Quantification of aroma compounds in an orange juice reconstituted from concentrate which had been stored at 37 °C for 4 weeks (forced storage) revealed an increase in the concentrations of, in particular, dimethyl sulphide, 2-methoxy-4-vinylphenol, α-terpineol, and 4-hydroxy-2,5-dimethyl-3(2H)-furanone (4-HDF) in comparison with the same orange juice before storage. On the other hand, clearly lower concentrations were found for octanal, decanal, (R)-α-pinene, linalool, and (E)-β-damascenone after storage, while the concentrations of vanillin and carvone remained nearly constant. Similar results were found for the same aroma compounds after storage of the orange juice at 20 °C for 1 year. Sensory experiments corroborated the importance of 2-methoxy-4-vinylphenol and dimethyl sulphide for the typical stale off-flavour of the stored orange juice, while the omission of e.g., α-terpineol in model mixtures could not be detected. Under both storage conditions (37 °C for 4 weeks or 20 °C for 1 year), the breakthrough odour thresholds of α-terpineol and 4-HDF were not reached, while the concentrations of dimethyl sulphide and 2-methoxy-4-vinylphenol clearly exceeded their breakthrough odour thresholds, thus confirming the crucial role of these odorants for the off-flavour of stored orange juice from concentrate. The aim of the presented research activities was to identify mechanical, thermal, and chemical factors possibly linked to the formation of WECs (White Etching Cracks). By means of a systematic variation of various influencing parameters, the significance of each of those was investigated. It is hoped that, once the parameters promoting WECs have been identified, the physical and chemical mechanisms responsible for WEC can be thoroughly understood in the near future. This would allow to prevent costly premature bearing failures, e.?g. given in wind turbines.Four research centers in Kaiserslautern, Münster and Hannover (Institute of Machine Elements, Gears and Transmissions (MEGT), Technische Universität Kaiserslautern, Materials Testing Group (AWP), Technische Universität Kaiserslautern, Institute of Physics, Westfälische Wilhelms-Universität Münster, and Institute for Machine Design and Tribology (IMKT), Leibniz University Hannover) provide their expertise and laboratory facilities for this purpose.At IMKT full bearing investigations with cylindrical roller thrust bearings and model tests with a special ring-roller-ring tribometer were performed under varied test conditions. In a theoretical work package, the stresses induced to the bearing surface were simulated.At “Physikalisches Institut” in Münster surfaces of the bearing washers (and rolling elements) were analysed applying Time-of-Flight secondary ion mass spectrometry (ToF-SIMS) to determine the actual respective chemical composition of the tribofilms.At MEGT component tests were carried out on a three-axis dynamic test rig with radially loaded cylindrical roller bearings. The internal bearing dynamics and, above all, the associated distribution of frictional energy were analysed by means of multi-body simulation.The AWP concentrated on synthesized multi-axial dynamic stresses aimed at mirroring the stresses in a rolling contact apllied on laboratory test specimens to investigate crack initiation and growth in depth.  相似文献

Contemporary inoculation scholarship has focused on the process of resistance to persuasion, and recently begun to examine various incidental effects that may accompany inoculation treatments. This study considers how talk fits within both of these areas, not only as a byproduct of inoculation, but also as a potentially important contributor to the process of resistance. Results indicate inoculation not only enhances talk about the target issue, but such talk appears to bolsters resistance to subsequent counterattitudinal messages.  相似文献

The disruption of six novel yeast genes has been realized in two genetic backgrounds. Six open reading frames (ORFs) from chromosome IV, YDR013w, YDR014w, YDR015c, YDR018c, YDR020c and YDR021w, were disrupted using the KanMX4 marker and PCR-targeting with long flanking regions homologous (LFH) to the target locus. The deletants were verified at the molecular level, using PCR and Southern analysis. Sporulation and tetrad analysis revealed that ORFs YDR013w and YDR021w (also known as FAL1) are essential genes. Microscopical observations showed that ydr013wDelta haploid cells were blocked after one or two cell cycles and presented heterogeneous bud sizes. The ydr021wDelta haploid cells gave rise to microcolonies of about 20 cells. The other four ORFs are non-essential. Basic phenotypic analysis of the non-lethal deletant strains did not reveal any significant differences in cell morphology, growth on different media and temperatures, sporulation and mating efficiency between parental and mutant strains in the FY1679 background.  相似文献

The CD53 antigen is a member of the tetraspan family of proteins with unknown function. Stimulation of rat IR938F B-cell lymphoma cells with monoclonal antibody MRC OX44 (anti-rat CD53) triggered a homotypic adhesion reaction which reached a maximum effect at 24 hr. This effect occurred at 37 degrees C but not at 4 degrees C. Adhesion was prevented by removal of divalent cations, Ca2+ and Mg2+, with EGTA and EDTA as chelating agents. The adhesion induced by MRC OX44 was inhibited by cycloheximide and actinomycin D, suggesting that de novo protein synthesis was required for this effect. The addition of mAb WT1 against rat LFA-1 (CD11a) antigen had no effect on adhesion, suggesting that the cell-cell interaction is not mediated by the expression of LFA-1 antigen. The intracellular signals required to induce adhesion were inhibited by two tyrosine kinase inhibitors, genistein and piceatannol. Wortmannin, a selective inhibitor of phosphoinositide 3-kinase activity, completely blocked adhesion. Two protein kinase C inhibitors, H7 and bisindolylmaleimide, inhibited the adhesion, suggesting that part of the signal is mediated by PKC. Electron microscopy of aggregated cells showed that the interaction is localized to short membrane regions, where contact areas of higher density in opposing zones from both cells were detected. We postulate that there is a common adhesion mechanism that is modulated by several tetraspan family members and associated proteins. This adhesion structure might represent a novel form of cell communication among lymphoid cells.  相似文献

2-nitro 7-methoxy naphtho [2,1-b] furan (R 7000) and 2-nitro 8-methoxy naphtho [2,1-b] furan, which are highly mutagenic on the bacterium Salmonella typhimurium have little activity on the yeast Saccharomyces cerevisiae. Application of an aroma extract dilution analysis on the entire volatile fraction isolated from an orange juice freshly reconstituted from concentrate revealed 40 odour-active constituents in the flavour dilution (FD) factor range of 4–2,048. Among them, ethyl butanoate and linalool showed the highest FD factor of 2,048, followed by octanal with an FD factor of 512. Thirty-six of the 40 odour-active compounds detected could be identified, all of which have previously been reported as volatile constituents of various orange juices. Quantification of 17 key odorants by stable isotope dilution assays followed by a calculation of odour activity values (OAVs) on the basis of odour thresholds in water or citrate buffer (pH 3.8), respectively, revealed the following most important odorants in the overall aroma of the freshly reconstituted juice: (R/S)-linalool, (R)-limonene and (S)-ethyl 2-methylbutanoate with the highest OAVs (>1,000) followed by octanal, (R)-α-pinene, ethyl butanoate, myrcene, acetaldehyde, decanal and (E)-β-damascenone with OAVs > 100. A model mixture containing all 14 aroma compounds with OAVs > 1 in their actual concentrations in the juice showed a good similarity with the aroma of the original orange juice under investigation, thus corroborating that the key odorants of a freshly reconstituted orange juice were characterised for the first time.  相似文献

Exposing cells to DNA damaging agents, such as ionizing radiation (IR) or cytotoxic chemicals, can cause DNA double-strand breaks (DSBs), which are crucial to repair to maintain genetic integrity. O-linked β-N-acetylglucosaminylation (O-GlcNAcylation) is a post-translational modification (PTM), which has been reported to be involved in the DNA damage response (DDR) and chromatin remodeling. Here, we investigated the impact of O-GlcNAcylation on the DDR, DSB repair and chromatin status in more detail. We also applied charged particle irradiation to analyze differences of O-GlcNAcylation and its impact on DSB repair in respect of spatial dose deposition and radiation quality. Various techniques were used, such as the γH2AX foci assay, live cell microscopy and Fluorescence Lifetime Microscopy (FLIM) to detect DSB rejoining, protein accumulation and chromatin states after treating the cells with O-GlcNAc transferase (OGT) or O-GlcNAcase (OGA) inhibitors. We confirmed that O-GlcNAcylation of MDC1 is increased upon irradiation and identified additional repair factors related to Homologous Recombination (HR), CtIP and BRCA1, which were increasingly O-GlcNAcyated upon irradiation. This is consistent with our findings that the function of HR is affected by OGT inhibition. Besides, we found that OGT and OGA activity modulate chromatin compaction states, providing a potential additional level of DNA-repair regulation.  相似文献

Recent microwave experiments have shown frequency dependent influences on the growth rate of bacteria. To determine whether microwaves are able to affect growth (or to induce lesions in cellular DNA of yeast cells), experiments were performed with millimeter microwaves at frequencies between 70 and 75 GHz. Saccharomyces cerevisiae cells were irradiated on millipore filter discs placed on agar plates in open petri dishes. A diploid strain of yeast (D5, Zimmerman), that is sensitive to genetic insult was used to study the effects of temperature and of microwave irradiation on cell survival, induction of mitotic recombination, and induction of cytoplasmic "petite" mutations. No evidence of altered survival, impaired function, or structural injury was seen at either frequency, even at power densities as high as 60 mW/cm2. Conventional heating had no deleterious effects until temperatures of specimens exceeded 50 degrees C. In addition, two haploid strains of yeast of opposite mating type were compared with respect to temperature and microwave treatment for formation of zygotes. The elevation of temperature due to the microwave treatment at 60 mW/cm2 and 2 mm distance was estimated to correspond to 3 degrees C.  相似文献

Decision making is influenced by social cues, but there is little understanding of how social information interacts with other cues that determine decisions. To address this quantitatively, participants were asked to learn which of two faces was associated with a higher probability of reward. They were repeatedly presented with two faces, each with a different, unknown probability of reward, and participants attempted to maximize gains by selecting the face that was most often rewarded. Both faces had the same identity, but one face had a happy expression and the other had either an angry or a sad expression. Ideal observer models predict that the facial expressions should not affect the decision-making process. Our results however showed that participants had a prior disposition to select the happy face when it was paired with the angry but not the sad face and overweighted the positive outcomes associated with happy faces and underweighted positive outcomes associated with either angry or sad faces. Nevertheless, participants also integrated the feedback information. As such, their decisions were a composite of social and utilitarian factors.
