Prediabetes is a high-risk condition for type 2 diabetes (T2D). Pancreatic β-cells adapt to impaired glucose regulation in prediabetes by increasing insulin secretion and β-cell mass expansion. In people with prediabetes, metformin has been shown to prevent prediabetes conversion to diabetes. However, emerging evidence indicates that metformin has negative effects on β-cell function and survival. Our previous study established the Nile rat (NR) as a model for prediabetes, recapitulating characteristics of human β-cell compensation in function and mass expansion. In this study, we investigated the action of metformin on β-cells in vivo and in vitro. A 7-week metformin treatment improved glucose tolerance by reducing hepatic glucose output and enhancing insulin secretion. Although high-dose metformin inhibited β-cell glucose-stimulated insulin secretion in vitro, stimulation of β-cell insulin secretion was preserved in metformin-treated NRs via an indirect mechanism. Moreover, β-cells in NRs receiving metformin exhibited increased endoplasmic reticulum (ER) chaperones and alleviated apoptotic unfold protein response (UPR) without changes in the expression of cell identity genes. Additionally, metformin did not suppress β-cell mass compensation or proliferation. Taken together, despite the conflicting role indicated by in vitro studies, administration of metformin does not exert a negative effect on β-cell function or cell mass and, instead, early metformin treatment may help protect β-cells from exhaustion and decompensation.  相似文献

In a long-term study on heterotrophic biofilms in tube reactors, this investigation focused on mass transfer at the bulk/biofilm interface, biofilm density and substrate conversion rates. Several biofilms were cultivated under different substrate and hydrodynamic conditions. Oxygen concentration profiles were measured with microelectrodes in the biofilm and in the boundary layer directly in the biofilm tube reactors. The thickness of the concentration boundary layer was found to depend on the surface structure of the biofilm. The hydrodynamic conditions and the substrate load during the growth phase of the biofilm in biofilm systems are two key parameters that influence the biofilm growth, particularly the structure, density and thickness. The measured substrate conversion rates, biofilm densities and the boundary layer thickness were used to formulate an equation for the mass transfer in biofilm tube reactors.  相似文献

ABSTRACT

For many cities in the Global South, colonialism played a dominant role in shaping their urban form. The historical objective of planning in colonial mother-cities was dealing with poor health and living conditions, therefore a planning approach similar to that followed in post-war Britain would appear beneficial in post-colonial cities, characterized by environmental and physical infrastructure unable to cope with massive population growth. Urban growth management is a discourse born in an attempt to control the growing industrial city in the early twentieth century, and in recent years applied through instruments such as urban edges or growth boundaries to limit urban sprawl and encourage higher density urban development. In South Africa, the principles of compaction and urban growth management formed part of the post-apartheid planning agenda towards transforming the inefficient and fragmented landscape inherited from separate spatial development. Consequently, urban edges and urban growth boundaries formed key components of municipal spatial planning frameworks since the early 2000s. The purpose of this paper is to explore the origin and status of urban edges in three metropolitan municipalities in South Africa to aid in understanding of these spatial instruments in the south.  相似文献

We present a novel approach to determine the surface roughness on varying scales using atomic force microscopy data. The key factor is to find a suitable background correction for the desired scale. Using the example of the surface of sized and unsized high-tenacity carbon fibers, we present an easy method to find backgrounds for widely varying scales and to evaluate respective topography and surface roughness with the same lateral resolution as the microscope itself. The analysis is done by subtracting a tunable background from the respective height data. By choosing an appropriate background to investigate the surface topography of a carbon fiber on a nm-scale, only small nano-structures with a width of around 20 nm remain after the background subtraction. Evaluating the mean roughness R a of these nano-structures, sized carbon fibers show an overall value of around 0.1 nm while unsized carbon fibers a value of around 0.4 nm. Total background corrected height analysis shows an even distribution of these nano-structures along the fibrils of the unsized fibers, whereas for the sized fibers the nano-structures are not present. The presented method allows analysis and visualization of the distribution of nano-structures on a carbon fiber surface for the first time. This feature is used to visualize the distribution of the sizing and can further be used to investigate the influence of different production parameters on the fiber topography or to evaluate the contribution of mechanical interlocking to the interfacial strength.  相似文献

Early carcinogenicity tests found no evidence of activity for picene but found considerable initiating and carcinogenic activity for DBA. More recent investigation suggests that both pentacyclics are complete carcinogens when administered as single s.c. injections in NMRI mice, despite findings that picene acts as neither an initiating nor promoting agent. To investigate this contradiction, the complete carcinogenicity of both isomers was compared by s.c. injection in female Sprague-Dawley rats. The results demonstrated that 1 w mol of DBA administered three times weekly for 20 doses, induced sarcomas in all test animals by 33 weeks (100%). Similar treatment with picene did not induce sarcoma in any test animals by 37 weeks (0%). These present results agree with the earlier studies. It follows then that the metabolic activation and carcinogenicity of DBA and picene are dependent on the presence of an L-region, but are independent of the presence of a bay-region.  相似文献

The charge distributions of an improved opposed flow unipolar diffusion charger were measured using a tandem differential mobility analyzer (DMA) set up in a size range of approximately 20–400 nm. The charger is intended to be used in a portable aerosol sizer to measure particle size distributions. The determined charge distributions were represented by lognormal distributions, and a set of equations and coefficients was developed to calculate the charge distributions. These equations can be easily implemented in software for size distribution measurements. The agreement between the mathematically derived and measured charge distributions is very good, with regression coefficients R 2 > 0.96. The investigations showed that approximately 55% of 20-nm particles remain uncharged, while up to 25 elementary charges need to be considered for multiple charge correction of 400-nm particles. Comparison with the Fuchs theory delivered satisfying agreement with the measured average charge levels, but charge distributions cannot be described by the Fuchs theory, likely caused by the charger geometry.

The objective of this study was to investigate the protective effect of five different emulsifiers on iron‐mediated lipid oxidation in 70% fish oil‐in‐water emulsions. The emulsifiers were either based on protein (whey protein isolate and sodium caseinate) or based on phospholipid (soy lecithin and two milk phospholipids with different phospholipid contents, MPL20 and MPL75). Lipid oxidation was studied at pH 4.5 and 7.0, and results were compared to lipid oxidation in neat fish oil. Results showed that all emulsions oxidised more than neat oil. Furthermore, emulsions prepared with proteins oxidised more at low pH than at high pH, and casein emulsions oxidised the least (Peroxide value (PV) at day 7 was 0.5–0.7 meq kg?1). Among emulsions prepared with phospholipids, emulsions with MPL75 were the most oxidised followed by emulsions prepared with lecithin and MPL20. Thus, PV in MPL75 emulsions was 5.0–5.5 meq kg?1 at day 7 compared with 0.9–1.9 meq kg?1 in MPL20 emulsions.  相似文献
