Recent progress in the de novo design of self-assembling peptides has enabled the construction of peptide-based viral capsids. Previously, we demonstrated that 24-mer β-annulus peptides from tomato bushy stunt virus spontaneously self-assemble into an artificial viral capsid. Here we propose to use the artificial viral capsid through the self-assembly of β-annulus peptide as a simple model to analyze the effect of molecular crowding environment on the formation process of viral capsid. Artificial viral capsids formed by co-assembly of fluorescent-labelled and unmodified β-annulus peptides in dilute aqueous solutions and under molecular crowding conditions were analyzed using fluorescence correlation spectroscopy (FCS). The apparent particle size and the dissociation constant (Kd) of the assemblies decreased with increasing concentration of the molecular crowding agent, i.e., polyethylene glycol (PEG). This is the first successful in situ analysis of self-assembling process of artificial viral capsid under molecular crowding conditions.  相似文献

This paper reports results of full scale testing to further explore potential GCL damage mechanisms in earth dam retrofit applications in seismically active areas; in particular, to a) investigate whether shear displacements could reduce the magnitude of GCL panel overlap during earthquake shaking; b) explore the influence of gravel particles on GCL thickness at localised point of contact; and c) observe the consequences of an accidental exposure of an uncovered GCL to short duration rainfall in terms of moisture content and effects during subsequent compaction. The results of these experiments indicate that even under severe shaking no movements were detected at the GCL panel overlap. Whereas gravel particles were observed to locally reduce the thickness of the GCL to 2.2 mm, no plowing of the particle into the GCL occurred due to a lack of shear displacement at the interface, resulting in no localised internal erosion through the barrier. Furthermore, hydration of GCL panels during construction due to surface wetting was observed to result in a state of hydration less than its post-construction state. These results indicate that although each of the three GCL damage mechanisms cannot be ruled out to ever be relevant in practice, the performance of the GCL retrofitted earth dam tested was satisfactory under even severe Level 2 earthquake shaking, and suggests that the retrofitting of small earth dams with GCLs is a promising strategy to improve their static and seismic resistance.  相似文献

ABSTRACT

It is important to perform neutron transport simulations with accurate nuclear data in the neutronics design of a fusion reactor. However, absolute values of large-angle scattering cross sections vary among nuclear data libraries even for well-examined nuclide of iron. Benchmark experiments focusing on large-angle scattering cross sections were thus performed to confirm the correctness of nuclear data libraries. The series benchmark experiments were performed at a DT neutron source facility, OKTAVIAN of Osaka University, Japan, by the unique experimental system established by the authors’ group, which can extract only the contribution of large-angle scattering reactions. This system consists of two shadow bars, target plate (iron), and neutron detector (niobium). Two types of shadow bars were used and four irradiations were conducted for one experiment, so that contribution of room-return neutrons was effectively removed and only large-angle scattering neutrons were extracted from the measured four Nb reaction rates. The obtained experimental results were compared with calculations for five nuclear data libraries including JENDL-4.0, JEFF.-3.3, FENDL-3.1, ENDF/B- VII, and recently released ENDF/B-VIII. It was found from the comparison that ENDF/B-VIII showed the best result, though ENDF/B-VII showed overestimation and others are in large underestimation at 14 MeV.  相似文献

Because Japanese cedar pollen (JCP) contains beta-1,3-d-glucan (BG), there is concern that its lingering presence in the atmosphere, especially during its scattering period, may cause false positives in the factor-G-based Limulus amebocyte lysate (LAL) assay used to test for deep mycosis (i.e., G-test). Hence, we examined whether the LAL assay would react positively with substances contained in JCP by using the G-test to measure JCP particles and extracts. BG was purified from the JCP extract on a BG-specific affinity column, and the percentage extractability was measured using three different BG-specific quantitative methods. The G-test detected 0.4 pg BG in a single JCP particle and 10 fg from a single particle in the extract. The percentage extractability of JCP-derived BG was not significantly different among the three quantitative methods. As the JCP particles should technically have been removed during serum separation, they should be less likely to be a direct false-positive factor. However, given that the LAL-assay-positive substances in the JCP extract were not distinguishable by the three BG-specific quantitative methods, we conclude that they may cause the background to rise. Therefore, in Japan false positives arising from JCP contamination should be considered when testing patients for deep mycosis.  相似文献

We focused on the work function of metals as an index of ion binding property between metals and hydrogen and analyzed the relation between the standard entropy change of metal hydrides and the work function of metals (alloys). The standard entropy change of saline hydrides and metallic hydrides roughly increased with the work function. It was indicated that hydrogen storage alloys consist of metal elements A and B whose work functions are small and large, respectively. The standard entropy changes of alloy-based metal hydrides with different composition ratio ΔS0 linearly increased with the work functions of the alloys Wc. Wc also linearly increased with the bulk modulus Bc. Then, it was found that ΔS0 was proportional to the standard heat of formation ΔH0 because dWc/dBc, dΔS0/dWc and dΔH0/dBc are constant.  相似文献

老人的健康与建筑室内环境存在重要联系,为了给适老建筑的暖通系统优化设计提供理论依据,有必要探究室内空气调节设备与老人健康的关系。对广州市104位本地老年人进行了问卷调查,收集了老年人的基本健康信息和家庭使用空气调节设备的情况,包括制冷、除湿、采暖和空气净化4种设备及其使用时长,使用习惯。收集到的信息用SPSS20.0进行了统计分析。结果包括广州市老年人家中四种设备拥有率、设备平均使用龄、设备使用时间以及常见日均使用时长;所调查的老年人健康比例达到55.8%。患病老人中心血管疾病的比例最高,其余为风湿性疾病和关节炎等疾病。相关性分析表明,广州地区夏天家庭采取制冷措施的老人患病可能性更小;制冷设备常见日均使用时长与身处空调区域时长的比值越大,老人患病的可能性越小。  相似文献
