A pattern net assisted mapping artificial neural network (PAMANN) model for estimation of parameters in problem with large data (1300 × 121 matrix size) is reported. A pattern net-based multilayer perceptron neural network (MLPNN) model for clustering the data, followed by mapping MLPNN model for mapping the target with the input, is developed as PAMANN model. A heat transfer problem with combined mode conduction and radiation in porous medium is solved numerically, and is called direct model. In the inverse model, a PAMANN model is developed by using data generated through the direct model. The PAMANN model is able to estimate two parameters (extinction coefficient β and convective coupling P2) after taking temperature profile as input. The model is tested for different number of neurons in hidden layer, and different levels of noise in input data. Twelve different algorithms are explored in training of mapping MLPNN, and compared for performance. Levenberg–Marquardt algorithm is found to estimate the parameters with high accuracy, but took high CPU time. Bayesian regularization is found to consume very high CPU time with moderate accuracy in estimation of parameters. Variations in hidden layer neuron number and noise in input data, were done to analyze the performance of mapping MLPNN with different training algorithms. Algorithms O-Step Secant, conjugate gradient with Polak-Ribiére updates, and conjugate gradient with Fletcher-Reeves updates are able to handle all variations of noise and number of neurons in hidden layer, with good accuracy of estimation and low CPU time consumption. Under high computational resource LM algorithm can be used for all cases. Up to 0.99132 value of regression coefficient is obtained in mapping MLPNN model with 15 hidden neurons, indicating the high accuracy of the model. With the help of PAMANN model, highly accurate (absolute error 1.78%) estimation of parameters is obtained. The model can handle upto 1% noise in input data, while giving accurate results.  相似文献

Phosphodiesterases (PDEs) hydrolyze cyclic nucleotides to modulate multiple signaling events in cells. PDEs are recognized to actively associate with cyclic nucleotide receptors (protein kinases, PKs) in larger macromolecular assemblies referred to as signalosomes. Complexation of PDEs with PKs generates an expanded active site that enhances PDE activity. This facilitates signalosome-associated PDEs to preferentially catalyze active hydrolysis of cyclic nucleotides bound to PKs and aid in signal termination. PDEs are important drug targets, and current strategies for inhibitor discovery are based entirely on targeting conserved PDE catalytic domains. This often results in inhibitors with cross-reactivity amongst closely related PDEs and attendant unwanted side effects. Here, our approach targeted PDE–PK complexes as they would occur in signalosomes, thereby offering greater specificity. Our developed fluorescence polarization assay was adapted to identify inhibitors that block cyclic nucleotide pockets in PDE–PK complexes in one mode and disrupt protein-protein interactions between PDEs and PKs in a second mode. We tested this approach with three different systems—cAMP-specific PDE8–PKAR, cGMP-specific PDE5–PKG, and dual-specificity RegA–RD complexes—and ranked inhibitors according to their inhibition potency. Targeting PDE–PK complexes offers biochemical tools for describing the exquisite specificity of cyclic nucleotide signaling networks in cells.  相似文献

The objective of the research study was to develop and characterize a biodegradable, thermo and pH dual responsive Oxaliplatin-loaded chitosan-graft-poly-N-isopropylacrylamide (CS-g-PNIPAAm) co-polymeric nanoparticles as a tumor-targeting drug delivery system. CS-g-PNIPAAm co-polymers were synthesized, characterized and optimized its thermo and pH responsive properties for tumor microenvironment conditions. Optimized co-polymer could be efficiently loaded with Oxaliplatin in nanoparticle form, evaluated for their morphology (TEM), particle size, zeta potential, loading efficiency and drug content. In vitro drug release study at tumor microenvironment and physiological pH and temperature conditions. The in vitro drug release was optimal at above lower critical solution temperature (LCST) and tumor microenvironment pH when compared to physiological pH & temperature. MTT assay and fluorescence microscopic study showed that drug release and cell uptake was significantly enhanced in tumor microenvironment. In conclusion, the obtained nanoparticles appeared to be of great promise in tumor targeted drug delivery of oxaliplatin.  相似文献

ABSTRACT

The need for development of a more efficient IC engine is always on the rise which can be related to oil being a non-renewable resource and its increasing carbon foot print causing global warming. In addition to the CO2, there are more toxic pollutants such as NOx, CO and HC due to improper combustion. These demands have paved way for development of alternative fuel engines such as bio-diesel, bio-ethanol and hybrids. Still these engines need to optimise their combustion to get the best of both worlds. And for the purpose of optimisation there must be a means to measure and estimate the combustion characteristics. Thus analysing the combustion noise gives some insight into the performance of the engine.  相似文献

Microbial attachment and colonization on separation membranes lead to biofilm formation. Some isolates within the biofilm microflora acquire greater resistance to the chemical cleaning protocols on prolonged use of membranes. It is thus likely that the constitutive microflora might compete with each other and result in certain species emerging as predominant, especially within older biofilms. To understand the microbial interactions within biofilms, the emergence of predominance was studied in the current investigation. An 18-mo-old reverse osmosis membrane was procured from a whey processing plant. The membrane pieces (2.54 × 2.54 cm2) were neutralized by dipping in Letheen broth. The resuscitation step was done in tryptic soy broth (TSB) at 37°C, followed by plating on tryptic soy agar (TSA) to recover the constitutive microflora. Distinct colonies of isolates were further identified using MALDI-TOF as Bacillus licheniformis, Exiguobacterium aurantiacum, Acinetobacter radioresistens, Bacillus subtilis (rpoB sequencing), and 1 unidentified species each of Exiguobacterium and Bacillus. Further, the competitive exclusion study helped establish the emergence of predominance using a co-culturing technique. Fifteen combinations (of 2 isolates each) were prepared from the isolates. Pure cultures of the respective isolates were spiked in a ratio of 1:1 in TSB and incubated at 37°C for 24 h, followed by plating on TSA. The enumerated colonies were distinguished based on colony morphology, Gram staining, and MALDI-TOF to identify the type of the isolate. Plate counts of B. subtilis emerged as predominant with mean log counts of 7.22 ± 0.22 cfu/mL. The predominance of B. subtilis was also validated using the process of natural selection in a multispecies growth environment. In this instance, the TSB culture with overnight-incubated membrane piece (with mixed-species biofilm) at 37°C for 12 h was inoculated in fresh TSB and incubated for the second cycle. Overall, 5 such sequential broth-culture incubation cycles were carried out, followed by pour plating on TSA plates, at the end of each cycle. The isolates obtained were identified using a similar methodology as mentioned above. The fifth subsequent transfer depicted the presence of only 1 B. subtilis isolate on plating, thereby validating its predominance under the conditions of the experiment.  相似文献

Deep mixing columns are commonly employed for soft ground improvement. However, the diameter of a single conventional column is a constant and the area replacement ratio does not vary with depth. Hence, the conventional column is not the ideal solution for multi-layered soft grounds, where different layers have remarkably different soil properties. Accordingly, this study proposes a better solution, which is the variable-diameter deep mixing column with a large diameter in the soil layer having high compressibility and a small diameter in the soil layer having relatively low compressibility. In this study, small-scale laboratory model tests were firstly employed to compare the performance of two-layered soft grounds improved by a variable-diameter column and a conventional column. The additional vertical stress in the soil and the column, the excess pore water pressure, and the ground settlement were analyzed. Then, a field application of variable-diameter columns for multi-layered soft ground improvement was presented; the design considerations, column installation, and monitored settlement were introduced and analyzed. The results indicated that the additional stress in the soil and the column in the highly compressible soil layer were much lower in the variable-diameter column-improved ground than in the conventional column-improved ground. Consequently, the variable-diameter column-improved ground yielded less total settlement and less post-construction settlement compared to the conventional column-improved ground.  相似文献

Doxorubicin is a highly effective chemotherapy agent used to treat many common malignancies. However, its use is limited by cardiotoxicity, and cumulative doses exponentially increase the risk of heart failure. To identify novel heart failure treatment targets, a zebrafish model of doxorubicin-induced cardiomyopathy was previously established for small-molecule screening. Using this model, several small molecules that prevent doxorubicin-induced cardiotoxicity both in zebrafish and in mouse models have previously been identified. In this study, exploration of doxorubicin cardiotoxicity is expanded by screening 2271 small molecules from a proprietary, target-annotated tool compound collection. It is found that 120 small molecules can prevent doxorubicin-induced cardiotoxicity, including 7 highly effective compounds. Of these, all seven exhibited inhibitory activity towards cytochrome P450 family 1 (CYP1). These results are consistent with previous findings, in which visnagin, a CYP1 inhibitor, also prevents doxorubicin-induced cardiotoxicity. Importantly, genetic mutation of cyp1a protected zebrafish against doxorubicin-induced cardiotoxicity phenotypes. Together, these results provide strong evidence that CYP1 is an important contributor to doxorubicin-induced cardiotoxicity and highlight the CYP1 pathway as a candidate therapeutic target for clinical cardioprotection.  相似文献

Laser ablation of high-temperature ceramic coatings results in thermal residual stresses due to which the coatings fail by cracking and debonding. Hence, the measurement of such residual stresses during laser ablation process holds utmost importance from the view of performance of coatings in extreme conditions. The present research aims at investigating the effect of laser parameters such as laser pulse energy, scanning speed and line spacing on thermal residual stresses induced in tantalum carbide-coated graphite substrates. Residual stresses were measured using micro-Raman spectroscopy and correlated with Raman peak shifts. Transient thermal analysis was performed using COMSOL Multiphysics to model the single ablated track and residual stresses were reported at low, moderate and high pulse energy regimes. The results showed that the initial laser conditions caused higher tensile residual stresses. Moderate pulse energy regime comprised higher compressive residual stresses due to off centre overlapping of the laser pulses. Higher pulse energy (250 μJ), higher scanning speed (1000 mm/s) and moderate line spacing (20 μm) caused accumulation of tensile residual stresses during the final stage of laser ablation. The deviation of experimental residual stresses from COMSOL numerical model was attributed to unaccounted additional stresses induced during thermal spraying process and deformation potentials in the numerical model.  相似文献
