Toxin-producing cyanobacteria are abundant in surface waters used as drinking water resources. Microcystins (MC) produced by certain cyanobacteria present acute and chronic toxicity, and their removal in drinking water treatment processes is of increasing concern. Previous studies have demonstrated that chlorine and potassium permanganate are feasible oxidants for the removal of MCs present in drinking water resources, although the oxidation might lead to toxic oxidation products. In this paper, the toxicity of the oxidation products of MC-LR and -RR has been studied using protein phosphatase 1 inhibition assay (PPIA). The HPLC and ELISA analyses correlated with the PPIA results for both toxins. The samples containing the oxidation products were fractionated by HPLC and the toxicity of the fractions was tested with PPIA. The results revealed that protein phosphatase 1 inhibition emerged only from intact MC, while the oxidation products were non-toxic. Similar results were obtained in experiments performed in natural waters: no reaction products or interactions exhibiting protein phosphatase 1 inhibition were detected.  相似文献

We report on the fabrication of covalently crosslinked and amine-reactive hollow microcapsules using 'reactive' layer-by-layer assembly to deposit thin polymer films on sacrificial microparticle templates. Our approach is based on the alternating deposition of layers of a synthetic polyamine and a polymer containing reactive azlactone functionality. Multilayered films composed of branched poly(ethylene imine) (BPEI) and poly(2-vinyl-4,4-dimethylazlactone) (PVDMA) were fabricated layer-by-layer on the surfaces of calcium carbonate and glass microparticle templates. After fabrication, these films contained residual azlactone functionality that was accessible for reaction with amine-containing molecules. Dissolution of the calcium carbonate or glass cores using aqueous ethylenediamine tetraacetic acid (EDTA) or hydrofluoric acid (HF), respectively, led to the formation of hollow polymer microcapsules. These microcapsules were robust enough to encapsulate and retain a model macromolecule (FITC-dextran) and were stable for at least 22 hours in high ionic strength environments, in low and high pH solutions, and in several common organic solvents. Significant differences in the behaviors of capsules fabricated on CaCO(3) and glass cores were observed and characterized using scanning electron microscopy (SEM) and energy dispersive X-ray spectroscopy (EDS). Whereas capsules fabricated on CaCO(3) templates collapsed upon drying, capsules fabricated on glass templates remained rigid and spherical. Characterization using EDS suggested that this latter behavior results, at least in part, from the presence of insoluble metal fluoride salts that are trapped or precipitate within the walls of capsules after etching of the glass cores using HF. Our results demonstrate that the assembly of BPEI/PVDMA films on sacrificial templates can be used to fabricate reactive microcapsules of potential use in a wide range of fields, including catalysis, drug and gene delivery, imaging, and biomedical research.  相似文献

River systems draining peaty catchments are considered a source of atmospheric CO2,thus understanding the behavior of the dissolved inorganic carbon pool (DIC) is valuable. The carbon isotopic composition, delta13C(DIC), and concentration, [DIC], of fluvial samples collected diurnally, over 14 months, reveal the DIC pools to be dynamic in range (-22 to -4.9% per hundred, 0.012 to 0.468 mmol L(-1) C), responding predictably to environmental influences such as changing hydrologic conditions or increased levels of primary production. delta(18)O of dissolved oxygen (DO) corroborates the delta(13)C(DIC) interpretation. A nested catchment sampling matrix reveals that similar processes affect the DIC pool and thus delta(13)C(DIC) across catchment sizes. Not so with [DIC]: at high flow, the DIC export converges across catchment size, but at low flow catchments diverge in their DIC load. Contextualizing delta(13)C with discharge reveals that organic soil-waters and groundwaters comprise end-member sources, which in varying proportions constitute the fluvial DIC pool. Discharge and pH describe well [DIC] and delta(13)C(DIC), allowing carbon to be apportioned to each end-member from continuous profiles, demonstrated here for the hydrological year 2003-2004. This approach is powerful for assessing whether the dynamic response exhibited here is ubiquitous in other fluvial systems at the terrestrial-aquatic interface or in larger catchments.  相似文献

Triclosan is a widely used antibacterial agent found in many personal hygiene products. Although it has previously been established that pure triclosan and free chlorine readily react, interactions between triclosan-containing consumer products and free chlorine have not previously been analyzed in great depth. Sixteen double-blinded solutions including both triclosan-containing (1.14-3.12 mg triclosan/g product) and triclosan-free products were contacted with free chlorine at pH 7. Products detected included (chlorophenoxy) phenols, 2,4-dichlorophenol, 2,4,6-trichlorophenol, and chloroform. The daughter product yields were found to be highly variable and were dependent on the antimicrobial product investigated, the free chlorine to triclosan ratio, and the temperature at which the study was conducted. Lowering the temperature from 40 to 30 degrees C resulted in a decreased average chloroform yield from 0.50 to 0.37 mol chloroform/mol triclosan consumed after 1 min of reaction time for an initial free chlorine concentration of 4.0 mg/L as Cl2. At 40 degrees C the average molar chloroform yields decreased to 0.29 and <0.1 when the initial free chlorine concentration was decreased to either 2.0 or 1.0 mg/L as Cl2, respectively. Field experiments, in which Atlanta, GA and Danville, VA tap waters were augmented with various soap products, exhibited results varying from the laboratory experiments in that different productyields were observed. These differences are attributed to the chlorine demand of constituents in the tap water. A simple exposure model suggests that exposure to chloroform can be significant under some conditions.  相似文献

A total of 426 strains of Micrococcaceae bacteria isolated from chorizo (a traditional Spanish fermented sausage) were identified. The chorizos were sampled from three regions of Castilla and León in Spain: Burgos, Segovia and Salamanca. Two factories were chosen in each region and the samples were taken at three stages of ripening. Staphylococcus xylosus was the most predominant species isolated (95%). Twelve strain types of S. xylosus were established according to their fermentation patterns, and two of them, S. xylosus type 2 and S. xylosus type 5, made up the majority of the strains of S. xylosus isolated (27 and 52%). Production of acetoin, nitrate reductase, urease activity, proteolytic and lipolytic activity were determined for all isolates. The percentage of strains of S. xylosus producing acetoin depends on the manufacturing location. In general, the proteolytic and lipolytic activities of the S. xylosus isolated from chorizo from Castilla and León were low and moderate; 97% of the strains showed nitrate reductase and urease activity. According to our results and to previous investigations, it seems that S. xylosus type 5, showing nitrate reductase and urease activity, low-moderate proteolytic and lipolytic activities and not producing acetoin would be suitable as a starter culture. Of the strains isolated in this study, 38% comply with these requirements.  相似文献
