OBJECTIVE: To evaluate the growth and insulin secretion from microencapsulated beta TC6-F7 cells in vitro and to assess the in vivo function of microencapsulated cells transplanted in rats with steptozotocin (STZ)-induced diabetes. METHOD: Alginate-poly-L-lysine encapsulated beta TC6-F7 cells were exposed to glucose, isobutylmethylxanthine (IBMX) and glucagon-like peptide I (7-36 amide) in a static in vitro challenge. In vivo, 2.5-3.5 x 10(7) encapsulated cells were implanted into diabetic rats. Graft function was evaluated by monitoring blood glucose concentrations and by an intraperitoneal glucose tolerance test. RESULTS: The cell density (number of cells per capsule) of cultured microencapsulated beta TC6-F7 cells increased almost 35-fold over a 55 day observation period to reach a plateau of approximately 3500 cells/capsule. While insulin secretion per capsule remained unchanged over the first 21 days of culture, a 7-fold increase was observed during the last 14 days of the 55 day observation period. Intraperitoneal transplantation of 3.5 x 10(7) encapsulated cells into diabetic rats resulted, within 24 hours, in reversal of hyperglycemia for up to 60 days. Post-transplantation blood glucose concentrations varied between 2 and 4 mM. Glucose clearance rates evaluated by an intraperitoneal glucose tolerance test at 30 days post-transplantation resulted in a markedly flat glucose clearance curve with blood glucose never rising above 4 mM. The glucose challenge of microencapsulated cells recovered 30 days post-transplantation resulted in a 2-fold increase in insulin response at glucose concentrations greater than 5.5 mM as compared to glucose-free media. In addition, immunostaining of recovered grafted tissue for insulin, reveals a strong presence of the peptide within the cell population. CONCLUSIONS: These data demonstrate the potential use of an immunoisolated beta-cell line for the treatment of diabetes.  相似文献

We investigate the effect of dynamic and uniaxial static loading on the wave speeds and rise times of laser generated acoustic waves traveling through a disordered, multilayer aggregate of 2 \(\mu {\mathrm {m}}\) diameter silica microspheres, where the excited dynamic amplitudes are estimated to approach the level of the static overlap between the particles caused by adhesion and externally applied loads. Two cases are studied: a case where the as-fabricated particle network is retained, and a case where the static load has been increased to the point where the aggregate collapses and a rearrangement of the particle network occurs. We observe increases in wave speeds with static loading significantly lower than, and in approximate agreement with, predictions from models based on Hertzian contact mechanics for the pre- and post-collapse states, respectively. The measured rise time of the leading pulse is found to decrease with increasing static load in both cases, which we attribute to decreased scattering and stiffening of the contact network. Finally, we observe an increase in wave speed with increased excitation amplitude that depends on static loading, and whether the system is in the pre- or post-collapse state. The wave speed dependence on amplitude and static load is found to be in qualitative agreement with a one-dimensional discrete model of adhesive spheres, although the observed difference between pre- and post-collapse states is not captured. This investigation, and the approach presented herein, may find use in future studies of the contact mechanics and dynamics of adhesive microgranular systems.  相似文献

The multilevel fast multipole algorithm (MLFMA) is very efficient for solving large-scale electromagnetic scattering problems. However, at low frequencies, or when the discretization is small compared with the wavelength, both the MLFMA and the underlying integral equation formulation typically suffer from a subwavelength breakdown. For the electromagnetic scattering from a homogeneous dielectric object, we obtain a stable and well-conditioned surface integral formulation using a variant of the classical Muumlller formulation and linear basis functions. To overcome the subwavelength breakdown of the MLFMA, we use both propagating and evanescent plane waves to represent the fields. The implementation is based on a combination of the spectral representation of the Green's function and Rokhlin's translation formula. We also present a new interpolation scheme for the evanescent part, which significantly improves the error-controllability of the MLFMA-implementation. This study investigated the role of the endogenous opioid system in maternal and affiliative behavior of group-living rhesus macaque (Macaca mularta) mothers with a history of abusive parenting. 18 mothers received an injection of the opioid antagonist naltrexone or saline for 5 days per wk for the first 4 wks of the infant's life. After treatment, mother-infant pairs were focally observed. Naltrexone did not significantly affect infant abuse or other measures of maternal behavior. Naltrexone increased the amount of grooming received by mothers from other group members and reduced the mothers' rate of displacement activities such as scratching, yawning, and self-grooming. These results concur with previous primate studies in suggesting that opioids mediate the rewarding effects of receiving grooming and affect anxiety-related behaviors. Environmental Fluid Dynamics Code, a numerical estuarine and coastal ocean circulation hydrodynamic and eutrophication model, was used to simulate the distributions of dissolved oxygen (DO), salinity, water temperature, and nutrients in the Caloosahatchee River Estuary. Modeled DO, salinity, and water temperature were in good agreement with field observational data from the Florida Department of Environmental Protection and South Florida Water Management District. Sensitivity analyses identified the effects of river discharge, atmospheric winds, and tidal forcing on the spatial and temporal distributions of DO. Simulation results indicated that vertical mixing due to wind forcing increased the bottom DO concentration. River discharge enhanced stratification in deep locations but propagated vertical mixing in the shallow upper estuary. Finally, tidal forcing heavily influenced bottom layer DO concentrations throughout the whole river estuary.  相似文献

Monthly nutrient enrichment experiments were performed from April through October, 1983, to identify growth limiting nutrients of natural phytoplankton assemblages collected from a station in the Pigeon Bay waters off the north shore of Lake Erie's western basin. Data from these experiments suggest that silica was the major limiting nutrient in April, phosphorus was the major limiting nutrient in May-September, and trace metals were limiting in October. These results show that nutrient enrichment experiments should be performed often enough to account for seasonal changes in the physicochemical environment as well as seasonal succession in phytoplankton. The conclusion that phosphorus was the major limiting nutrient in summer is different from the conclusions of the late 1960s and early 1970s that nitrogen was the major limiting nutrient in the summer in the western basin and suggests a possible shift from nitrogen to phosphorus limitation. Pigeon Bay nutrient trend data show that summer nitrate:soluble reactive phosphorus ratios have increased from approximately 4 in the late 1960s to over 40 in the late 1970s and early 1980s, corroborating that a shift has probably occurred. This shift in nutrient limitation is probably due to reduced bioavailable phosphorus loadings and increased nitrate loadings to the western basin. The southern portion of the western basin is physicochemically different from the northern portion and may not have responded in the same manner.  相似文献

Three new 10-hydroxy fatty acids, all optically active, have been prepared by the anaerobic microbiological hydration of acis-9 double bond. Substrates that formed these new hydroxy fatty acids are linoleic, linolenic, and ricinoleic acids. The hydroxyl group has the D configuration and the methyl esters are levorotatory. Infrared, mass spectral, specific rotation and ultraviolet data on these compounds were determined. There was no migration of the unreated double bonds at C12 and C15 in linoleic or linolenic acids. The presence of a double bond in the 10-hydroxy fatty acids significantly increased the optical rotation of the methyl esters. The hydratase enzyme showed unusual specificity among Δ9 unsaturated acids. While it hydrates methylene interrupted and hydroxy unsaturated acids, it failed to hydrate either 9-decenoic, 12,13-epoxy- or 12-keto-cis-9-octadecenoic acids or sterculic acid. Genetic studies of the nematode Caenorhabditis elegans (C. elegans) have identified several important components of the cell death pathway, most notably CED-3, CED-4, and CED-9. CED-4 directly interacts with the Bcl-2 homologue CED-9 (or the mammalian Bcl-2 family member Bcl-xL) and the caspase CED-3 (or the mammalian caspases ICE and FLICE). This trimolecular complex of CED-4, CED-3, and CED-9 is functional in that CED-9 inhibits CED-4 from activating CED-3 and thereby inhibits apoptosis in heterologous systems. The E1B 19,000-molecular weight protein (E1B 19K) is a potent apoptosis inhibitor and the adenovirus homologue of Bcl-2-related apoptosis inhibitors. Since E1B 19K and Bcl-xL have functional similarity, we determined if E1B 19K interacts with CED-4 and regulates CED-4-dependent caspase activation. Binding analysis indicated that E1B 19K interacts with CED-4 in a Saccharomyces cerevisiae two-hybrid assay, in vitro, and in mammalian cell lysates. The subcellular localization pattern of CED-4 was dramatically changed by E1B 19K, supporting the theory of a functional interaction between CED-4 and E1B 19K. Whereas expression of CED-4 alone could not induce cell death, coexpression of CED-4 and FLICE augmented cell death induction by FLICE, which was blocked by expression of E1B 19K. Even though E1B 19K did not prevent FLICE-induced apoptosis, it did inhibit CED-4-dependent, FLICE-mediated apoptosis, which suggested that CED-4 was required for E1B 19K to block FLICE activation. Thus, E1B 19K functions through interacting with CED-4, and presumably a mammalian homologue of CED-4, to inhibit caspase activation and apoptosis.  相似文献
