Escherichia coli O157 strains have emerged as important human enteric pathogens. Strains that express the O-antigen 157 are commonly associated with severe clinical manifestations, including bloody diarrhea, hemorrhagic colitis, and hemolytic uremic syndrome. E. coli O157 strains may be transmitted in a variety of ways, including food, water and person-to-person or animal-to-person contact. Fecal contamination is one source of environmental contamination and is responsible for the presence of these pathogens in the environment. We used a specific and sensitive PCR assay based on the rfbE gene to detect low levels of these pathogens in wastewater. The set of primers used was designed to amplify an intragenic segment of the rfbE gene. The amplification assay detected 200 CFU of E. coli O157 in pure water. The prevalence of E. coli O157 in the effluents of 44 wastewater treatment plants was determined (7%).  相似文献

研发了一种新型短程硝化反硝化工艺——ANITATMShunt,它通过特殊的自控系统来控制N2O的释放。采用500 L的SBR中试装置处理消化污泥脱水上清液,经过18个月的稳定运行表明:通过短程硝化反硝化途径可以实现90%的脱氮率,并且释放的N2O不足总脱氮量的0.7%。将通过pH值、温度和在线监测的NO-2-N浓度实时计算的亚硝酸浓度与亚硝酸浓度设定值进行比对,以便对曝气过程进行调控,从而抑制了N2O的释放并实现了对SBR短程硝化反硝化工艺的自动控制。同时证实了在低溶解氧条件下,由氨氧化菌(AOB)在短程硝化反硝化过程中产生的N2O并非与高亚硝酸盐浓度有直接关系,而是与游离亚硝酸浓度有关。  相似文献

Combined photochemical arylation, “nuisance effect” (SNAr) reaction sequences have been employed in the design of small arrays for immediate deployment in medium-throughput X-ray protein–ligand structure determination. Reactions were deliberately allowed to run “out of control” in terms of selectivity; for example the ortho-arylation of 2-phenylpyridine gave five products resulting from mono- and bisarylations combined with SNAr processes. As a result, a number of crystallographic hits against NUDT7, a key peroxisomal CoA ester hydrolase, have been identified.  相似文献

This article reports an extraction–purification of napins from an industrial rapeseed meal and the assessment of their antimicrobial activity against Fusarium langsethiae. The best extraction conditions are observed at pH 2, 12% (w/w) of rapeseed meal after 15 min of extraction in water at room temperature. Under these conditions the extraction is highly selective, allowing a simple purification process (ammonium sulfate precipitation followed by desalting size exclusion chromatography) to get purified napins. These napins possessed significant anti-Fusarium activity (IC50 = 70 μM) and a compact secondary structure rich in α-helix, which may explain this bioactivity.  相似文献

The effect of thermal profile on microstructure is studied in the frame of thin films deposited by inkjet-printing technology. The role of sintering temperature and thermal ramp is particularly investigated. Fast heating ramps exhibit coarse grains and pores, especially when a hybrid microwave curing is performed. This enhanced growth is attributed to the quick activation of densifying sintering regimes without undergoing thermal energy loss at low temperature. Microstructural evolution of various sintered inkjet-printed films is correlated with electrical resistivity and with the Young’s modulus determined by nanoindentation. A strong link between those three parameters is highlighted during experiments giving credit to either a surface or a fully volumetric sintering, according to the process. Sintering is then mainly triggered by surface mass transfer or by grain boundary diffusion. Silver thin films with an electrical resistivity 4–5 times higher than the bulk, has been reached in a few minutes with a Young’s modulus of 38?GPa.  相似文献

This paper investigates the various techniques used in the literature to calculate the effective Lewis number of two-component (H2/CO and H2/CH4) and three-component fuels (H2/CO/CH4 and H2/CO/CO2) over a wide range of equivalence ratios (0.6 ≤ φ ≤ 1.4) under laminar flame conditions. The most appropriate effective Lewis number formulation is identified through comparison with experimentally extracted Lewis numbers (Le). The paper first identifies the proper methodology to extract the experimental Le from the burned Markstein length of an outwardly propagating flame. Second, the different methodologies for the calculation of the effective Le are presented and compared to experimental results for H2/CH4 and H2/CO mixtures. Based on the experimental results, it is shown that the calculation of the effective Le of mixtures can be divided into a three-step procedure depending on the equivalence ratio: (1) calculation of the Le for each fuel and the oxidizer; (2) use of the Le mixing rule; and (3) assessment of the necessity or not of combining the fuel's and oxidizer's Lewis numbers. The paper shows that, in rich mixtures, the oxidizer Le needs to be taken into account. Lastly, the methodology is validated for H2/CO/CH4 and H2/CO/CO2 fuels.  相似文献
