The peripherin gene has three potential ATG translation initiation sites at positions 38, 56, and 290. The second ATG has been proposed to be the initiation codon used for translation of the protein, but there is no experimental evidence for this conjecture. We have isolated a full-length peripherin cDNA (designated as p61-11) from a rat brain cDNA library. Upon sequencing, we found that this cDNA contains a point mutation at the second potential translation initiation codon, which changes this ATG to ACG. When expressed in SW13 cl.2 vim- cells, a cell line without any detectable cytoplasmic intermediate filaments, the protein product of p61-11 cannot form a filamentous network and the major product is 45 kDa in size, which is most likely initiated from the third ATG. The protein product from the first ATG (57 kDa in size) of p61-11 is also detected albeit in smaller amounts. We introduced a frame-shift mutation upstream of the third ATG in p61-11 to create p61-11FS and showed that the third ATG is able to initiate translation efficiently even in the presence of the first ATG, and the 45 kDa protein leads to a diffuse nonfilamentous staining pattern in vim- cells confirming that the first ATG may not be the preferred translation initiation codon, since it cannot suppress a downstream ATG. We increased the translation efficiency from the first ATG of p61-11 by mutating the three nucleotides preceding this first ATG and thereby placing it in a better Kozak consensus sequence for translation initiation. The resulting 57 kDa protein is able to form a filamentous network in vim- cells. We corrected the mutation in the original p61-11 by polymerase chain reaction and generated two peripherin constructs: perM1M2 (which contains all three translation initiation codons) and per delta 1M2 (the first ATG is deleted, but the other two are present). When transfected, their protein products, about 57 kDa in size, form filamentous networks in the absence of other cytoplasmic intermediate filaments. Since there is no 45 kDa protein detected for these latter two constructs, it is reasonable to conclude that in the presence of the second ATG, little or no translation is initiated from the third ATG. Taken together, these results strongly suggest that the second ATG is the preferred translation initiation codon for the peripherin gene.  相似文献

According to hopelessness theory, hopelessness expectancy is the proximal, sufficient cause of hopelessness depression. Consequently, hopelessness expectancy is viewed as mediating the influence of all other factors on hopelessness depression. Using a longitudinal research design, hopelessness expectancy was examined as a mediator of the relation between illness attributions and hopelessness depression in a sample of 57 adults with rheumatoid arthritis. Although hopelessness expectancy was a strong predictor of hopelessness depression, it moderated rather than mediated the relation between attributions and depression. Finding support for a moderating rather than a mediating model is inconsistent with theory but is consistent with the findings of J. H. Riskind et al (1987). In this study a set of thin Hydroxyapatite (HA) [Ca10(PO4)6(OH)2] coatings was deposited on heated silicon and titanium substrates using Ion Beam Assisted Deposition (IBAD). The effects of substrate temperature and processing parameters on the microstructural properties and composition of the coatings are being studied. Analytical techniques include transmission electron microscopy (TEM), scanning transmission electron microscopy (STEM) with an energy dispersive X-ray spectroscopy (EDS), as well as scanning electron microscopy (SEM) with EDX, X-ray diffraction (XRD), and Fourier Transform Infrared Spectroscopy (FTIR). The current results indicate that as substrate temperature increases the Ca/P ratio of the coatings both on titanium and silicon substrates increases. The crystallinity of the coatings and the number of calcium phosphate compounds within the coating including HA also increases. STEM-EDS revealed an atomically diffused intermediate layer at the interface between the coating and substrate. XRD results along with TEM selected area diffraction (SAD) revealed that the coatings are composed of HA, other calcium phosphate, and calcium oxide compounds.  相似文献

This study examined whether reactance would negatively influence treatment outcome in 347 patients diagnosed with chronic forms of depression and treated at 9 sites with either Nefazodone, cognitive-behavioral analysis system of psychotherapy (CBASP), or combination therapy. Contrary to our hypotheses, reactance positively predicted treatment outcome in CBASP on 2 of 4 scales. These effects were independent of the therapeutic alliance, which also positively predicted outcome. Reactance did not predict outcome in the groups receiving medication alone or in combination with CBASP. The findings suggest that reactance may be an asset in psychotherapy among chronically depressed individuals and that reactant patients can benefit from directive psychotherapy when therapists flexibly respond to perturbations in the therapeutic relationship. Results support the importance of Aptitude × Treatment interactions in psychotherapy outcome. The direction and significance of such interactions may vary with different forms of psychopathology. This paper demonstrates a technique for controlling the electron emission of an array of field emitting vertically aligned carbon nanofibers (VACNFs). An array of carbon nanofibers (CNF) is to be used as the source of electron beams for lithography purposes. This tool is intended to replace the mask in the conventional photolithography process by controlling their charge emission using the “Dose Control Circuitry” (DCC). The large variation in the charge emitted between CNFs grown in identical conditions forced the controller design to be based on fixed dose rather than on fixed time. Compact digital control logic has been designed for controlling the operation of DCC. This system has been implemented in a 0.5 μm CMOS process. Chandra Sekhar A. Durisety received his B.E. (Hons.) Instrumentation from Birla Institute of Technology and Sciences, Pilani, India in 1997 and his M.S in Electrical Engineering from University of Tennessee, Knoxville in 2002. Since 2003, he has been working towards his Ph.D degree also in Electrical Engineering at Integrated Circuits and Systems Lab (ICASL), University of Tennessee, Knoxville. He joined Wipro Infotech Ltd, Global R & D, Bangalore, India in 1997, where he designed FPGA based IPs for network routers. Since 1999, he was involved in the PCI bridge implementation at CMOS chips Inc, Santa Clara, CA, and the test bench development for Sony’s MP3 player, while at Toshiba America Electronic Components Inc., San Jose, CA. His research interests include multi-stage amplifiers, data converters, circuits in SOI and Floating Gate Devices. Rajagopal Vijayaraghavan received the B.E degree in electronics and communication engineering from Madras University in 1998 and the M.S degree in electrical engineering from the University of Texas, Dallas in 2001.He is currently working towards the Ph.D degree in electrical engineering at the University of Tennessee. His research interest is in the area of CMOS Analog and RF IC design. His current research focuses on LNAs and VCOs using SOI based MESFET devices. Lakshmipriya Seshan was born in Trivandrum, India on April 30, 1979. She received her B.tech in Electronincs & Communication Engg from Kerala University, India in June 2000 and M.S in Electrical Engg from University of Tennessee in 2004. In 2004, she joined Intel Corporation as an Analog Engineer, where she is engaged in the design of low power, high speed analog circuits for various I/O interface topologies. Syed K. Islam received his B.Sc. in Electrical and Electronic Engineering from Bangladesh University of Engineering and Technology (BUET) and M.S. and Ph.D. in Electrical and Systems Engineering from the University of Connecticut. He is presently an Associate Professor in the Department of Electrical and Computer Engineering at the University of Tennessee, Knoxville. Dr. Islam is leading the research efforts of the Analog VLSI and Devices Laboratory at the University of Tennessee. His research interests are design, modeling and fabrication of microelectronic/optoelectronic devices, molecular scale electronics and nanotechnology, biomicroelectronics and monolithic sensors. Dr. Islam has numerous publications in technical journals and conference proceedings in the areas of semiconductors devices and circuits. Benjamin J. Blalock received his B.S. degree in electrical engineering from The University of Tennessee, Knoxville, in 1991 and the M.S. and Ph.D. degrees, also in electrical engineering, from the Georgia Institute of Technology, Atlanta, in 1993 and 1996 respectively. He is currently an Assistant Professor in the Department of Electrical and Computer Engineering at The University of Tennessee where he directs the Integrated Circuits and Systems Laboratory (ICASL). His research focus there includes analog IC design for extreme environments (both wide temperature and radiation immune), multi-gate transistors and circuits on SOI, body-driven circuit techniques for ultra low-voltage analog, mixed-signal/mixed-voltage circuit design for systems-on-a-chip, and bio-microelectronics. Dr. Blalock has co-authored over 60 published refereed papers. He has also worked as an analog IC design consultant for Cypress Semiconductor Corp. and Concorde Microsystems Inc.  相似文献

Silicon thinned to 50 mum and less is flexible allowing the fabrication of flexible and conformable electronics. Two techniques have been developed to achieve this goal using thinned die: die flip chip bonded onto flexible substrates [polyimide and liquid crystal polymer (LCP)] and die flip chip laminated onto LCP films. A key to achieving each of these techniques is the thinning of die to a thickness of 50 mum or thinner. Conventional grinding and polishing can be used to thin to 50 mum. At 50 mum, the active die becomes flexible and must be handled by temporarily bonding it to a holder die for assembly. Both reflow solder and thermocompression assembly methods are used. In the case of solder assembly, underfill is used to reinforce the solder joints. With thermocompression bonding of the die to an LCP substrate, the LCP adheres to the die surface, eliminating the need for underfill.  相似文献

A new high-level noise source was developed specifically for the calibration of Johnson noise power thermometers (JNPT), but it is useful in other applications requiring a stable, wideband, well-defined noise power spectral density. The design concept for the noise source is based on the fact that the open-circuit thermal noise voltage of a feedback resistor, connecting the output to the input of an inverting amplifier, is available at the amplifier output from an equivalent low output impedance caused by the feedback mechanism. The noise power spectral density level at the noise source output is equivalent to the density of the open-circuit thermal noise of a 100-Omega resistor at a temperature of about 64 000 K. The noise source has an output power spectral density that is flat to within 0.1% (0.0043 dB) in the frequency range 1-150 kHz which brackets typical passbands of the signal-processing channels of JNPTs. The noise source was evaluated by comparing its output noise power with the thermal noise power generated by a platinum resistor of known ohmic value at the temperature of the aluminum freezing point (933.52 K). The measured noise power was within 0.06% of the value computed from the fundamental thermal noise equation of Nyquist. The power spectral density varies less than 0.03% for ambient temperature fluctuations in the range 15 degrees -35 degrees C.  相似文献

To investigate the possible role of positron emission tomography (PET) with fluorine-18 fluorodeoxyglucose (FDG) in the prognostic evaluation of primary breast cancer, we studied 86 patients with T1-3 (TNM classification) breast tumours before surgery and compared the tumour FDG uptake, calculated as a standardized uptake value (SUV), with postoperative histopathological findings, steroid hormone receptor status of the tumour, thymidine labelling index (LI) and tissular expression of p53. SUV was significantly higher in infiltrating ductal carcinomas (n = 68; median SUV = 5.6) than in lobular ones (n = 18; median SUV = 3.8), and in grade 3 carcinomas (n = 26; median SUV = 6.2) than in grade 1-2 ones (n = 60; median SUV = 4.9). Moreover, SUV was significantly higher in carcinomas with high levels of p53 (n = 12; median SUV = 9.5) than in those with low levels (n = 48; median SUV = 4.25). By contrast, there was no significant correlation between SUV and the steroid hormone receptor status or LI of tumours. Our data demonstrate that FDG uptake, expressed as SUV, is associated with certain prognostic factors in breast cancer, such as histopathological grading and p53 expression, which can be assessed only by means of postoperative in vitro examinations. Hence, the information provided by FDG-PET is to some extent related to relevant information on tumour biology. The clinical value of these data will have to be confirmed by analysis of the independence of SUV from other prognostic factors by means of a multivariate analysis performed on a larger series of patients with adequate follow-up. If SUV is confirmed as an independent variable, FDG-PET could assume an important role in the determination of appropriate therapeutic strategies for primary breast cancer.  相似文献

An improved procedure for the synthesis of 3 beta-hydroxyandrost-5-ene-7,17-dione, a natural metabolite of dehydroepiandrosterone (DHEA) is described. The synthesis and magnetic resonance spectra of several other related steroids are presented. Feeding dehydroepiandrosterone to rats induces enhanced formation of several liver enzymes among which are mitochondrial sn-glycerol 3-phosphate dehydrogenase (GPDH) and cytosolic malic enzyme. The induction of these two enzymes, that complete a thermogenic system in rat liver, was used as an assay to search for derivatives of DHEA that might be more active than the parent steroid. Activity is retained in steroids that are reduced to the corresponding 17 beta-hydroxy derivative, or hydroxylated at 7 alpha or 7 beta, and is considerably enhanced when the 17-hydroxy or 17-carbonyl steroid is converted to the 7-oxo derivative. Several derivatives of DHEA did not induce the thermogenic enzymes whereas the corresponding 7-oxo compounds did. Both short and long chain acyl esters of DHEA and of 7-oxo-DHEA are active inducers of the liver enzymes when fed to rats. 7-Oxo-DHEA-3-sulfate is as active as 7-oxo-DHEA or its 3-acetyl ester, whereas DHEA-3-sulfate is much less active than DHEA. Among many steroids tested, those possessing a carbonyl group at position 3, a methyl group at 7, a hydroxyl group at positions 1, 2, 4, 11, or 19, or a saturated B ring, with or without a 4-5 double bond, were inactive.  相似文献
