Point centromeres, found in some ascomycete yeasts such Saccharomyces cerevisiae, are very different in structure from the centromeres of other eukaryotes. They are tiny and nonrepetitive and contain only two short conserved sequence motifs. Until recently, point centromeres were thought to have a single evolutionary origin, in the budding yeast family Saccharomycetaceae. Most yeasts outside this family have centromeres that are many kilobases in size. Some have centromeres consisting of a large inverted repeat sequence, others have centromeric clusters of retrotransposons, and a third group including Candida albicans has centromeres with no conserved sequence features. It was recently reported that Scheffersomyces stipitis has point centromeres with a strongly conserved 125-bp core sequence, which is unexpected because Sstipitis is only distantly related to the known point-centromere species. We show here that the 125-bp core sequence is actually part of the long terminal repeat (LTR) of the Ty5-like retrotransposon Tps5, which forms a cluster in the centromeric region of each Sstipitis chromosome. Thus, the LTR of a centromere-associated retrotransposon confers centromere-like mitotic stability when cloned into a plasmid. The centromeric regions of Sstipitis contain three types of Tps5 element (Tps5a, Tps5b, and Tps5c) and a noncoding nonautonomous large retrotransposon derivative.  相似文献

The last decade has witnessed an extraordinary shift in housing policy in Canada. Responsibility for social housing has been devoluted from the federal government to theprovincial and territorial governments, who in turn shift administration and manangement to regional and municipal agencies. And while the proportion of needy families is increasing, the deficit-minded Federal government only maintains its financial commitments to existing projects with no new funds presently available. Market solutions are being promoted by both the public and private sectors through a wide range of activities. The impact of calcium–magnesium–alumino-silicate (CMAS) degradation is a critical factor for development of new thermal and environmental barrier coatings. Several methods of preventing damage have been explored in the literature, with formation of an infiltration inhibiting reaction layer generally given the most attention. Gd2Zr2O7 (GZO) exemplifies this reaction with the rapid precipitation of apatite when in contact with CMAS. The present study compares the CMAS behavior of GZO to an alternative thermal barrier coating (TBC) material, GdAlO3 (GAP), which possesses high temperature phase stability through its melting point as well as a significantly higher toughness compared with GZO. The UCSB laboratory CMAS (35CaO–10MgO–7Al2O3–48SiO2) was utilized to explore equilibrium behavior with 50:50 mol% TBC:CMAS ratios at 1200, 1300, and 1400°C for various times. In addition, 8 and 35 mg/cm2 CMAS surface exposures were performed at 1425°C on dense pellets of each material to evaluate the infiltration and reaction in a more dynamic test. In the equilibrium tests, it was found that GAP appears to dissolve slower than GZO while producing an equivalent or higher amount of pore blocking apatite. In addition, GAP induces the intrinsic crystallization of the CMAS into a gehlenite phase, due in part to the participation of the Al2O3 from GAP. In surface exposures, GAP experienced a substantially thinner reaction zone compared with GZO after 10 h (87 ± 10 vs. 138 ± 4 μm) and a lack of strong sensitivity to CMAS loading when tested at 35 mg/cm2 after 10 h (85 ± 13 versus 246 ± 10 μm). The smaller reaction zone, loading agnostic behavior, and intrinsic crystallization of the glass suggest this material warrants further evaluation as a potential CMAS barrier and inclusion into composite TBCs.  相似文献

Terror Management Theory (TMT) suggests that mortality salience (MS), or death reminders, should impact environmental behaviour and decision-making by increasing consumption and resource usage, shifting aesthetic preferences toward cultivated landscapes, and affecting adherence to environmental norms. We examined MS effects on residential flood risk perceptions in Toronto, Canada, following the major urban flood in July 2013. Survey responses were analyzed to assess risk perceptions across nine categories; analyses through the Wilcoxon-Mann–Whitney test show statistically insignificant differences between MS and control groups for all subcategories. Coupled with the TMT literature, the results indicate that existential anxiety is more likely to affect flood risk perceptions through worldview defense when the social dimension of flooding is made clear. Toronto residents’ societal beliefs about flooding play a major role in guiding flood management strategies. Our results provide scholarly and practical direction for flood risk communication strategies to promote the adoption of private flood-prevention measures, suggesting that anthropocentric framing of flooding issues will not induce negative environmental behaviour and may increase concern for flooding issues.  相似文献

Generating sentences from a library of signs implemented through a sparse set of key frames derived from the segmental structure of a phonetic model of ASL has the advantage of flexibility and efficiency, but lacks the lifelike detail of motion capture. These difficulties are compounded when faced with real-time generation and display. This paper describes a technique for automatically adding realism without the expense of manually animating the requisite detail. The new technique layers transparently over and modifies the primary motions dictated by the segmental model and does so with very little computational cost, enabling real-time production and display. The paper also discusses avatar optimizations that can lower the rendering overhead in real-time displays.  相似文献

(Ti,Cr)N nanolayer coatings were deposited on Ti–6Al–4V, 17-4PH and Inconel 718 substrates using cathodic arc physical vapor deposition for improved erosion and corrosion resistance. Coating corrosion performance was highly dependent on the coating thickness and packing factors and correlated with increased chromium content within the (Ti,Cr)N nanolayer coatings. The change in cathode current predominantly affected coating thickness and the bias affected the packing factor. Erosion tests of the coated and uncoated substrates at both 30° and 90° erodent impingement angles were conducted using angular aluminum oxide media at particle velocities up to 145 m/s. Chromium evaporator current and substrate bias were varied to change film stoichiometry and microstructure for erosion performance evaluation. When chromium evaporator current was varied, the increase in chromium content led to an increase in binary CrN phase volume and a decrease in TiN phase volume. The increase in CrN phase volume decreased both hardness and erosion performance at both impingement angles. Lower bias values resulted in better erosion performance. At 30° erodent impingement, all coated samples outperformed the uncoated substrate; whereas, for 90° impingement, only coatings deposited at low bias values (? 25 V, ? 50 V, and ? 100 V) and high Ti:Cr ratios (> 2.4) outperformed the uncoated substrate. Nineteen ecologically relevant streamflow characteristics were estimated using published rainfall–runoff and regional regression models for six sites with observed daily streamflow records in Kentucky. The regional regression model produced median estimates closer to the observed median for all but two characteristics. The variability of predictions from both models was generally less than the observed variability. The variability of the predictions from the rainfall–runoff model was greater than that from the regional regression model for all but three characteristics. Eight characteristics predicted by the rainfall–runoff model display positive or negative bias across all six sites; biases are not as pronounced for the regional regression model. Results suggest that a rainfall–runoff model calibrated on a single characteristic is less likely to perform well as a predictor of a range of other characteristics (flow regime) when compared with a regional regression model calibrated individually on multiple characteristics used to represent the flow regime. Poor model performance may misrepresent hydrologic conditions, potentially distorting the perceived risk of ecological degradation. Without prior selection of streamflow characteristics, targeted calibration, and error quantification, the widespread application of general hydrologic models to ecological flow studies is problematic. Professional plumbers play an essential role in the implementation of water efficiency. If North America is to achieve high water efficiency standards and more ambitious plumbing codes, plumbers will need to be actively included in the water efficiency discussion. Repositioning the industry will required a cultural shift because the plumbing community has been mostly ignored in discussions of the larger environmental agenda and priorities. This repositioning will require substantial rethinking and retraining. New knowledge will need to be transmitted about emerging water efficient technologies, public policies and practices, as well as the rationale for use in residential and Industrial, Commercial, and Institutional (ICI) sectors. The GreenPlumbers Program (GPP) initiated this knowledge transfer process. Originating in Australia and expanded to the United States, the GPP is a national training and accreditation program for professional plumbers. Their focus has been on upgrading skills and awareness of water efficiency, conservation, and the professional plumbers’ roles in the contemporary environmental context. In this paper we report on the efficacy of the GPP’s curriculum and the process of transferring explicit water efficiency knowledge. Semi-structured interviews and a survey were used to gather the data. We considered how the program participants incorporated the GPP curriculum into their ‘day-to-day’ practices and operations post-certification. We also investigated participants’ motivating factors and cross-referenced these findings to their overall assessment of the program. Recommendations focus on how the GPP can best influence and contribute to a more comprehensive water efficiency agenda.  相似文献
