The triterpenoid (+)-ambrein is the major component of ambergris, a coprolite of the sperm whale that can only be rarely found on shores. Upon oxidative degradation of (+)-ambrein, several fragrance molecules are formed, amongst them (−)-ambrox, one of the highest valued compounds in the perfume industry. In order to generate a Saccharomyces cerevisiae whole-cell biocatalyst for the production of (+)-ambrein, intracellular supply of the squalene was enhanced by overexpression of two central enzymes in the mevalonate and sterol biosynthesis pathway, namely the N-terminally truncated 3-hydroxy-3-methylglutaryl-CoA reductase 1 (tHMG) and the squalene synthase (ERG9). In addition, another key enzyme in sterol biosynthesis, squalene epoxidase (ERG1) was inhibited by an experimentally defined amount of the inhibitor terbinafine in order to reduce flux of squalene towards ergosterol biosynthesis while retaining sufficient activity to maintain cell viability and growth. Heterologous expression of a promiscuous variant of Bacillus megaterium tetraprenyl-β-curcumene cyclase (BmeTC-D373C), which has been shown to be able to catalyse the conversion of squalene to 3-deoxyachillol and then further to (+)-ambrein resulted in production of these triterpenoids in S. cerevisiae for the first time. Triterpenoid yields are comparable with the best microbial production chassis described in literature so far, the methylotrophic yeast Pichia pastoris. Consequently, we discuss similarities and differences of these two yeast species when applied for whole-cell (+)-ambrein production.  相似文献

Current developments in cement chemistry increasingly rely on predictive thermodynamic modeling of the phase composition in cementitious composites with the aim of linking the performance of the material with the phase composition of the material. This approach requires identification of the cement phases that form in hydrating cementitious materials using standard techniques, such as X‐ray diffraction (XRD) and thermal analysis (DTA, TGA), but also state‐of‐the‐art synchrotron‐based techniques, in particular for those cases in which the signals of solid solutions overlap in XRD and TGA measurements. In this study, two ordinary Portland cements, with different chemical compositions and subject to different hydration times (~10, ~50 yr), were investigated aiming at identifying the most stable Fe‐containing cement phase in the cement pastes. The Fe‐containing cement phases and their solid solutions with the Al analogues in the complex cement matrix were analyzed with X‐ray absorption spectroscopy, indicating the formation of a mixed Fe–Al siliceous hydrogarnet as the major Fe‐containing phase. The presence of this phase after long hydration periods and upon selective dissolution of the pastes further indicates that, independent of the chemical compositions of cements, formation of the mixed Fe–Al siliceous hydrogarnet is thermodynamically favored in aged pastes, which is supported by published thermodynamic calculations.  相似文献

Hydratases provide access to secondary and tertiary alcohols by regio‐ and/or stereospecifically adding water to carbon‐carbon double bonds. Thereby, hydroxy groups are introduced without the need for costly cofactor recycling, and that makes this approach highly interesting on an industrial scale. Here we present the first crystal structure of a recombinant oleate hydratase originating from Elizabethkingia meningoseptica in the presence of flavin adenine dinucleotide (FAD). A structure‐based mutagenesis study targeting active site residues identified E122 and Y241 as crucial for the activation of a water molecule and for protonation of the double bond, respectively. Moreover, we also observed that two‐electron reduction of FAD results in a sevenfold increase in the substrate hydration rate. We propose the first reaction mechanism for this enzyme class that explains the requirement for the flavin cofactor and the involvement of conserved amino acid residues in this regio‐ and stereoselective hydration.  相似文献

Although topology optimization is well established in most engineering fields, it is still in its infancy concerning highly non-linear structural applications like vehicular crashworthiness. One of the approaches recently proposed and based on Hybrid Cellular Automata is modified here such that it can be applied for the first time to thin-walled structures. Classical methods based on voxel techniques, i.e., on solid three-dimensional volume elements, cannot derive structures made from thin metal sheets where the main energy absorption mode is related to plastic buckling, folding and failure. Because the main components of car structures are made from such thin-walled beams and panels, a special approach using SFE CONCEPT was developed, which is presented in this paper.  相似文献

This paper presents novel algorithms which are able to generate recommendations within a heterogeneous service environment. In this work explicitly set preferences as well as implicitly logged viewing behavior are employed to generate recommendations for Digital Video Broadcast (DVB) content. This paper also discusses the similarity between the DVB genres and YouTube categories. In addition it presents results to show the comparison between well known collaborative filtering methods. The outcome of this comparison study is used to identify the most suitable filtering method to use in the proposed environment. Finally the paper presents a novel Personal Program Guide (PPG), which is used as a tool to visualize the generated recommendations within a heterogeneous service environment. This PPG is also capable of showing the linear DVB content and the non-linear YouTube videos in a single view.  相似文献

Observed differences between measured and calculated elastic constants for Ti3SiC2 are investigated using Density Functional Theory and Inelastic Neutron Scattering. The agreement between the calculated lattice dynamics and the dynamics measured by inelastic neutron scattering is considered good except at energies below ~20 meV where discrepancies suggest anharmonic potentials. This suggestion is confirmed by Density Functional Theory—Molecular Dynamics simulation which shows multiple site occupancy of the Si atoms within the basal plane at finite temperature and produces a calculated inelastic spectrum in better agreement with the measured spectrum in the low‐energy region. The highly anharmonic potential energy surface of the Si atoms offers an explanation for the failure of elastic constants, calculated based on the harmonic approximation, to agree with initial experimental measurements.  相似文献

The production of raw materials causes positive (economic) effects (added value, new jobs) as well as negative impacts on the environment and the quality of life (noise, land use etc.). Spatial planning aims at minimising land use conflicts between mining sites on one hand and housing developments and the conservation of the environment on the other hand. During the planning permission approval process the economic effects of the raw material production on a specific site and the interests of protecting nature and environment have to be counterbalanced. Main arguments for existing public interests of raw materials production are the coverage of the regional demand, the reduction of heavy traffic, regional added value and the creation of new jobs. The more particular the economic net effects of a new raw materials production can be justified, the stronger the position of the developer in the planning permission approval process will be.  相似文献

The increasing prevalence of inflammatory diseases and the adverse effects associated with the long-term use of current anti-inflammatory therapies prompt the identification of alternative approaches to reestablish immune balance. Apigenin, an abundant dietary flavonoid, is emerging as a potential regulator of inflammation. Here, we show that apigenin has immune-regulatory activity in vivo. Apigenin conferred survival to mice treated with a lethal dose of Lipopolysaccharide (LPS) restoring normal cardiac function and heart mitochondrial Complex I activity. Despite the adverse effects associated with high levels of splenocyte apoptosis in septic models, apigenin had no effect on reducing cell death. However, we found that apigenin decreased LPS-induced apoptosis in lungs, infiltration of inflammatory cells and chemotactic factors’ accumulation, re-establishing normal lung architecture. Using NF-κB luciferase transgenic mice, we found that apigenin effectively modulated NF-κB activity in the lungs, suggesting the ability of dietary compounds to exert immune-regulatory activity in an organ-specific manner. Collectively, these findings provide novel insights into the underlying immune-regulatory mechanisms of dietary nutraceuticals in vivo.  相似文献
