization of the results is enabled in this way. It is shown that superiority of the FL speed control is less pronounced than it is often portrayed in the literature on the basis of limited comparisons.
