Tested the hypothesis that Ss with severe mental illnesses would achieve better vocational outcomes with an accelerated approach to supported employment (AASE), as compared to gradual approaches (GA) with prevocational training. 86 Ss (mean age 35.1 yrs) with a diagnosis of a serious mental illness, were randomly assigned to either the AASE, or the GA, which included a minimum of 4 mo prevocational training. Data were obtained on indicators of vocational outcomes over 2 yrs, and for a limited number of Ss (n=36), during the 4th yr of inception into the program. Initially, only 5% of Ss preferred prevocational training. After 1 yr, AASE Ss showed better outcomes for a range of indicators, including achievement of competitive employment, duration of employment and mean earnings. During the 4th yr, 59% of these Ss were competitively employed, as compared to only 6% of GA Ss. Rehabilitation is more effective using AASE than GA. Certain tasks, such as formal program development and theorem proving, fundamentally rely upon the manipulation of higher-order objects such as functions and predicates. Computing tools intended to assist in performing these tasks are at present inadequate in both the amount of knowledge they contain (i.e., the level of support they provide) and in their ability to learn (i.e., their capacity to enhance that support over time). The application of a relevant machine learning technique—explanation-based generalization (EBG)—has thus far been limited to first-order problem representations. We extend EBG to generalize higher-order values, thereby enabling its application to higher-order problem encodings.Logic programming provides a uniform framework in which all aspects of explanation-based generalization and learning may be defined and carried out. First-order Horn logics (e.g., Prolog) are not, however, well suited to higher-order applications. Instead, we employ Prolog, a higher-order logic programming language, as our basic framework for realizing higher-order EBG. In order to capture the distinction between domain theory and training instance upon which EBG relies, we extend Prolog with the necessity operator of modal logic. We develop a meta-interpreter realizing EBG for the extended language, Prolog, and provide examples of higher-order EBG.  相似文献

Developed a fidelity index of program implementation for assertive community treatment (ACT). In Study 1, 20 experts rated the importance of 73 elements proposed as critical ACT ingredients, also indicating ideal model specifications for elements. Agreement among experts on ratings of importance was high (intraclass r?=?.98). In Study 2, a 17-item subset of the expert-identified critical ingredients was used to construct a fidelity index with 3 subscales: Staffing, Organization, and Service. Internal consistencies ranged from .50 to .72, with a .81 reliability for the total scale. Fidelity was linearly related to program "generations," suggesting "program drift." In 18 ACT programs, fidelity also was associated with measures of reduction in days in psychiatric hospitals. The correlation was significant for the total scale and for the Organization and Staffing subscales but not for the Service subscale. The cytochrome P450 monooxygenase CYP106A2 from Bacillus megaterium ATCC 13368 catalyzes hydroxylations of a variety of 3-oxo-Δ(4) -steroids such as progesterone and deoxycorticosterone (DOC), mainly in the 15β-position. We combined a high-throughput screening and a rational approach for identifying new substrates of CYP106A2. The diterpene resin acid abietic acid was found to be a substrate and was docked into the active site of a CYP106A2 homology model to provide further inside into the structural basis of the regioselectivity of hydroxylation. The products of the hydroxylation reaction were analyzed by HPLC and the V(max) and K(m) values were calculated. The corresponding reaction products were analyzed by NMR spectroscopy and identified as 12α- and 12β-hydroxyabietic acid. CYP106A2 was therefore identified as the first reported bacterial cytochrome P450 diterpene hydroxylase. Furthermore, an effective whole-cell catalyst for the selective allylic 12α- and 12β-hydroxylation was applied to produce the hydroxylated products.  相似文献
