Tumor-specific enhanced delivery of chemotherapeutics and modulators to tumor cells and activated pancreatic stellate cells (aPSCs), respectively, represents safer and more effective therapy for pancreatic cancer. Herein, a membrane type 1-matrix metalloproteinase (MT1-MMP)-cleavable spacer is used to assemble low-density cRGDfK onto thermosensitive liposomes loaded with phosphorylated calcipotriol (PCAL) and doxorubicin (DOX), yielding MR-T-PD. The liposome-linked cRGDfK prodrug on MR-T-PD surface is first activated by MT1-MMP, which is selectively expressed on tumor endothelial cells, to release cRGDfK. The free cRGDfK specifically promotes tumor angiogenesis, leading to 3.4-fold higher accumulation and a wider distribution of MR-T-PD in tumors. Furthermore, MR-T-PD rapidly releases PCAL and DOX into the interstitium under heat treatment. The released DOX enters tumor cells to induce apoptosis, whereas the PCAL prodrug is converted to CAL by alkaline phosphatase on the surface of aPSCs; CAL can then enter aPSCs to induce quiescence and promote the antitumor effect of DOX. Finally, by enhancing the exposure of DOX and CAL to tumor cells and aPSCs, respectively, in a tumor-specific manner, MR-T-PD exerts superior efficacy (a 5.9-fold decrease in tumor weight) without causing additional side effects. Overall, this prodrug-based smart liposome system represents a promising paradigm for pancreatic cancer therapy.  相似文献

Fullerenes are candidates for theranostic applications because of their high photodynamic activity and intrinsic multimodal imaging contrast. However, fullerenes suffer from low solubility in aqueous media, poor biocompatibility, cell toxicity, and a tendency to aggregate. C70@lysozyme is introduced herein as a novel bioconjugate that is harmless to a cellular environment, yet is also photoactive and has excellent optical and optoacoustic contrast for tracking cellular uptake and intracellular localization. The formation, water-solubility, photoactivity, and unperturbed structure of C70@lysozyme are confirmed using UV-visible and 2D 1H, 15N NMR spectroscopy. The excellent imaging contrast of C70@lysozyme in optoacoustic and third harmonic generation microscopy is exploited to monitor its uptake in HeLa cells and lysosomal trafficking. Last, the photoactivity of C70@lysozyme and its ability to initiate cell death by means of singlet oxygen (1O2) production upon exposure to low levels of white light irradiation is demonstrated. This study introduces C70@lysozyme and other fullerene-protein conjugates as potential candidates for theranostic applications.  相似文献

5G蜂窝网络发展迅猛,其覆盖面积将逐渐增大,因此使用5G蜂窝网络进行定位是有研究潜力的研究方向。本文提出一种新的深度学习技术来实现高效、高精度和低占用的定位,以代替传统指纹定位过程中繁重的指纹库生成以及距离计算。该方法建立了一个特殊的卷积神经网络,并根据5G天线信号的接收信号强度指示、相位和到达角等特征量,选择合适的输入数据格式构造样本组建训练集,对该卷积神经网络进行训练。训练得到的卷积神经网络可以替代指纹定位中的庞大指纹库,非常有利于直接在5G移动设备端实现定位。虽然卷积神经网络在训练过程中需要大量时间,但在训练完毕后直接进行分类定位的速度非常快,可以保障定位实现的实时性。本文所实现的卷积神经网络权重与偏置所占内存不到0.5 MB,且能够在实际应用环境中以95%的定位准确率以及0.1 m的平均定位精度实现高精度定位。  相似文献

Due to the demand of miniaturization and integration for ceramic capacitors in electronic components market, TiO2-based ceramics with colossal permittivity has become a research hotspot in recent years. In this work, we report that Ag+/Nb5+ co-doped (Ag1/4Nb3/4)xTi1−xO2 (ANTOx) ceramics with colossal permittivity over a wide frequency and temperature range were successfully prepared by a traditional solid–state method. Notably, compositions of ANTO0.005 and ANTO0.01 respectively exhibit both low dielectric loss (0.040 and 0.050 at 1 kHz), high dielectric permittivity (9.2 × 103 and 1.6 × 104 at 1 kHz), and good thermal stability, which satisfy the requirements for the temperature range of application of X9R and X8R ceramic capacitors, respectively. The origin of the dielectric behavior was attributed to five dielectric relaxation phenomena, i.e., localized carriers' hopping, electron–pinned defect–dipoles, interfacial polarization, and oxygen vacancies ionization and diffusion, as suggested by dielectric temperature spectra and valence state analysis via XPS; wherein, electron-pinned defect–dipoles and internal barrier layer capacitance are believed to be the main causes for the giant dielectric permittivity in ANTOx ceramics.  相似文献

建立了具有两端轴承基座松动故障的拉杆转子模型(松动轴承基座质量称为松动质量块,松动轴承基座与地面连接的刚度称为松动刚度),并推导了其运动微分方程.基于分离变量法和Sturm-Liouville理论得到了有限长滑动轴承油膜力近似解析解,运用Newmark法分析了该转子系统的非线性动力学行为.首先,将无故障转子、一侧轴承基座松动和两侧轴承基座松动转子系统动力学进行比较,结果显示存在松动故障的转子具有更加复杂的动力学行为,而且存在松动故障的转子振幅比无故障转子的振幅大.其次,对两端轴承基座松动情况进行了分析,结果表明转子系统存在着周期、周期三、周期五、周期六、周期七、准周期和混沌等运动形式.同时发现,随着转子转速升高,松动质量块的振动幅度也随之增大.当两端松动质量和松动刚度不同时,松动质量较大或者松动刚度较小的一侧,转子的振幅和松动质量块的振幅较大,随着两侧松动质量和松动刚度差值的增加,两侧转子的振幅和松动质量块的振幅的差值也随之增加.  相似文献

Lithium (Li) metal, as an appealing candidate for the next-generation of high-energy-density batteries, is plagued by its safety issue mainly caused by uncontrolled dendrite growth and infinite volume expansion. Developing new materials that can improve the performance of Li-metal anode is one of the urgent tasks. Herein, a new MXene derivative containing pure rutile TiO2 and N-doped carbon prepared by heat-treating MXene under a mixing gas, exhibiting high chemical activity in molten Li, is reported. The lithiation MXene derivative with a hybrid of LiTiO2-Li3N-C and Li offers outstanding electrochemical properties. The symmetrical cell assembling lithiation MXene derivative hybrid anode exhibits an ultra-long cycle lifespan of 2000 h with an overpotential of ≈30 mV at 1 mA cm−2, which overwhelms Li-based anodes reported so far. Additionally, long-term operations of 34, 350, and 500 h at 10 mA cm−2 can be achieved in symmetrical cells at temperatures of −10, 25, and 50 °C, respectively. Both experimental tests and density functional theory calculations confirm that the LiTiO2-Li3N-C skeleton serves as a promising host for Li infusion by alleviating volume variation. Simultaneously, the superlithiophilic interphase of Li3N guides Li deposition along the LiTiO2-Li3N-C skeleton to avoid dendrite growth.  相似文献

Poly(l ‐lactic acid) (PLLA) is a biodegradable and biocompatible thermoplastic polyester produced from renewable sources, widely used for biomedical devices, in food packaging and in agriculture. It is a semicrystalline polymer, and as such its properties are strongly affected by the developed semicrystalline morphology. As a function of the crystallization temperature, PLLA can form different crystal modifications, namely α′‐crystals below about 120 °C and α‐crystals at higher temperatures. The α′ modification is therefore of special importance as it may be the preferred polymorph developing at processing‐relevant conditions. It is a metastable modification which typically transforms into the more stable α‐crystals on annealing at elevated temperature. The structure, kinetics of formation and thermodynamics of α′‐ and α‐crystals of PLLA are reviewed in this contribution, together with the effect of α′‐/α‐crystal polymorphism on the properties of PLLA. © 2018 Society of Chemical Industry  相似文献

In this study, we developed a unique defect healing method for 3D printed ceramic compact via cold isostatic pressing (CIP) after debinding, and typical features of interlayer interface defects of 3D-printed zirconia compact were characterized and found to be reduced significantly. The characteristic sintering kinetics window and microstructure evolution of the healed sintered bodies were systematically investigated, which was found to be quite different from conventional shaping methods. The three sintering stages are probed by their feature microstructure details such as the mechanically flattening surface at the early sintering stage, the heterogeneous microstructure and high porosity in the interlayer interface region at the middle stage, and the slightly ripple-like structural features combined with the healed interlayer defects at the final stage. The evolution of the pore structure of the healed 3D printed bodies were traced and the mechanical properties such as the Young's modulus, hardness, and fracture toughness were measured to understand the significance of the heal effect.  相似文献
