A strip-based biosensor using the enzyme-linked immunosorbent assay technique was fabricated to detect Escherichia coli O157:H7. Two types of antibody specified to E. coli O157:H7 were used to form sandwich-binding complexes. To fabricate an immuno-strip, capture antibody (monoclonal antibody) was immobilized onto signal generation pad and polyclonal antibody conjugated with horseradish peroxidase (HRP) was utilized as detection antibody. Four different functional membranes have been used to fabricate immuno-chromatographic assay strip. A sample application pad was a glass fiber membrane pre-treated with polyvinyl alcohol. A conjugate release pad was fabricated using a glass membrane. A signal generation pad was made on nitrocellulose membrane. Finally, a cellulose membrane was used as an absorption pad. Under optimal conditions of analysis, a color signal in proportion to the E. coli O157:H7 concentration was measured using a detector. The measurement range was 1.8x10(3)-1.8x10(8)CFU/mL.  相似文献

The inhibitory effects of lipid accumulation on ethanol extract from stem bark of Japanese horse chestnut (JHC) were evaluated. Exposure to JHC extract (10–100 μg/mL) for a 72 h incubation period did not alter cell viability compared to the untreated control. JHC extract, with concentrations of 25, 50, and 100 μg/mL, inhibited lipid accumulation in 3T3-L1 adipocytes in a dose dependent manner. The expression of PPARγ and C/EBPα, important adpogenic key markers was significantly reduced when JHC extract was added to cells for 8 days compared with the untreated control group. These results suggest that JHC extract might be a potential therapeutic agent as a natural anti-obesity material.  相似文献

The development of new electrocatalysts for electrochemical oxygen reduction to replace expensive and rare platinum‐based catalysts is an important issue in energy storage and conversion research. In this context, conductive and porous metal–organic frameworks (MOFs) are considered promising materials for the oxygen reduction reaction (ORR) due to not only their high surface area and well‐developed pores but also versatile structural features and chemical compositions. Herein, the preparation of bimetallic conductive 2D MOFs (CoxNiy‐CATs) are reported for use as catalysts in the ORR. The ratio of the two metal ions (Co2+ and Ni2+) in the bimetallic CoxNiy‐CATs is rationally controlled to determine the optimal composition of CoxNiy‐CAT for efficient performance in the ORR. Indeed, bimetallic MOFs display enhanced ORR activity compared to their monometallic counterparts (Co‐CAT or Ni‐CAT). During the ORR, bimetallic CoxNiy‐CATs retain an advantageous characteristic of Co‐CAT in relation to its high diffusion‐limiting current density, as well as a key advantage of Ni‐CAT in relation to its high onset potential. Moreover, the ORR‐active bimetallic CoxNiy‐CAT with excellent ORR activity is prepared at a large scale via a convenient method using a ball‐mill reactor.  相似文献

This study investigated the effects of soy bean extract fermented by Bacillus subtilis MORI (BTD-1) on blood glucose, hemoglobin A1c (HbA1c), plasma insulin, and pancreatic β islets in db/db mice. The BTD-1 (500 mg/kg) group showed significantly lower fasting blood glucose level (p<0.01) and postprandial 2 h blood glucose level (p<0.01) compared with the db control group. The BTD-1 (500 mg/kg) group showed significantly lower HbA1c level compared with the db control group (p<0.01). After 8 weeks of BTD-1 administration, the pancreatic islet architecture was preserved and the immunofluorescent intensities of insulin in BTD-1 (500mg/kg) group apparently increased compared to in the db control group. Plasma insulin levels were found to be significantly higher in the BTD-1 (500 mg/kg) group than in the db control group (p<0.05). In summary, our results suggest that BTD-1 has an anti-diabetes effect in a non-insulin dependent diabetes mellitus mouse model.  相似文献

The immuno-strip biosensor was developed by combination of immuno-chromatography assay and sandwich enzyme linked immunosorbent assay (ELISA) techniques to detect enrofloxacin, a widely used broad-spectrum antibiotic. A proposed analytical system, consisted of immuno-reaction step and enzyme reaction step, has several advantages such as a very cheap manufacturing cost, a short analysis time (approximately 20 min), an easy-to-use, a high sensitivity, and a qualitatively confirmation by bare eyes. Enzyme reaction step in a proposed analytical tool could enhance the signal of immunosensor, resulting in the high sensitivity. A proposed system was successfully applied to detect enrofloxacin in a range of 100–10000 ppb. The lower detection limit was 100 ppb, which is the maximum residue limit of enrofloxacin in animal product.  相似文献
