The Treatment of Dissociation in Sexually Abused Children From a Family/Attachment Perspective.

This article outlines an approach to treatment of sexually abused children with dissociative symptoms. Dissociated self-states are seen as competing interpersonal approaches to handling the many emotional sequela of abuse, including anger, fear, and regressive needs. Parents' responses to their sexually abused children, complicated by guilt and their own histories of trauma, can promote dissociative coping in the children as they have difficulty processing their own real feelings of anger, fear, and responsibility. Children and parents may alternatively take victimizer, victim, and rescuer roles, thus mutually reinforcing a dissociative style of coping with these events. This article illustrates how sensitivity to these family dynamics, along with a problem-solving approach to the child's symptoms, can treat dissociative psychopathology in these children. Microwave-Assisted Synthesis of Phenothiazine and Quinoline Derivatives

Application of a dynamic microwave power system in the chemical synthesis of some phenothiazine and quinoline derivatives is described. Heterocyclic ring formation, aromatic nucleophilic substitution and heterocyclic aldehydes/ketones condensation reactions were performed on solid support, or under solvent free reaction conditions. The microwave-assisted Duff formylation of phenothiazine was achieved. Comparison of microwave-assisted synthesis with the conventional synthetic methods demonstrates advantages related to shorter reaction times and in some cases better reaction yields.     

Dissociative symptoms in children

Acute nonfatal erysipelas was diagnosed in 4 sows in a 1,000-sow commercial farrow-to-finish operation in Indiana. Sows were pyrexic, lethargic, lame, and had multiple, 1.3- to 7.6-cm, erythematous rhomboid skin lesions. Outbreak was attributed to failure to properly vaccinate pigs. Further morbidity and mortality were prevented by treatment of clinically affected sows and all pigs in close proximity with procaine penicillin G daily for 3 consecutive days and vaccination of all pigs with questionable vaccination status, using bacterin of killed Erysipelothrix rhusiopathiae. Periodic review of herd management protocols is important to ensure that recommended vaccination schedules are being followed and animals are receiving quality vaccinations. Human error can contribute to many production problems and should be included on the differential diagnoses list.     

The application of structural equation modeling to maternal ratings of twins' behavioral and emotional problems.

The application of structural equation modeling to twin data is used to assess the impact of genetic and environmental factors on children's behavioral and emotional functioning. The models are applied to the maternal ratings of behavior of a subsample of 515 monozygotic and 749 dizygotic juvenile twin pairs, ages 8 through 16 yrs, obtained through mailed questionnaires as part of the Medical College of Virginia Adolescent Behavioral Development Twin Project. The importance of genetic, shared, and specific environmental factors for explaining variation is reported for both externalizing and internalizing behaviors, as well as significant differences in the causes of variation in externalizing behaviors among young boys and girls. The usefulness of applying structural equation models to data on monozygotic and dizygotic twins and the potential implications for addressing clinically relevant questions regarding the causes of psychopathology are discussed. The effects of child sexual abuse: Comment on Rind, Tromovitch, and Bauserman (1998).

Comments on the article by B. Rind, P. Tromovitch, and R. Bauserman (see record 1998-04232-002) which examined the long-term effects of childhood sexual abuse (CSA) by meta-analyzing studies of college students. The authors reported that effects "were neither pervasive nor typically intense" and that "men reacted much less negatively than women" (p. 22) and recommended value-neutral reconceptualization of the CSA construct. The current analysis revealed numerous problems in that study that minimized CSA-adjustment relations, included use of a healthy sample, an inclusive definition of CSA, failure to correct for statistical attenuation, and misreporting of original data. Rind et al.'s study's main conclusions were not supported by the original data. As such, attempts to use their study to argue that an individual has not been harmed by sexual abuse constitute a serious misapplication of its findings. Combinatorial design of chemical-dependent protein switches for controlling intracellular electron transfer

One challenge with controlling electron flow in cells is the lack of biomolecules that directly couple environmental sensing to electron transfer efficiency. To overcome this component limitation, we randomly inserted the ligand binding domain (LBD) from the estrogen receptor (ER) into a 2Fe-2S ferredoxin (Fd) and used a bacterial selection to identify protein variants that support electron transfer in cells. Mapping LBD insertion sites onto structure revealed that Fd tolerates domain insertion adjacent to or within the tetracysteine motif that coordinates the 2Fe-2S metallocluster. With both protein designs, cellular electron transfer (ET) was enhanced by the ER antagonist 4-hydroxytamoxifen, albeit to different extents. One variant arising from ER-LBD insertion within the tetracysteine motif acquired an oxygen-tolerant 2Fe-2S cluster, suggesting that ET is regulated through post-translational ligand binding. These metalloprotein switches are expected to be useful for achieving fast regulation of ET in engineered metabolic pathways and between electroactive bacteria and conductive materials.     

Fluorescence Detection of a Protein‐Bound 2Fe2S Cluster

A fluorescent biosensor is described for 2Fe2S clusters that is composed of green fluorescent protein (GFP) fused to glutaredoxin 2 (Grx2), as illustrated here. 2Fe2S detection is based on the reduction of GFP fluorescence upon the 2Fe2S‐induced dimerization of GFP‐Grx2. This assay is sufficiently sensitive to detect submicromolar changes in 2Fe2S levels, thus making it suitable for high‐throughput measurements of metallocluster degradation and synthesis reactions.
